Modification of material flow for a production line
Zadák, Zdeněk ; Jelínek, Oldřich (referee) ; Kašpárek, Jaroslav (advisor)
The main aim of this master´s thesis is the analysis and design of internal supply production lines in the company Edscha Automotive Kamenice s.r.o. The company manufactures components for the automotive industry. Internal material flow are mapping and described in chapter 3. This chapter describes the current state of supply buffers on production lines. Buffers are supplying by small materials. Next part of this work is describing the method of removal of finished production and using system Kanban in company. Chapter 4 of this thesis is devoted to the design part. The design part is first described the creation of models in Excel and MS software Tecnomatix Plant Simulation. Subsequently they are compiled proposals for the supply of production lines, small material and transport the finished products. Proposals in this part evaluated and a recommendation for implementing into operation. This proposal is then simulated in software Tecnomatix Plant Simulation and compared with the current situation. It is also determined by the number of handlers on the functioning of material flow. Furthermore, they are given further suggestions for improvements, some of which require a little change in the current system Kanban and supply.
Effect of piracetam on advanced dementia with predominated cardiovascular symptoms
Kovtun, Vladimír ; Ďoubal, Stanislav (advisor) ; Zadák, Zdeněk (referee) ; Mrňa, Boris (referee)
From 2001 to 2004 a retrospective non-invasive clinical trial on the effects off Piracetam was carried out in the nursing home in Česká Kamenice. The effect of Piracetam was observed on 150 patients aged 84,29 ( +- 3,5 years ) who suffered from an advanced form of vascular dementia. The patients were diagnosed in accordance with all currently valid criteria. At the time of the clinical trial the patients had been suffering from the disease for more than 6 months. A placebo was administered to the reference group composed of patients suffering from the same disease. The clinical trial lasted for a period of 12 months . The treatment with Piracetam did not demonstrate any positive effects which would be statistically substantial. This is probably due to an increased number of advanced forms of dementia syndrome with an irreversible loss of the brain's ability to compensate. For the above mentioned reasons an early diagnosis of demetia is of high importance because of a possibility to positively influence the disease. We assume that it is a task of an immense significance which goes beyond the boarders of the Czech Republic. Also, it raises the importance of geriatrics and gerontopharmacy on the societal ladder. Obviously, it is necessary to conduct further and much more extensive studies, which would...
Relationship of apolipoprotein E polymorfism on the functional fitness in the elderly
Šnejdrlová, Michaela ; Češka, Richard (advisor) ; Hubáček, Jaroslav (referee) ; Zadák, Zdeněk (referee)
(EN) Background: The life expectancy is defined by a combination of genotype (25%) and environmental influences, including lifestyle (75%). Nevertheless a stronger genetic influence is expressed in long-living individuals. Epsilon gene for apolipoprotein E (APOE) belongs among the most studied so called "candidate genes of longevity". The association of APOE polymorfism is proved in relation to many diseases (hypercholesterolaemia, cardiovascular disease, Alzheimer disease), thus we tried to evaluate the relation of APOE polymorfism and senior fitness. Methods: In our pilot study we examined a total number of 128 patients (93 women, 35 men), all of them older than 80 years, average age 88.6 ±5.3 years. Examinations: A) detailed examination of functional fitness (ADL, MMSE, Yeassavage's depression scale , MNA, 6-MWT). Based on this complex examination, patients have been sorted to 5 functional categories according to W. Spirdus, these categories objectively express the level of self - sufficiency. B) laboratory examinations C) examintaion of APOE gene polymorphism using DNA restriction analysis, DNA isolated from peripheral leucocytes Results: Our pilot study proved statisticaly significant relation (p = 0,0248) between allele E4 and functional fitness in senior age. According to the results of...

The role of cholesterol, its precursors and oxidative products in clinically serious conditions
Vyroubal, Pavel ; Zadák, Zdeněk (advisor) ; Matějovič, Martin (referee) ; Dítě, Petr (referee)
The role of cholesterol, its precursors and oxidative products in clinically serious conditions Objective: The aim of the study was to elucidate the role and importance of hypocholesterolemia in clinically serious conditions. It was a monocentric, prospective clinical study. Material and Methods: Two groups of patients were recruited into the study - one group consisted of patients with coronary heart disease (CHD), who underwent miniinvasive cardiosurgical operation without extracorporeal circulation (n=22) and the other group of patients, who sustained polytrauma (n=31). Fifty three patients were recruited into the study. We performed the determination of sterols (total cholesterol, HDL cholesterol, LDL cholesterol, triacylglycerols) and their precursors (β-sitosterol, campesterol, lathosterol, skvalen), interleukin IL-6, insulin, C-peptid and cortisol in the blood serum. The short version of ACTH stimulation test was performed. The oxidative burst of granulocytes was evaluated. The blood samples were taken on the day of admission, the first, the fourth and the eighth post-operative and post-traumatic day. Results: There was a significant early decline of total cholesterol (TC) and LDL cholesterol level followed by full recovery during observed period. There was a decline of cholesterol synthesis...
The significance of the analysis of noncholesterol sterols and profile of fatty acids in various pathophysiological states
Vecka, Marek ; Tvrzická, Eva (advisor) ; Zadák, Zdeněk (referee) ; Kazdová, Ludmila (referee)
At present time, there is growing interest in two classes of lipids - sterols and fatty acids, which posses many different roles in living organisms. The concentrations of cholesterol biosynthetic precursors (lathosterol, desmosterol) reflect level of cholesterol biosynthesis, whereas the plasma phytosterol (campesterol, β-sitosterol) concentrations can be used for estimation of fractional absorption of dietary cholesterol. These groups are sometimes called together as noncholesterol sterols. The dissertation thesis dealt with the significance of analysis of noncholesterol sterols and fatty acid profile in various pathophysiological states. The methodology part of the thesis covered some aspects of analysis of these compounds with capillary gas chromatography. We found that for the minor fatty acid analysis, both the type of the column and the software used play an important role. The gas chromatography step contributes by the least part to the total error of the procedure, whereas the effect of other steps (extraction, thin-layer chromatography and methylation process) is more pronounced. The comparison of fatty acid profiles in lipoprotein particles with low and very low density revealed differences, but these differences were highly interindividual. We also found that it is possible to replace the...
The trans-fatty acids in subcutaneous fat and in fat content of breast milk as an indicator of their nutritional intake
Dlouhý, Pavel ; Anděl, Michal (advisor) ; Dostálová, Jana (referee) ; Hrubý, Stanislav (referee) ; Zadák, Zdeněk (referee)
Trans fatty acids (TFA) are unsaturated fatty acids, which have one or more double bonds in the trans configuration. Their main dietary sources are partially hardened fats and food products containing partially hardened fats (margarines, shortenings, bakery products, and fast foods). TFA are thought to increase the risk of coronary heart disease and type 2 diabetes. These fatty acids have adverse effects on blood lipid levels, increasing LDL-cholesterol while decreasing HDL-cholesterol. Consumption of TFA can also impair insulin sensitivity. High intake of TFA is associated with systemic inflammation, activation of immune functions and endothelial dysfunction. Hypotheses also exist that TFA may have adverse effect on the metabolism of essential fatty acids and foetus development. The concentration of TFAs in subcutaneous fat is a very good indicator of dietary exposure of TFA. This concentration reflects long-term dietary intake of these fatty acids. Levels of TFA in human milk are more variable; however, it is quite simple to obtain a sample of human milk. The purpose of our studies was to determine (a) content of TFA in subcutaneous fat samples from persons with coronary atherosclerosis and from persons with no sign of coronary dinase and (b) the content of TFA in early human milk of breast-feeding women...
