Indicatours of sustainable development social pillar at the local level

The aim of the thesis is to find the answer, whether it is plausible to monitor social cohesion of the municipality and how to measure this cohesion, which is one of the essential pillars of sustainable development. In order to find out, analysis of approaches and measurements of the sustainable development social pillar in international and Czech environment was carried out. Social pillar was identified and described also at the local level. Within the framework of the sustainable development social pillar at the municipal level three main thematic blocks are proposed: • Social environment quality of the municipality • Social-demographic population structure of the municipality • Contentment and participation in the public affairs These themes should be monitored by the set of indicators proposed in this thesis. It aimed to cover all dimensions of social cohesion which are significant and well-founded in the context of the territorial planning process. These indicators can be followed in the direct relationship with the population size and regional importance of the observed territorial unit at the local level and they are not separated from the concept of indicators monitoring at other – territorially higher – levels. Outcomes, analysis and interpretation of attained values could become the basis for the “territorial sustainable development analysis” (part of territorial planning instruments according to contemporary legal regulations), representing social pillar. All phenomenon and processes must be evaluated in the context with other sustainable development pillars and afterwards they should become another suitable instrument for measurement of the fulfilling the territorial planning aims.

Study of substance transport via membrane transporters

Title of diploma thesis: Study of substance transport via membrane transporters Wide range of specialized transport proteins expressed mainly in the kidney proximal tubule ensures the excretion of endogenous and exogenous substances. OCT2 (organic cation transporter 2) is localized on the basolateral membrane of the kidney proximal tubule, the transporter transports broad spectrum of structurally different substances. Drug-drug interactions may occur at the level of this transporter due to its broad substrate specificity. Within this diploma thesis the accumulation of the radiolabeled antiviral agent [3 H]lamivudine and the model hOCT2 substrate [3 H]MPP+ was evaluated using accumulation and transport studies. The canine kidney cells MDCKII or human cell line HeLa were used as model cell lines. Mentioned cell lines expressed the human OCT2 transporter stably or transiently. The antibacterial chemotherapy agent trimethoprim was used as competitive inhibitor of OCT2 transporter. The dissertation is divided into two parts. The theoretical part introduces a bio, author's work, awards received, preferred literature genre, and selected methods chosen for particular work with text. The practical part focuses on the project O čertovi that develops the reading in kinder garden.

The role of the program The leader in me in developing competencies

The subject of this work is to describe and present the program The Leader in me as one of the possible starting points of the current educational effect on students of all levels of the educational system in terms of their equipment with competencies. The theoretical part defines the basic concepts related to the framework educational program, key competencies in education and the school educational program. Furthermore, the principle of 7 habits of effective behavior is described here and it is stated how they can be applied in the school environment with the help The practical part of the work focuses on the methods leading to the acquisition of key competencies within the classes of Personal - Social Education and on the possibilities and impact of introducing of the program The Leader in me into practice at the Primary School Bratří Čapků in Ústí nad Orlicí. The information was collected through interviews and follow-up questionnaires. Legal aspects of the circular economy Abstract The diploma thesis deals with the issue of legal aspects of the circular economy. This concept is based on resource efficiency with the intention to minimize waste generation. This approach is considered to be the future of economies not only in the European Union, but also in the rest of the world. The thesis aims to provide a comprehensive overview of the circular economy and the legal background of this concept in the EU and the Czech Republic. Given that the legislation of the circular economy has so far been reflected at the EU and national level predominantly in the field of waste management, it forms a significant part of this thesis. The projection of the principles of the circular economy into the legal order as well as the future development of this concept is evaluated. Recognized advantages and disadvantages in the current circular economy legislation are formulated at the end of the thesis. Subsequently, the thesis sets recommendations for the future direction of the circular economy and for its efficiency enhancement.

Transformation of the values of the individual by Friedrich Wilhelm Nietsche and Søren Aabye Kierkegaard.

Goal of this work is to show that the values in European and a traditional context could be transformed. I chose two philosophers F.Nietzsche and S.Kierkegaard where I can document this change. Just the two of them proved to be very important for the 20th century .In the main part of my work, I am describing and comparing the evolution of the axiological views of both philosophers. My study on existential philosophers includes topics such as freedom, sense of life, base of christianity, perception of human being. Nietzsche and Kierkegaard are philosophers who have a lot in common, but they would not get on well with each other. The things that they share are the same time what they deal with in a difááé=ferent way. They treat topics such as existence, futility, strength of faith. However Kierkegaard studies in depth existentialism, immersion in irrationality, desolation. In Nietzsche we can find the will of power, the nihilist opinions and the criticism of christianity.

New types of magnetic sorbents for phosphoprotein analysis

The method for the study of protein phoshorylation sites was elaborated. This method is based on the IMAC separation of phosphopeptides from protein proteolytic digests using new magnetic sorbents and on their subsequent identification by mass spectrometry (MS). Magnetic non-porous hydrophilic poly(2-hydroxyethyl methacrylate-co-glycidyl methacrylate) particles prepared by the dispersion polymerization and modified with iminodiacetic acid (IDA) with immobilized Fe(III) and Ga(III) ions were employed for the enrichment of phosphopeptides from the proteolytic digests of two model proteins, porcine pepsin A and bovine α-casein. The optimum conditions for phosphopeptide adsorption and desorption in both cases were investigated and compared. The phosphopeptides separated from both proteolytic digests were analyzed by matrix-assisted laser desorption/ionization time-of-flight MS. For the immunochemical separation of phosphoproteins, protein fraction containing antibodies was obtained from egg yolk of hens immunized with O-phosphoryl-L-serine conjugated to key limpet hemocyanin. Antibodies were purified using affinity chromatography on immobilized α-casein and their presence was proven by MS. Specificity of the obtained antibodies was examined using ELISA tests. The History of the Pharmacies of Prior Region Rychnov nad Kněžnou

Závěr Cílem předkládané diplomové práce bylo podat celkový obraz o historii i současné situaci lékáren na Rychnovsku tak, aby poskytla základní informace nejen dnešním zájemcům o dějiny lékárenství, ale aby též mohla být zdrojem poznatků do budoucna. Na začátku byly stručně zpracovány dějiny Rychnovska ukazující vývoj od dob zemědělství po zavedení průmyslu a rozmach regionu, jako významné a hojně navštěvované kulturní a turistické oblasti. Podrobněji pak byly rozepsány dějiny jednotlivých obcí a měst, kde lze konkrétně vidět postupný vývoj a zejména dnešní situaci jasně ukazující na potřebu lékárny v dané obci či městě. Zachytit historii starých historických lékáren se podařilo na základě studia historických materiálů (metodika zpracování je v předešlé kapitole). Roku 1980 bylo na území bývalého okresu Rychnov nad Kněžnou 9 lékáren. V současnosti existuje v okrese Rychnov nad Kněžnou 24 lékáren. Již tento údaj ukazuje oprávněnost zpracování diplomové práce, navazující na již dříve zpracované materiály. Bez diplomové práce by dějiny 24 lékáren nebyly zachyceny a v budoucnosti by nikdo neznal dějiny lékáren v letech 1989- 2004. Jedná se o historicky velice zajímavé období. Teprve budoucí generace ocení velikost této doby, kdy se prakticky v celé České republice i v jednotlivých regionech počet lékáren...

The Nazi ideology in relation to persons with mental disabilities

Diploma thesis "Nazi ideology in relation to mentally ill people" focuses on starting points and ideas of this ideology, which in its consequences led to social exclusion and subsequent systematic extermination of people with a certain handicap. Although the thesis is limited in time from 1933 - 1945, it also follows development line of selected scientific disciplines, which were significantly reflected in the Nazi ideology. Primarily it concerns historical science, medical disciplines, genetics, racial hygiene, eugenic origins and other related disciplines. The thesis also takes into account the economic situation of that time with its many social problems which arose as a result of the new geopolitical arrangement after the World War I and the changes in the social world of that period and it also captures the process of changes in the social status of mentally ill people in the course of history. Last but not least, it describes the practical consequences of these changes, which were "forced sterilization" and "euthanasia" of both children and adults. Concept of the individual in relation to God in Kierkegaard and Nietzsche

This work deals with philosophy of Søren Kierkegaard and Friedrich Nietzsche. It focuses on the problem of their relationship with God and religion. The aim is to show where these thinkers agree on and differ in relation to God, Christianity and change of the values of an individual. The first chapter is devoted to foundations of Nietzsche and Kierkegaard's work. The second chapter compares common and different values. The work compares masters and slaves' morality in the concept of Friedrich Nietzsche and morality built on authentic faith at Søren Kierkegaard.
