Abstract: The thesis adresses the problem of multiple sequence alignment (MSA). It contains the specication of the proposed method MSAMS that allows to find motifs in biological sequences, to split sequences to blocks using the motifs, to solve MSA on the blocks and nally to assemble the global alignment from the aligned blocks and motifs. Motif search and MSA are both solved using genetic algorithms. The thesis describes the implementation of the method, conguration of its settings, benchmarking on the BAliBASE database and comparison to the ClustalW program. Produkce polyklonálních protilátek proti rekombinantnímu proteinu X viru bramboru

The gene encoding the coat protein (CP) of Potato virus X was cloned into expression vector pMPM-4A Omega. PVX CP was expressed in Escherichia coli and was used for raising rabbit polyclonal antibodies. For immunization we purified CP fractions from bacterial lysate either by simple fractionation or by excision from SDS gel. The obtained sera and antibodies were tested for the detection of PVX in laboratory hosts Nicotiana tabacum, cv. Petit Havana SR 1 and N.benthamiana and in various cultivars of natural host Solanum tuberosum by ELISA as well as by Western blots. The antibodies were specific for PVX and are usable for the detection of the broad isolates spectrum of the Potato virus X by ELISA and Western blot.

Phonological changes between Old and Middle English (an algorithmic approach)

The purpose of this thesis was to create an automatic analyzer of phonetic changes in the historical development of English, namely between Old and Middle English. The analyzer gets an OE and ME form of a word at the input and produces a suggestion of an explanatory sequence of changes connecting these two forms as the output. As sound changes operate mainly on the spoken language, three main subtasks emerge: to create a grapheme-to-phoneme convertor for Old English, the same (although structurally more complicated) for Middle English and a core algorithm that searches for possible sequences that would explain the development of the word between its OE and ME spoken forms. There is a certain regularity in the diachronic phonetic changes, and therefore these can be translated into a set of rules. A general framework is proposed and implemented that works with these rules in a certain formalized fashion that allows their integration within standard algorithms. The form of the rules is largely adopted from regular expressions, with some alterations and additions, the most important being the possibility of using wildcards that are based on phonetic properties. Teplotní účinek na rheologické vlastnosti pryžového prvku

Deformation and temperature data of a vibro-damping element measured in laboratory condition at harmonic loading with constant prload F st =6kN were elaborated with the aim to build termo-mechanical characteristic of rubber-like material. It is shown that the increase of temperature during harmonic loading at various amplitudes F0=(0,75,3) kN and frequencies fe(10,30) Hz is exponential in time and proportional to the square of loading amplitude. The proposed mathematical model of dynamic characteristic of specimen gained by the regression procedure consists of linear, quadratic and fourth root function of displacement amplitude Xo and of the linear correction function of temperature.

The capability of yeast colonies to produce ammonia depends on correct function of SPS sensor composed of Ssy1p, Ssy5p and Ptr3p. The obtained data indicate that SPS sensor influences the expression of amino acid transporter genes in yeast colonies by different way when compared with its function in yeast liquid cultures

Analysis of the Nonprofit Sector and its Potential for Regional Development

Nonprofit sector is regarded to be ever more significant part of a democratic society. Within the European context it is seen as a partner of public and commercial sectors also in relation to the development of specific regions. On the level of local action groups, which apply the LEADER methodology, it is directly defined as an inseparable partner of regional cooperation and development. The aim of the thesis is a detailed survey of nonprofit sector in the area of Local action group NAD ORLICÍ. Additionally, it focuses on analyzing the potential of nonprofit sector for the following development of the region. Several researches were conducted in order to collect as much data as possible. Analyses of publicly accessible databases and information sources as well as of records acquired from the respective specialized institutions were performed. Simultaneously, independent questionnaire surveys were carried out, focusing on representatives of local authorities and representatives of non-governmental non-profit organizations. Both researches were mainly quantitative, having covered also some aspects of activities and cooperation between the non-governmental non-profit organizations and public sector by use of a simple qualitative analysis. The analyse of cattle breeding in an enterprise - case study

This work deals with an analysis of breeding cattle in the agricultural cooperative Agra Březnice u Bechyně. Data on milk production and reproduction was obtained from records of performance tests and livestock records. The results of performance and reproduction in the years 2010 to 2012 shows that the monitored breeding achieved when compared to the average population of Czech Pied cattle bred in the Czech Republic, above-average values. The efficiency of milk production in the reporting period increased by an average of 394 kg. Milk composition is at the population level. The fat content was in the period, on average, 0.02% lower and the protein content was about 0.01% lower than the values of the population in the Czech Republic. The reproduction of purebred breeding is used in a closed herd turnover. The reproductive performance achieves significantly less value than the population average length of service period. The other reproductive indicators, with the exception of insemination index reached in the period better values than the average of these values achieved in the Czech Republic. The results showed that the most common reason for withdrawal from dairy herds are problems with disease of the legs and udder disease. These problems need to be addressed in the short term better approach to monitoring and prevention in the long term appears to be the most important completion of a new barn. The breeding value of bulls used in reproduction was satisfactory level.

Latinisms in contemporary German language and their lexicographic treatment in a bilingual dictionary

A role of Latinisms in contemporary German language is the subject of this thesis. The thesis has been elaborated on the basis of findings acquired in the course of drafting dictionary entries within a project focused on development of the German-Czech Academic Dictionary. The role of the Latinisms within the German language vocabulary is analysed on the background of a confrontation of entries included in the monolingual dictionary Duden-Deutsches Universalwörterbuch with a structuralist theory of centre and periphery of language. The analyses, elaborated on the data obtained from the Mannheim German Reference Corpus (DeReKo) and the collocation database (CCDB) developed on the basis of the Corpus, are used for the purpose of the confrontation with the above- mentioned premise of the central or peripheral position of Latin loan words.

Terminology problems in myofascial trigger points concept

Title: Terminology problems in myofascial trigger points concept Objectives: The main aim of this work is to organize information in the field of myofascial pain syndrome and myofascial trigger points, and to show unclearness in using terminology. Another aim is to describe trigger points from the view of the etiology, the diagnosis and the therapy. Subsidiary aim is to evaluate the relation among trigger points, tender points and acupuncture points. Methods: Design review was used in this thesis. Data was captured on the base of self studying available sources. Collecting available information from databases specialized in medicine and complementary medicine. Results: The presentation of myofascial pain syndrome and myofascial trigger points data obtained from available sources. Introducing these information in terms of terminology, etiology, diagnosis and therapy as well as a brief evaluation of relation among myofascial trigger points, tender points and acupuncture points.
