Development of an UHPLC method for determination of abiraterone acetate and its oxidative degradation

Aim of this thesis was to develop and validate a method for determination of pharmaceutical active substance abiraterone acetate for UHPLC and also to perform forced degradation with 0,3 % hydrogen peroxide and elevated temperatures. Because of poor solubility of abiraterone acetate in water, mixture of acetonitrile of water in ratio of 1:1 was chosen as solvent. Opimized separation was performer on the column Acquity CSH PHENYL-HEXYL (1.7 µm, 100 × 2.1 mm) and its temperature was 45 řC. Mobile phaze was composed of acetonitrile and 0.1 % aqueous solution of formic acid, flow of mobile phase was 0,3 ml/min, sample injection volume was 1 µl and gradient elution was used. UV/VIS detector with diode array was used at the 265 nm wavelenght. Quadrupole mass detector was used as well. For validation of the method repeatability, recovery and limits of detection and quantitation were tested. Calibration dependence was tested in the concentration in range of 0.1-1∙10-4 mg/ml. Forced degradation was carried out on two sets of samples. Hydrogen peroxide of 0.3 % mass concentration was added to one set. Both of the sets were exposed to laboratory temperature and to temperature o 50 řC for 1, 2, 3 days. Study of the amount of antioxidants in extracts from Melissa officinalis

This diploma thesis deals with the determination of selected antioxidants, namely rosmarinic acid, caffeic acid, cinnamic acid and t-4-hydroxycinnamic acid in aqueous and ethanolic extracts from Melissa officinalis. The aim of the work was to find out whether the above- mentioned antioxidants were present in the extracts prepared at home. A new UHPLC method with UV and MS detection was developed to monitor the content of rosmarinic acid, caffeic acid, cinnamic acid and t-4-hydroxycinnamic acid in the extracts. A BEH C18 column (2,1 mm × 100 mm, particle size 1,7 µm) was selected for separation, the mobile phase consisted of methanol (component A) and 0.1% aqueous formic acid solution at pH 2.6 (component B), the ratio of components being changed according to the gradient program. The method was validated and repeatability, limits of detection and limits of quantification, linearity, yield and robustness were determined after optimization. Analysis of 20 aqueous and 20 ethanolic extracts was performer differing in extraction time, type of solvent and lighting conditions. IgG glycopeptides separation using hydrophilic interaction/ion-exchange liquid chromatography

Glycoproteomics is a challenging branch of proteomics because of the micro- and macro-heterogeneity of protein glycosylation. Hydrophilic interaction liquid chromatography (HILIC) is an advantageous alternative to reversed-phase chromatography for intact glycopeptide separation prior to their identification by mass spectrometry. Nowadays, several HILIC columns differing in used chemistries are commercially available. However, there is a lack of comparative studies assessing their performance, and thus providing guidance for the selection of an adequate stationary phase for different glycoproteomics applications. Here, we compare three HILIC columns recently developed by Advanced Chromatography Technologies (ACE) with unfunctionalized (HILIC-A), polyhydroxy functionalized (HILIC-N), and aminopropyl functionalized (HILIC-B) silica with a C18 reversed phase column in the separation of human immunoglobulin G glycopeptides. HILIC-A and HILIC-B exhibit mixed-mode separation combining hydrophilic and ion-exchange interactions for analyte retention. Analysis of the degradation products of the active substances in historical pharmaceutical relicts from 18th and 20th century.

Historical pharmaceutical preparations analyzed in this thesis were a senna extract more than 200 years old, an ointment "Naso-Merfen" 75 years old, and an ointment "Sulfathiazol" 42 years old. The active substances in the analyzed samples were sennosides A and B (senna extract), ephedrine and menthol (the ointment "Naso-Merfen"), and sulfathiazole (the ointment "Sulfathiazol"). The senna extract was analyzed by RP-HPLC and HPLC-MS. Separation conditions were optimized, especially for separation of the sennoside A and B enantiomers. The active substances were not detected in the sample. One degradation product and substances characteristic for senna were identified. Their presence in the historical and contemporary sample was compared. Detailed ESI− -MSn fragmentation mechanisms of sennoside A and B have been proposed. The sample of the ointment "Naso-Merfen" was analyzed by HILIC-UV, HPLC-MS, GC-MS, and AAS. Separation conditions were optimized. The active substances were quantified. Traumatic experiences of children in the context of the Czech republic

The topic of the work is the traumatic experiences of children in the context of the Czech Republic. The work is divided into two parts, theoretical and practical. The aim of the theoretical part is to get acquainted with the influence on behavior and education after experiencing traumatic experiences, describes the typical forms and manifestations that belong to the negative experiences in childhood as is mental, physical abuse, sexual abuse, neglect, but also substance abuse by a household member or suicide of a family member. The theoretical part also draws attention to the possible impact on the child after experiencing traumatic experiences, in the areas of education or behavior. The work emphasizes the importance of prevention of this social problem and presents legislative support, crisis lines, centers and support in the field of education. The practical part is focused on the evaluation of anonymous questionnaires. The aim of this work was to find out the awareness and personal experience of teachers with violence, physical, mental abuse, neglect, sexual abuse in their students. Strategy oriented to support positive behavior at lower secondary school

The bachelor thesis Strategies oriented to support positive behavior at lower secondary school aims to present strategies that these schools use to prevent problem behavior. The theoretical part of the thesis defines the psychosocial development of an individual in the period of early adolescence, the concepts of problem behavior, class climate and the PBIS system. The research sample was created by eight respondents who teach at lower secondary school. Four respondents started implementing some elements of the PBIS system in year 2020/2021. Data for the research were collected by interviews in spring 2021 using Microsoft Teams. In our research we focused on preventive precaution and rulemaking. We found that the most common forms of preventive precaution are training course, classroom hours and correctly set rules. We found that rules have an important place in preventing problem behaviors and that all schools used strategies to support positive behavior. The research helped us to fulfil the goal. Selfconcept of individuals placed in the diagnostic institute

This diploma thesis is focused on individuals placed in a diagnostic institute and their self- concept. The aim was to evaluate and describe their view of themselves, whether they perceive positively or negatively, in which area they evaluate above average and where they move on the edge of below average. We consider the conception of ourselves to be the essence on which it is necessary to work on, build it and try to strengthen it if these individuals are to be corrected and their current behavior changed. The first part of this work clarifies the theoretical concepts related to the issue and the second part includes the research itself to determine the self-concept of individuals placed in a diagnostic institute. The evaluation of self-concept was based on a questionnaire survey (Questionnaire of self- concept of children and adolescents Piers Harris) and analysis of products of mental activity (drawing a tree). The results of the questionnaires show that the respondents from our research sample do not perceive positively, on the contrary, on many scales they show values corresponding to the average, below average to significantly below average. The elements resulting from the analysis of the drawings only highlight and confirm the results of the questionnaires.
Support of social and emotional development of shildren with ADHD in pre-school education

The thesis focuses on children in pre-school educational institutions who are diagnosed with ADHD (Attention Deficit Hyperactivity Disorder), as well as on the methods of supporting their social and emotional development. The thesis consists of two parts, the theoretical and the practical one. The theoretical part deals with the basic information of ADHD, children's development and the methods of working with children suffering from this syndrom. We can find there three main chapters. The first chapter deals with pre-school children's development. The second chapter concentrates on the definition, causes, occurrence, symptoms, diagnosis and treatment of ADHD. Finally, the third chapter focuses on the methods of working with ADHD children, creating a positive environment, integration of these children, general reccomendations for working with ADHD children and on educational methods and techniques of controlling their behaviour. The practical part investigates how these methods and techniques are used by teachers in pre-school educational institutions. The theoretical knowledge gained from literature is completed by practical findings which I have obtained from interwieving five kindergarten teachers. Analysis of amino acids and polyamines by HPLC and CZE

(EN) This work is focused on two specific classes of amines. First group comprises of cyclic polyaminocarboxylates from the DOTA (1,4,7,10-tetraazacyclododecane- 1,4,7,10-tetraacetic acid) family, which are important reaction intermediates in magnetic resonance imaging (MRI) contrast agent synthesis. In the development of new contrast agents, carboxyl groups are very often protected with tert-butyl ester groups. Among these derivatives, tBu3DO3A is of the highest importance. Therefore, reverse-phase high performance liquid chromatographic (RP-HPLC) and non- aqueous capillary zone electrophoretic (CZE) methods were evaluated for qualitative and quantitative analysis of tBu3DO3A (1,4,7,10-tetraazacyclododecan-1,4,7- tris(tert-butylacetate) and two typical reaction by-products, i.e. tBu4DOTA (1,4,7,10- tetraazacyclododecan-1,4,7,10-tetrakis(tert-butylacetate) and tBu2DO2A (1,4,7,10- tetraazacyclododecan-1,4-bis(tert-butylacetate). These optimized methods were successfully applied to monitoring of real reaction mixtures during synthesis of a new MRI contrast agent. No further sample pretreatment was necessary.
