Chiral analysis of beta-alanyl-D,L-tyrosine and its derivatives by capillary electrophoresis with 2-hydroxypropyl-beta-cyclodextrin stereoselector

A new capillary electrophoretic method was developed for separation of enantiomers of antimicrobial dipeptide betha-Ala-D,L-Tyr and its derivatives using 50 mM Tris-phosphate, pH 2.50, as background electrolyte and 2-hydroxypropyl-betha-cyclodextrin at concentrations 20 – 60 mg/mL as chiral selector. In addition, association constants of complexes of the enatiomers of dipeptide betha-Ala-D,L-Tyr and its derivatives with 2-hydroxypropyl-betha-cyclodextrin chiral selector were determined.

Study of single nucleotide polymorphisms in candidate genes for Crohn's disease

Mgr. Petra Dušátková, Studium jednonukleotidových polymorfizmů v kandidátních genech pro Crohnovu chorobu Abstract Crohn's disease (CD) is ranked among inflammatory bowel diseases. The etiology of the disease is not completely understood yet. However, it is assumed that genetic predispositions may play important role in the etiology of CD. This work is a part of a project studying causality of single nucleotide polymorhisms within candidate genes for CD in the Czech population. We tested in total 333 patients with CD and 499 healthy subjects for SNPs: c.3020insC, p.Gly908Arg, p.Arg702Trp within the gene CARD15, g.-308G>A within the gene TNFA, p.Arg381Gln within the gene IL23R a p.Ala197Thr within the gene ATG16L1. For determination of the genotypes we used alellic discrimination in TaqMan sonds format. Variants c.3020insC, p.908Arg within the gene CARD15 were significantly associated with CD (OR = 4,4; 95%CI 3,0 - 6,4, OR = 2,7; 95%CI 1,4 - 5,0, respectively). SNP p.702Trp was associated with CD after adjustation for other two polymorphisms within the gene CARD15 (OR = 1,7; 95%CI 1,0 - 2,7). We found the protective effect of the p.381Gln in the gene IL23R (OR = 0,6; 95%CI 0,3 - 1,0). Variant p.197Ala within the gene ATG16L1 increased the risk of CD (OR = 1,3; 95%CI 1,0 - 1,9). The diploma thesis "Sudetendeutsche Landsmannschaft in the 21st century in the Czech republic and Germany" deals with the position and influence of this organization on present Czech-German relationship. In the theoretical part of this work there is described the creation, structure, aims and needs as well as present problems of Sudetendeutsche Landsmannschaft. Further in the text is the analysis of the position of this organization in the Czech Republic and in Germany. The cooperation of the representatives of Sudeten Germans with German political parties and the attitude of Czech and German governments towards this association is the main part of the second - 4 - chapter. The last chapter deals with the influence of Sudetendeutsche Landsmannschaft, especially of sudetendeutsche question, on the selected periods of Czech-German relationship. Strained elections in 2002 and complicated entry of the Czech Republic to the European Union which was caused by the attempts of the Sudetendeutschen representatives to violate the Beneš Desrees approved that Czech-German relationship on the government degree can be cooled under some circumstances. With the significant potential of microalgae as a major biofuel source of the future, a considerable scientific attention is attracted towards the field of biotechnology and bioprocess engineering. Nevertheless the current photobioreactor (PBR) design methods are still too empirical. With this work I would like to promote the idea of designing a production system, such as a PBR, completely \emph{in silico}, thus allowing for the in silico optimization and optimal control determination. The thesis deals with the PBR modeling and simulation. It addresses two crucial issues in the current state-of-the-art PBR modeling. The first issue relevant to the deficiency of the currently available models - the incorrect or insufficient treatment of either the transport process modeling, the reaction modeling or the coupling between these two models. A correct treatment of both the transport and the reaction phenomena is proposed in the thesis - in the form of a unified modeling framework consisting of three interconnected parts - (i) the state system, (ii) the fluid-dynamic model and (iii) optimal control determination. The proposed model structure allows prediction of the PBR performance with respect to the modelled PBR size, geometry, operating conditions or a particular microalgae strain. The proposed unified modeling approach is applied to the case of the Couette-Taylor photobioreactor (CTBR) where it is used for the optimal control solution. The PBR represents a complex multiscale problem and especially in the case of the production scale systems, the associated computational costs are paramount. This is the second crucial issue addressed in the thesis. With respect to the computational complexity, the fluid dynamics simulation is the most costly part of the PBR simulation. To model the fluid flow with the classical CFD (Computational Fluid Dynamics) methods inside a production scale PBR leads to an enormous grid size. This usually requires a parallel implementation of the solver but in the parallelization of the classical methods lies another relevant issue - that of the amount of data the individual nodes must interchange with each other. The thesis addresses the performance relevant issues by proposing and evaluation alternative approaches to the fluid flow simulation. These approaches are more suitable to the parallel implementation than the classical methods because of their rather local character in comparison to the classical methods - namely the Lattice Boltzmann Method (LBM) for fluid flow, which is the primary focus of the thesis in this regard and alternatively also the discrete random walk based method (DRW). As the outcome of the thesis I have developed and validated a new Lagrangian general modeling approach to the transport and reaction processes in PBR - a framework based on the Lattice Boltzmann method (LBM) and the model of the Photosynthetic Factory (PSF) that models correctly the transport and reaction processes and their coupling. Further I have implemented a software prototype based on the proposed modeling approach and validated this prototype on the case of the Coutte-Taylor PBR. I have also demonstrated that the modeling approach has a significant potential from the computational costs point of view by implementing and validating the software prototype on the parallel architecture of CUDA (Compute Unified Device Architecture). The current parallel implementation is approximately 20 times faster than the unparallized one and decreases thus significantly the iteration cycle of the PBR design process.

Role of Psb28 proteins in the biogenesis of the Photosystem II complex in the cyanobacterium Synechocystis sp. PCC 6803

The thesis focuses on the role of Psb28 proteins, namely the Psb28-1 and its homolog Psb28-2, in the biogenesis of the Photosystem II complex (PSII) in the cyanobacterium Synechocystis PCC 6803. The aims of this work were to localize the proteins within the cells, and to determine their function. A fraction of both Psb28 proteins was identified in the monomeric PSII core complexes but most proteins were found in the unassembled protein fraction associated with thylakoid membranes. Psb28-1 was mostly detected as a dimer while Psb28-2 as a monomer. Psb28-1 also differed from Psb28-2 by its higher affinity to the PSII core complex lacking CP43 antenna. Characterization of Psb28-less mutants suggested regulatory function of the proteins in PSII biogenesis in connection with chlorophyll biosynthetic pathway. Analysis of preparations isolated using FLAG-tagged versions of Psb28 proteins showed their association with Photosystem II - Photosystem I supercomplexes, especially under increased irradiance, and supported a role of Photosystem I in the PSII biogenesis.

Life management - important concept in psychology

The thesis brings forward an analysis of the phenomenon of life management (LM) and its research. The introduction ponders over the level of knowledge of the theme and its interpretation; the theoretical part deals with comparison of related theories or, to say it in another words, of theoretical foundations of the phenomenon of LM, taking their similarities and differences into account, where the conclusion of the thesis is that the phenomenon of life management is absolutely necessary; and the practical part deals with field research. In total, 125 subjects took part in it. The research combined various methods - interviews, life axes and questionnaires. The participants were asked to answer questions regarding the management of their own life, and the basic characteristics of it. Based on an analysis of categories, a theory of life management was laid down based on the most relevant pieces of knowledge. LM is a complex and individual phenomenon. Obviously adverse external factors, e.g. poverty, unemployment or illness, weren't necessarily signs of inadequate life management in the studied population. This diploma thesis is focused on return on investment associated with the construction of a residential building in Brno. First, the author finds all costs associated with the construction of a residential building. Subsequently, the residential building will be evaluated according, as a complex and as an individual flats. It will also be based on the prevailing prices of rents calculated yield value of the house. Then the comparative method provides the prevailing price of apartments in the area and find out how much revenue would amount to, if the house is sold off for flats. Finally the resulting values are compared among themselves and determine the best option to deal with new residential building in Brno.

A lot of up-to-date industrial technologies associated with treatment of solid and liquid metals are based on thermal and force effects of electromagnetic field. The fundamental process is induction heating of metals that precedes a number of consequent operations such as tempering, drying, melting, stirring, hardening, hot pressing etc. The paper summarizes physical essence of the above processes (that usually represent complex coupled problems) and presents their mathematical and computer models as well as possible methods of their solutions.
