Zirconia-based and Chiral Stationary Phases in Pharmaceutical Analysis
Kučera, Radim ; Sochor, Jaroslav (advisor) ; Blešová, Marie (referee) ; Pospíšilová, Marie (referee)
Disertační práce Summary High performance liquid chromatography achieved during its existence enormous boom. Modern, PC controlled and user-friendly chromatographic devices allow performing a number of tens analyses on one apparatus daily. There is possible to analyse wide spectrum of compounds from non-polar to polar and from low molecular to high molecular analytes. Appropriate choice of chromatographic column, mobile phase and detection technique makes the analysis of complicated mixtures possible and allow obtaining both qualitative and quantitative information. HPLC is due to these characteristics utilized not only in the field of pharmacy but also in other areas e.g. biology, medicine, industry, environment monitoring and quality control of various products. Separation column is the most important part of the chromatographic system. The availability of stable, high performance chromatographic phases is the essential requirement for the development of a rugged and reproducible method. Nowadays, most HPLC separations have been done in the reversed-phase mode on the silica-based columns. This phenomenon is due to many positive properties of silica - mechanical strength, well mastered manufacture of silica particles, easy modification of silica surface, compatibility with water rich mobile phases etc. The...

Epigenetic regulation of HLA class II genes and their role in autoimmune diseases.
Čepek, Pavel ; Kotrbová - Kozak, Anna Katarzyna (advisor) ; Horníková, Lenka (referee)
Abstract Background: Type 1 diabetes (T1D) is a multifactorial autoimmune disease. Its incidence in Europe is continuously rising. The highest T1D risk is associated with HLA (human leukocyte antigen) class II genes. HLA class II molecules play a key role in regulation of immune response. They contribute to the selection of T cell repertoire by presenting antigenic peptides to the CD4+ T lymphocytes. HLA class II expression is controlled by regulatory module that is situated 150 - 300 base pairs upstream of the transcription- initiation site in all HLA class II genes. Polymorphisms in this region are linked to some autoimmune diseases. There were identified several promoter alleles (named QAP) in the HLA DQA1 gene promoter region. Most of the polymorphisms appear to be conserved within haplotype. Individual QAP alleles may have a different promoter strength by which they influence expression of HLA DQA1 gene alleles. Promoter strength can be modulated by DNA methylation. Aims:Our aim was to define methylation profile of HLA DQA1 promoters and determine the mRNA expression of individual alleles of HLA DQA1 gene in T1D patients. The mRNA expression level of HLA DQA1 gene alleles was determined using quantitative PCR. Methods: 30 diabetic pacients (age range 21 to 76 years), were included in this pilot...

Adaptation of new employees in the SIEMENS Ltd. company
Havlíková, Karolína ; Hubinková, Zuzana (advisor) ; Lorencová, Hana (referee)
One of the most important factors of the prosperity in each company is people -- its employees. It is well known, that when the employees are unsatisfied, they don't use so much effort in their work or in a worse case they loose interest to stay in the company and they start to seek for another one. Remarkable influence on stabilization of employees in a company certainly has their successful adaptation to the new employment. This diploma work focuses on an analysis of the adapting process in the company Siemens Ltd. Using a questionnaire there were inefficient components of the contemporary programme for adaptation of new employees discovered and recommendations for the management of Siemens Ltd. were proposed.

Transdermal in vitro permeation of terbinafine II.
Doubravová, Hana ; Doležal, Pavel (advisor) ; Musilová, Marie (referee)
Rigorosal Thesis 2006, Doubravová Hana ¨ ABSTRACT Thesis provides a detailed overview of the current perspective on the performance and state of stripping methods in the study of penetration of substances through the skin. Using the pre-certified protocol of stripping experiment were found that part of terbinafine penetrates through the epidermis to the dermis. For both single vehicle evaluated, the ispropylmyristate and isolane, this results are quantitatively comparable with those obtained for the preparation Lamisil cream. Arbitrarily chosen, the addition of 1 percent of transkarbam 12 in isopropylmyristate and/or isolane yielded in significant increase of amounts of terbinafine deposited in the dermis.

Earlier evolutionary dead-ends, now the creators of a reproductive strategy: the origin and reproduction of the all-male water frog lineage Pelophylax esculentus
Doležálková, Marie ; Choleva, Lukáš (advisor) ; Mikulíček, Peter (referee) ; Kratochvíl, Lukáš (referee)
Asexual modes of reproduction are usually based on the principle of copying (cloning) DNA from the female and passing it on to the offspring. For most asexually reproducing vertebrates the progeny develop from an unreduced and often unfertilised egg. This is driven by the mechanisms of parthenogenetic and gynogenetic reproduction. While in the former the clonal germ cell develops spontaneously and separately, in the latter a sexual partner is needed to activate the cleavage of the ovum, although without the fusion of the sperm and egg. Therefore in both cases there is no fertilization and the clonal progeny consist solely of daughters, hence the majority of previous studies have only focused on asexual female lineages. However, on rare occasions asexual clonal males can arise when the right fertilization occurs. Whilst these offspring are usually infertile, fertile diploid asexual males have been discovered in just three genera of hybrid origin in vertebrates. One of these unique cases is the European water frog complex of the genus Pelophylax, whose distribution includes the Czech Republic. In areas around the upper Odra River populations of hybrid males were recently discovered who form stable all-male lineages, similar to those formed by asexual females. The results of this study show that males produce...

Personalities and approaches in czech economy - foundations for social economy
Králík, Tomáš ; Dohnalová, Marie (advisor) ; Šloufová, Romana (referee)
The abstract This work deals with the origins of Czech social economy. Its aim is to discover whether some of its characteristics are to be found in the theoretical works of Czech economists from the year 1848 to 1948. As there is no definition of social economy in the Czech environment, it was necessary to employ the foreign definition according to the International Research Network EMES.

Biopower and autonomism
Poppr, Martin ; Petříček, Miroslav (advisor) ; Kouba, Petr (referee)
Martin Poppr : Biopower and autonomism (abstract of bachelor thesis) The main subject of the thesis is the relationship between an individual and the society. My goal is to find out the origin of the social authority, society cares, and the society control. My work will as well question the relationship between the knowledge and the power, and if both of them are in some way relied to the violence. My work will be based in the philosophy of Michel Foucault, his archeology of knowledge and his studies of the society. I will try to find out how is the knowledge defined by its own discourse. The discourse is a special form of a social power. I will focus my work on the question, how is every human being influenced by the social power and the power of discourse. I will also stay focused on the topic reading few more authors. I will follow Gilles Deleuze developing Foucault's concept of "bio-power", leading us from the age of "Enlightenment" to the present. Foucault himself finds as a good way, how to study the power, to study the forms of the political resistance. That is why I will also focus my attention on the philosophy of "autonomism".
