Laboratory determination of bone markers in the course of osteoporosis
Moučková, Štěpánka ; Kotaška, Karel (referee) ; Dršata, Jaroslav (advisor)
1 Abstrakt Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Štěpánka Moučková Supervisor: ing. Marta Pechová Title of bachelor thesis: Laboratory determination of bone markers in osteoporosis The aim of this study is to compare preliminary values of bone formation and resorption markers in newly diagnosed osteoporotic patients with values after their treatment. We compared the serum levels of N-MID osteocalcin and aminoterminal propeptide of procollagen type I (PINP) as markers for bone formation, and serum carboxyterminal telopeptide (β-CTx) as a marker of bone resorption. Data were collected from 66 patients - 60 women (47 postmenopausal and 13 premenopausal) and 6 men. Patients were divided into 4 groups according to age and applied treatment. Analysis of individual markers was performed using Roche Cobas e411 immunochemical analyzer. Simultaneously were measured serum levels of 25-OH vitamin D3, kalcium (TCa) and parathyrin. Results show that patients treated with bisphosphonates have a significant change in markers of bone turnover, while in patients without proven osteoporosis the levels of bone markers remain unchanged after the supplementation of vitamin D and calcium. Values of vitamin D without supplementation are often below the...

Immobilization of heavy metals in the cement matrix with the admixture of natural zeolite
Semerádová, Nikol ; Bayer, Patrik (referee) ; Vyšvařil, Martin (advisor)
Heavy metals released to the natural ecosystem constitute significant risks not only for organisms, but also for water supplies in the nature. There are a number of analytical methods for the determination of heavy metals. Each of them is suitable for different element and its character. Since the results of the analysis are applied during the solidification and stabilization, it is necessary to ensure the highest correlation between hazardous waste and solidification medium for maximum inhibition of toxic ions. This work investigates the stabilization of heavy metals binders based on cement base with admixture of the natural zeolite and its possible use for the solidification of heavy metals in soluble form.

Linzerová, Petra.: Biological Effect of Plant Metabolites III. Izolation of Alkaloids from Eschscholzia californica Cham. A Diploma Work, Charles University in Praque, Faculty Of Pharmacy in Hradec Králové, Department of Pharmaceutical Botany And Ecology, Hradec Králové 2009, 54 pages Aims of this diploma thesis were to isolate alkaloids from J1 extract obtained from a herb with roots from a plant Eschscholzia californica Cham., to perform the identification of gained alkaloid and to participate in determination of activity of isolated alkaloid against the erythrocytic acetylcholinesterase. After finding suitable mix of reagents we divided the extract on a chromatografic column filled with silica gel by system of chloroform : ethanol with gradual increasing of concentration ethanol. With obtained eluents we made a orientation thin layer chromatography with developing system acetic acid : ethanol : water (60 : 30 : 10). Alkaloids were detected by UV radiation and spray with Dragendorff reagent. After decantation of fractions with similar spectrum of matters we cleared selected mixture. We obtained two alkaloids; NMR spectroscopy will be used to determine the structures of these alkaloids. Keywords: Eschscholzia californica Cham., benzo[c]phenantridin alkaloids, column chromatography, Alzheimer...

Determination of vitamins A, D and E using sequential injection chromatography method.
Trefilová, Kateřina ; Chocholouš, Petr (referee) ; Sklenářová, Hana (advisor)
The working isntruction was created for separation vitamins A, D and E by means of monolithic column engaged in system sequentional injectional analysis. Following conditions of separation were chosen after performance of optimalization: Analysis at monolithic column 25 mm long Chromolith Flash RP-18e column, 4,6 mm I.D. (Merck, Germany), with speed (10 µl/s) with applied mobile phase acetonitril metanol (1:1, v/v). Monolithic column was in SIA system and was engaged between moreway valve and flow Z cell. Spectrophotometric detection was performed at wave length for absorbing maximum for vitamins A - 324 nm, D - 265 nm, E - 285. Total SIA analysis time was 400 s (from that 230 s lasted separation process). From basic chromatographic validation parameters were evaluated the following symetric factor As, resolution Rs, amount theoretical trays and repeatability. Symetric factor for vitamin A - 1.309; vitamin D2 - 1.302; vitamin D3 - 1.286; vitamin E - 1.158. Peak resolution for vitamins A-D2 je 3.607; D2-D3 4.232; D-E 1.532. Amount theoretical stages for vitamin A 998, vitamin D2 1621, vitamin D3 2091 and vitamin E 2376. Repeatability was worked out from 6 sprays, and relative standard deviation of particular substances was calculated from the highest point of the graph peak. Repeatability was for...
