Current problems concerning administration of uninsurable allowances at regional (district and municipal) office

The basic mission o f the state is to create the conditions for the stability and to regulate the social climate, to absorb the social tension and by the development o f social activities to stabilise the situation in the society in order to enable the social subjects fully develop, affect and realise their targets by their own forces. These efforts intend to establish the conditions for the social defensibility of the social reform and/or minimize its impacts. In order to assure this target, the state disposes by the wide system of legal, financial and institutional instruments. The aim o f this thesis is the global evaluation of the actual legislation on the security of the people in socially indisposed situation; this people enters under the application of the Act No 117/1995 Coll., on social state aid, Act No 108/2006, on social services, Act. No 110/2006 Coll. about the existential minimum and Act No 111/2006 Coll. on aid in material necessity. When I evaluated the practical problems with the realisation of these systems I have resulted from the experience o f the employees of the Employment office in Tábor and of the Department o f social affairs and public health o f the Office o f region South Bohemia. Anti-Plague texts in the late Middle Ages and the Early modern period.

The theme of this work are late medieval and early modern time anti-plague treatises and official regulations against plague. In times when many countries and even continents were affected by plague, many city councils and rulers themself had the treatises printed in order to reduce the risk of transmission of the infection and development of the illness. The treatises adjusted regulations of towns and villages. These official decrees against plague were supplemented with plague treatises that were written for inhabitants of towns affected by black death. Many of them were written in Latin and therefore incomprehensible to uneducated people. It changed when a doctor Jan Černý wrote such a treatise in Czech language. The treatises advised not only how to prevent plague but also how to treat it. I divided my work into two parts. In the first part I describe particular plague epidemics that took place especially in Bohemia and Moravia. I progressed chronologically from antiquity to the 18th century and described whole course of epidemics in various regions. In the second part I focused mainly on different ways how these treatises deal with prevention and treatment of the disease. I made a comparison of various plague treatises from Prague, České Budějovice and Nepomuk. In order to gather data and informations I focused on historical sources and literature that give us informations from people who had first-hand experience of plague. I do the overall comparison and critique of these sources in seperate chapter. I believe this work can work as an educational material for students and teacher

Change of heat source in a heating palnt 2.5 MW

The scope of this Master´s thesis is estimation of changing of heat source in a heating plant 2.5 MW. In the present heating plant burns natural gas, placing of the heating plant makes it possible development source of burning biomass. The scope of thesis is establish with regard to developed capital and operational costs, next effects to environment in scene of heat source, measuring and regulation of technological unit, determination of acceptable size of fuel depot and composition of fuel and ashes economy. The results of this Master´s thesis can be used as a basis about determination toward investment.

The role of ATM in breast cancer

Incidence of breast cancer is continuously increasing in the Czech Republic. Tumor development is a result of gene alterations'accumulation, particulary associated with genes involved in regulation of cell growth and division. Hereditary carcinomas account for approximately 5-10% of all breast tumors and in 60-80% cases are caused by a germline mutation in the major predisposition genes BRCA1 and BRCA2. Nevertheless, other genes, mostly of lower penetrance, may play a role in breast pathogenesis such as the ATM tumor-suppressor gene. ATM is the apex of the repair pathway of DSB. This protein kinase activates through phosphorylation of its substrates cell cycle checkpoints, which leads either to the delay of the cell cycle progression until DSB are repaired or to the promotion of apoptosis. To sum up, the ATM gene seems to have a role in breast cancer development in a minority of the high-risk families in our population which is significantly lower compared to BRCA1/2 and it also seems to be involved in pathogenesis of sporadic breast cancer. Despite the ATM gene's length, we do not perform the preventive screening of this gene in breast cancer high-risk families. Nevertheless, we offer the molecular diagnostics of ATM to ataxia telangiectasia patients.

The thesis deals with the functioning of depositories in the enviroment of the Czech market from the perspective of legislative and practical. The introduction is devoted to directives that make up a harmonized legal framework and the Czech legislativ, which specifically prescribes behaving of depositories. The main part is devoted to the depositary functions, such as operating in the Czech market conditions. This part of work is focused not only on the distribution of the depositary market, but also on the collected amount of the fees, the efficiency of regulation in the real market enviroment and in the importace of depositories in the Czech market. The final section is devoted to the findings, including the final assessment of the depositaries. As a first of its kind, this thesis gives a comprehensive view of activity of depositories and their importace for the Czech market, as the depositaries represent entities that control whether funds act in accordance with the legislation. Depository oversees as to prevent fraud and to qualified investors or collective investment contributed to the further development of the capital market, which is necessary for the further development of all economic sectors.

Cloning and characterisation of selected Class II formins

Formins are proteins involved in regulation and construction of actin filaments of eucaryotic organism. They parcipitate in regulating cytokinesis, polar tip growth, and thus participate in development of whole organisms. There are 2 classes of formins in Arabidopsis thaliana. Both classes include FH1 and FH2 domains (formin homology 1 a 2). Class I formins have N-terminal transmembrane domain, unlike class II formins. Some formins of class II have a N-terminal PTEN domain (Phosphatase and Tensin Homolog). Sequence analyses predicted membrane binding via phosphatase or C2 subdomain of PTEN. This thesis was focused on the formin AtFH14, specifically its PTEN domain. Based on predicted sequence, a DNA fragment encoding the PTEN domain was amplified, sequenced and cloned to destination vectors for YFP and EOS phusions. Marked protein was visualized by transient expression in Nicotiana benthamiana. Stably transformed Arabidopsis lines were prepared for stably expression of protein. The tagged protein was localized in cortical cytoplasm, cytoplasmatical strands, probably in nuclear membrane or perinuclear cytoplasm, as well as in peculiar "folicle-like" structures that might be due to binding of PTEN at the periphery of some membrane organelles. Biological activity of plant metabolites. XVII. Influence of alkaloids from different plant taxons on acetylcholinesterase.

Within the frame of the rigorous thesis, a screening of alkaloids from some botanical taxons used or toxikologically significant in Europe (and also in traditional Chinese medicine) on inhibition of acetylcholinesterase (AChE) was performed. Using a suitable extraction method the summary extracts from morfological parts of Laurus nobilis L. (leaf), Buxus sempervirens L. (sprigs and leaf), Lycopodium clavatum L. (aerial parts), Nelumbo nucifera Gaertn. (stamens and bulb), Fritillaria thunbergii L. (tuber), Fritillaria cirrhosa L. (tuber), Stemona spp. (root) a Stephania tetrandra S. Moore (root) have been obtained. Individual extracts were prepared by digestion and further analyzed by TLC on silica gel in neutral and basic system for alkaloid content (Dragendorff-reactant). Inhibitory activities of the extracts and of the standards (physostigmine, galanthamine) were subsequently determined by means of autobiographical method using TLC (silica gel, only neutral developing system). Corruption and public contracts in the field of ICT

The bachelor thesis deals with the problem of corruption and public contracts in the field of ICT. The work focuses on the corruption situation in the Czech Republic, generally on corruption and laws relating to public contracts. The goal of this project was to unify the information on public procurement and to demonstrate the corruption situation in the Czech Republic on particular examples. The work presents to the analysis of the individual contracts uncovering of corruption in these cases and making proposals to improve the situation in the form of amendments to the Public Procurement Act. Meeting these goals required the search for information on suspect contracts on the Internet, browse closely the Public Procurement Act and the knowledge of the author's own experience. In the introductory part the author defines the concepts of law in the field of public procurement, laws and regulations dealing with public contracts, the authority supervising the Public Procurement Act, procurement processes in various types of contracts and the work of anti-corruption police departments. In the following section of the theoretical part, the author discusses the general problem of corruption and corrupt practices in the field of ICT .The analysis of Public Procurement Act is the last chapter of the theoretical part of the work. In the following practical part, the author discusses several examples of contracts that have been or have not been proven corrupt or illegal and compares expenses of the ICT projects in the private and public spheres. In the last chapter, after obtaining all available information, the author proposes several ideas for improving The Public Procurement Act The author's contribution of the work lies in a clear summary of information, referring to the errors in the procurement processes and then in the analysis of the situation of the "suspicious" cases and detection of illegal conduct, in these cases. The author came to the knowledge that allow the conclusion to summarize in a few paragraphs incentives for the development of amendments to applicable laws of the relevant authorities.

The role of p38-Mapk14/11 as an enabler of primitive endoderm (PrE) maturation and as a sensor of metabolism during mouse preimplantation embryo development

ANNOTATION This thesis describes the role of p38 mitogen activated kinases alpha/beta, also referred to as Mapk14/11 (p38-Mapk14/11), in regulating primitive endoderm (PrE) cell-fate and its sensitivity towards exogenously supplemented amino acids during mouse preimplantation embryo development. This study utilizing various pharmacological inhibitors, gene transcript over-expression and immunofluorescence assisted confocal microscopy imaging, has revealed a novel and pivotal role p38-Mapk14/11 in promoting germane maturation of the blastocyst stage inner cell mass (ICM)into the defined epiblast (EPI) and PrE and its potential requirement under amino acid deprived conditions.

Introduction to bio art

Bio art is a contemporary art practice that works with living organisms (except humans) or is created in cooperation with living organisms. It is relatively new art movement that is rooted deeply inside the history of evolution, domestication of animals and plants, technological discoveries, their development and science. Never before was art questioning its ethics and aesthetics in such profound, molecular and sub-molecular way. In this work I will consider the historical influences and conditions that led to current developments in the field. I will also try to explain the problematics of defining what bio art is and try to clarify the associated terminology. Most relevant and ground breaking works in the field will be mentioned as some new recent developments. Aesthetics of bio art is also an important subject often set aside by the ado around ethics of it. It is an extremely important subject to be considered when talking about bio art and I will try to sketch out a brief outline of it with references to philosophy and contemporary bio art theory.
