Viscosimetry of hyaluronan-amphiphile systems

This bachelor thesis investigates the interaction between the polysaccharide sodium hyaluronate (HA) and some amphiphilic molecules. As amphiphilic molecules the aminoacids lysine and 6 – aminocaproic acids were chosen. The presence of the carboxylic group on HA and the aminogroup on the aminoacids suggests electrostatic interaction between these two compounds. This supposal offers the possibility to modificate HA and uses it as a new type of a carrier of bioactive compounds, for example medicals. The HA of two molecular weights (1.75MDa and 70kDa) were chosen for the study of the interactions with the aminoacids. Solutions of HA and stock solutions of the aminoacids were prepared for their mutual mixing in the various volume ratios. The solutions and the mixtures were measured in three types of environment: pure water, 0.15M solution of NaCl and in phosphate buffer (PBS) of pH 6. The interactions were investigated by means of reometry. Our results show, the presence of interactions between HA and the aminoacids mainly in the system consisting the 1.75 MDa HA and lysine in water. As a result of the interactions, a decrease in the viscosity of the mixtures comparing to the HA solutions was observed.
Draft measures against floods in the basin of a small watercourse Mlýnka

This thesis is concerned with protection against flooding in the basin of a small watercourse Mlýnka. The aim is to get information about runoff in the watershed and the proposal dry reservoir as flood measure. The first part of thesis deals with the survey watershed, getting materials, schematisation watershed, constructing rainfall-runoff process. In the second part is designed dry reservoir. Schematisation has been made using ArcGIS, rainfall-runoff process was done in software HYDROG and drawings have been processed by AutoCAD.
Flexible Network Flow Measurement

This thesis deals with designing the probe used for measuring network flows. It contains theoretical analysis of network measurment topic, description of algorithms and principles used for network flow measurement. Emphasis on the probe architecture lies on efficient indexing algorithm and flow record flexibility, such that user is able to define format of flow record.
Compression of IP Flow Records

My Master's thesis deals with the problems of flow compression in network devices. Its outcome should alleviate memory consumption of the flows and simplify the processing of network traffic. As an introduction I provide a description of protocols serving for data storage and manipulation, followed by discussion about possibilities of compression methods that are employed nowadays. In the following part there is an in-depth analysis of source data that shows the structure and composition of the data and brings up useful observations, which are later used in the testing  of existing compression methods, as well as about their potential and utilization in flow compression. Later on, I venture into the field of lossy compression and basing on the test results a new approach is described, created by means of flow clustering and their subsequent lossy compression. The conclusion contains an evaluation of the possibilities of the method and the final summary of the thesis along with various suggestions for further development of the research.
River regulation with flood protection

This work describes the assessment of the capacity of the river Chrudimka on the river kilometers 87,837 to 93,173. The hydraulic computation was done for flow with periodicity of 10, 50 and 100. The calculation in the HEC-RAS program is described. Based on the results, flood protection is proposed. Flood protection is verified by a control hydraulic calculation The results are outlined in the drawing documentation
Graph theory and its applications

It is often necessary to be oriented in the complicated relations between parts of a unit. This problem can be solved by the graphs’ theory very well. Graph G is an ordered pair (V,E) where V is a non-empty set of vertices (our parts of the unit) and E is a set of two-element subsets of the set V called arcs (i.e. relations between parts of the unit). G= (V,E) The graph aplication is frequently hidden. It is not found in the solution of the problem, but could be used to express and substantiate it quite easily. In my work I will deal with optimization problems of a graph. Problem of the maximum network flow that we solve using directed graph can be an example. As for non-directed graphs, we will be interested in the search of a minimum frame of a graph.
Specifics of the hydromorphological monitoring on small water courses and its improvement

This thesis deals with hydroecological monitoring of watercourses. The HEM – Hydroecological monitoring is part of evaluation of status of watercourses, which is a requirement of the European Water Framework Directive. The content of this thesis is firstly to find out the status monitoring of hydromorphological quality of watercourses in the Czech Republic and a subsequent survey of methodology HEM. The aim of this thesis is comparison with the older methodologies and ways to improve the practical implementation of monitoring on smaller waterways. Measurement and knowledge will be applied to Troubsko creek.
Hydromorphological monitoring of small water course and its evaultaion

This thesis is dealing with the hydroecological monitoring watercourses. The HEM – Hydroecological monitoring is part of the assessment of the status of watercourses, whitch is a requirement of the European Water Framework Directive. The content of this thesis is to evaluate a small stream throught two chosen methods (HEM and Bavarian method). The methods is compare with each other and with the state of the stream in 2008. The aim of the study is to compare the suitability of methods and their application.
Application of graph theory to the design and optimization data network architectures

This masters'sthesis deals with graph theory and utilization of this theory for design and optimization of data network structures. Introduction chapter describes graph theory in general view, i.e. fundamental terms used for graph description, graph distinguishing, etc. Next part describes graph algorithms, for example a shortest path finding. After this I write about actual routing protocols where the graph algorithms are used. Last but one part deals with queuing theory and final part describes practical presentation of using graph theory for design and optimization of data network structure in Matlab programme environment.
Rheology of hyaluronane solutions

Předmětem tohoto studia bylo prozkoumat reologické chování vodných roztoků vysokomolekulárního hyaluronanu. Byl studován vliv zvyšující se koncentrace biopolymeru v roztoku, která se pohybovala v rozmezí od 1 do 3 hmotnostních procent, a také vliv vzrůstající iontové síly rozpouštědla, způsobené přídavkem chloridu sodného, na viskoelasticitu a stabilitu těchto roztoků. Pro obsáhlejší popis viskoelasických vlastností roztoků byla použita, vedle běžných oscilačních měření, také metoda ceepových testů, ze které bylo možno určit důležité veličity, jako je procentuální poměr viskozní a elastické složky vzorku, rovnovážná poddajnost, viskozita při nulovém smykovém napětí a retardační čas. Ty byly následně porovnávány s výstupy z jiných typů měření, jako jsou právě oscilační a tokové křivky, nebo nesly dopňující informace důležité pro detailnější popis viskoelastických vlastností těchto roztoků. Ke studiu stability vzorků během namáhání pak byla použita metoda peak-hold, která ukázala na velmi dobou mechanickou i časovou odolnost roztoků hyaluronanu a naznačila hranice, za kterými už dochází k trvalému poškození struktury a degradaci řetězců hyaluronanu a je s němi proto potřeba při manipulaci s roztoky tohoto biopolymeru pro jejich další použití v aplikacích počítat.
