Nanostructured diamond-like carbon coatings were deposited on several different substrate materials (silicon, glass, polycarbonate and steel) in single (13.56 MHz) and dual capacitive r.f. discharges (LF=100-300kHz, HF =13.56 MHz). The objective of our study was to find the optimum combination of the high and low frequency (continuous or pulsed) excitation enabling to cover uneven substrate surfaces and significant lowering of the internal stress in films.

Study of laser mixture in the large pressure region

This work studies discharge plasma in a mixture of gases, similar to that used in the so-called CO2-lasers. This mixture consists of CO2, N2 and He. The effect of the mixture composition and discharge parameters (especially pressure, in the range of 266 Pa - 100 kPa) on the distribution of energy in the vibrational levels of nitrogen was examined. This is important parameter for modelling of the discharge plasma. The effect of the mixture composition on the degree of dissociation of the CO2 molecules was also studied. The relative concentration of CO was applied to find the conditions leading to a minimal dissociation of the carbon dioxide. Measurements of radial profiles were also made. Results from two discharge tubes made from different materials were compared. Two types of discharge were utilized to acquire a wide pressure range - low pressure DC glow discharge in the range of 266 Pa to 1330 Pa and dielectric barrier discharge in the range of 5 kPa to 100 kPa. Both discharges are used in commercial CO2-lasers. We observed a descending dependence of the vibrational temperature on the pressure and a big step caused by increased occurrence of standing ionizing waves in the mixtures with low nitrogen ratio. We present an innovative Bayesian method for estimation of statistical parameters of time series data. This method works by comparing coefficients of Karhunen-Lo\`{e}ve expansion of observed and synthetic data with known parameters. We show one new method for generating synthetic data with prescribed properties and we demonstrate on a numerical example how this method can be used for estimation of physically interesting features in power spectra calculated from observed light curves of some X-ray sources.

Investigation of lightning-generated elecromagnetic waves in a broad frequency range

In this work I present a study dedicated to the penetration of whistler- mode waves to the ionosphere. An algorithm of automatic detection of whist- lers in spectrograms computed from the data measured on the DEMETER sa- tellite is described. A method of causative lightning detected by the EUCLID lightning detection network assignment to a detected whistlers is described. Results of statistical study dedicated to relationship between the detected whistlers and assigned causative lightning. Based on the proccessing of data from 364 passes of the DEMETER satellite over monitored area is shown, that mean whistler amplitude decreases with distance between the causative lightning, increses with causative lightning current and in the evening is ap- proximately three times higher than in the morning. A study dedicated to subprotonospheric whistlers is presented. We found, that subprotonospheric whistler causative lightnings currents are very high compared to that of usual 0+ whistlers.

Study on neuropathophysiological changes in mammalian host caused by bird schist osome infection

Trichobilharzia regenti belongs to a small group of parasitic helminthes localized in nervous tissue of their hosts. Like in other bird schistosomes, repeated penetration of larvae (cercariae) into the skin lead to development of skin allergic reaction in humans (cercarial dermatitis). Soon after the penetration in the skin, cercariae transform to schistosomula which are able to resist the host response and, in case of T. regenti, migrate to central nervous system (CNS). During CNS involvement, the infections of both specific avian and non-specific mammalian hosts can result in neuromotor disorders and even death of the host. The present PhD thesis deals with T. regenti infections of non-specific mammalian host. The first part of the experimental work was focused on antibody reactivity and antigen specificity of sera from mice experimentally infected with T. regenti. Development of antigen-specific IgM and IgG1 antibodies and elevated levels of total serum IgE indicated a Th2 immune response. Cercarial antigens stimulated IL-4 release from basophiles obtained from healthy volunteers. Western blot analysis revealed that IgG and IgE antibodies in the sera specifically recognized antigen of 34 kDa in both homogenate of cercariae and cercarial excretory/secretory products. Fourier transform infrared spectroscopy: application in a study of transient species in discharge and ablation plasma

The present dissertation consists of two thematically related parts. The first one (includes two publications) deals with the study of chemical pro- cesses and spectroscopy of highly reactive particles produced in the discharge plasma. This part includes the analysis of ro-vibronic CN radical transitions in the infrared region and the study of chemical reactions in pulsed dischar- ges. The discharge was used as a tool for research of decomposition of simple precursors (acetonitrile, formamide and BrCN) and the subsequent formation of intermediates and reaction products in plasma. The obtained experimental results were interpreted using a numerical model developed in context of this work and used to simulate the kinetics of the studied systems. The second part (includes seven works) is aimed at high-resolved spectro- scopy of metals in the ablation plasma. A total of six different metals were studied: Au, Ag, Cu, Cs, K and Na. The main motivation for spectroscopic research on metals in the infrared region is to obtain information on atomic metals transitions, which are particularly important for astronomical identi- fication of lines in the spectra of stars and their spectroscopic assignments. Studies of minor capsid proteins of the mouse polyomavirus

Mouse polyomavirus (MPyV) is a small non-enveloped virus. Its capsid consists of 72 pentamers of the major capsid protein VP1. The central cavity of each VP1 pentamer contains one minor capsid protein, either VP2, or VP3. The minor capsid proteins are dispensable for capsid formation, but their presence is required for infection of the host cell, presumably because of their anticipated functions during virus entry. After internalization, MPyV virions traffic to endoplasmic reticulum (ER). VP2 and VP3 have been proposed to function as factors responsible for penetration of ER membranes, which is required for subsequent delivery of the viral DNA into the nucleus, a key step of the early phase of MPyV infection. Three hydrophobic domains were predicted in the sequence of VP2 and VP3. First in the unique Nterminal part of VP2, second and third in the common part of VP2 and VP3. The third domain corresponds to C-terminal VP1binding alpha-helix. It has been previously found in our laboratory, that VP2 and VP3 fused to N-terminus of EGFP, when expressed in mammalian cells, display properties similiar to the wild-type VP2 and VP3, namely affinity to intracellular membranes and high cytotoxicity. Study of Chemical Processes in Titan Atmosphere Initiated by Discharge in Electrode Configuration Like Gliding Arc Discharge

The aim of this work is the study of plasma processes and the synthesis of organic compounds due to electric discharge generated in gas mixture corresponding to the composition of the atmosphere of Saturn's largest moon Titan. This study focuses on the mimic of Titan's atmosphere at atmospheric pressure and ambient laboratory temperature. The chemical composition of Titan's atmosphere is very similar to atmosphere of prehistoric Earth. Many articles have been published with theoretical model-research, and laboratory experiments are the pursuit of their interconnection. The main aim of thesis is the identification of synthesized gaseous organic, amino, imino and cyano compounds by use to various analytical methods such as the PTR-MS, FTIR and GC-MS. The OES and electric measurements were applied to the determination of selected electric discharge parameters. The gaseous products and radicals formed in an atmospheric discharge fed by different mixtures of N2:CH4 (0,5 up to 5 % of CH4) operated in a flowing regime at the total gas mixture flows from 50 to 200 sccm at different discharge currents from 15 up to 40 mA were determined. A part of experiments was carried out with admixtures of CO2 and hydrogen. This first part of results has been obtained using OES in dependence on the gas mixture composition and supplied power. The bands of the nitrogen second positive and the first negative systems, CN violet system and Swan system of C2 were recorded. Besides them, atomic lines H, H, and C (in the second order) were also observed. These spectra allowed calculation of rotational and vibrational temperatures. FTIR in situ analysis of the gaseous products showed presence of various nitrile compounds and hydrocarbons in all experiments. The HCN, C2H2, NH3 were the main products generated in our system. The dependences of their concentrations on various experimental parameters were measured. The other part of this work was devoted to estimate the influence of CO2 traces addition on the reactivity in the gaseous mixtures mentioned above. Besides the main products mentioned above, CO2 and CO were detected and also some more complicated oxygen molecules has been confirmed but not estimated because of FTIR spectra complexity. In the case of hydrogen traces addition into the reaction gas mixture, no other compounds were determined. Impurities of CO2 as well as hydrogen have a great positive influence on the production efficiency of the major generated compounds at all conditions. The more detailed gaseous products analyses were carried out using the in situ PTR-MS. This diploma thesis is dealing with the negative consequences of implementation of information and communication technologies and its penetration into education. The purpose of the thesis is identification and analysis of problems dealing with the intensive use of ICT in education of children in elementary school. The objectives of the thesis were fulfilled using theoretical and empirical research methods. The theoretical methods subsumed study and analysis of scientific literature, articles and study of results of theme related experiments. The empirical research methods contained interview, questionnaire, and students' observation. The conclusion of the thesis contains evaluation and summary of the explorative research results, research findings and recommendations for further education and pedagogical practice.
