The subject of diploma thesis is the construction of technical project documentation for building realization - Multifunctional building in the town of. The object has three floors with residential attic and basement garage. The land is in the city center Uherské Hradiště and This is a building in the vacant space between existing objects. Construction is rectangular in shape, roofed with a gable roof and is designed from traditional building materials.

Bachelor thesis deals with drafting project documentation for the new family house Troubelice. Family house is detached and is located in the cadastral Troubelice. The house is located in a new area of the village designated for houses. This is a partial basement building, which has two floors. The building has a rectangular shape and has a flat mono-layer roof with attic gable. House is designed for one family of 4 members. The basement serves as technical facilities and storage areas. The first floor serves mainly as a social part of the house. In the first floor is a garage. The second floor serves as a a quiet zone and there is a balcony. It is a brick construction system. The drawing part is processed in AutoCAD.
Isolation of yeasts from the vineyard and their using in the production of wine

The aim of this thesis was the isolation and identification of yests obtained from the wine berries by different molecular methods. The yeasts for identification were collected from the species of Veltlínské zelené white wine from the wineyard from Milotice, district Hodonín. Identification based on analysis DNA by PCR method and followed by RFLP using restriction enzymes HaeIII, HinfI, HhaI a TaqI. To obtain values lenghts of the restriction fragments was used gel electrophoresis. For getting genetic similarity of analyzed yeasts was used BioNumerics software, that processe the results using cluster analysis using Jaccard´s coefficients and the yeasts were identified by comparing the characteristic lenghts with number of articles and educational databases.

Musk compounds in water biota

Synthetic musk compounds (MUSK) or synthetic fragrances are organic substances commonly used as fragrant constituents of perfumes, detergents, cosmetics and personal care products. These compounds are discharged after use via domestic wastewater and sewage treatment plants to the aquatic environment. The result is accumulation in the environment and occurrence in a food chain especially of aquatic and marine ecosystems. Synthetic musk compounds are also found in surface water, fish tissues and also in body fluid (blood, urine, milk) and tissues of human body. Quantitative detection and studying properties of these compounds is an important activity because we have low knowledge about their fate in the environment (persistence, bioaccumulation) and toxicity. This diploma thesis is focused on three commonly used synthetic fragrances – tonalide, musk ketone and musk xylene. The base of this study was to perform a method optimisation for the determination of selected compounds in water and water biota (fish). The aim was determination of selected musk compounds in real samples (water, fish) and evaluation of the results. Pressurized solvent extraction (PSE) for fish samples and solid phase microextraction (SPME) for water samples were used for isolation of analytes from samples. The identification and quantification of analytes was carried out by high resolution gas chromatography – mass spectrometry.
Isolation of compounds from Cannabis sativa and their biological effect

The object of this thesis is a traditional and also controversial plant Cannabis sativa. The theoretical part is focused on botanical side of this plant and its presently used medical forms. All previously isolated substances and their observed biological activity are described in detail. The experimental part of this thesis describes the isolation and identification of substances of the chloroform extract fraction obtained from this plant. Chromatographic methods were used for isolation – analytical and semipreparative HPLC. Obtained substances were identified using UV and IR spectrophotometry. Then the biological effect of obtained substances was observed on chosen microorganisms.
mRNA isolation of plants

The bachelor thesis includes the information about different methods of the isolation of nucleic acids and focuses on the method which allows isolating of nucleic acids from Chinese lantern plant using paramagnetic particles. It includes the information about the best conditions for gaining sufficient amount of nucleic acid. The emphasis is put on the apparatus which is used for automated pipetting. The aim is to create a program for automated isolation of nucleic acids using paramagnetic particles.

Tato diplomová práce je zaměřena na sestavení přehledu konstrukcí a teplených cest kosmických lodí a kosmických vozidel, které se v současné době používají. Na základě specifických požadavků a standardů jsou vypracovány koncepční návrhy zástavby tepelného spínače do konstrukce družice. V jednotlivých kapitolách jsou popsány určité členy, které se na vedení tepla podílí a jsou důležité pro navrhované koncepty z pohledu konstrukce. Diplomová práce popisuje postupy výpočtu tepelných vodivostí a rozložení působící tíhové síly do míst uchycení pro jednotlivé koncepty. Na závěr provádí hodnocení a výběr potenciálně nejvhodnějšího návrhu.

The subject of the thesis is a price comparison of a variant technical solution of a specific building. The thesis consists of two parts. The theoretical part describes the basic terms used in the processing of calculation and budget, price theory, types of prices and software used for budget. The practical part consists of the proposal of variable technical solutions and their comparison based on the chosen criteria. The conclusion is the evaluation of various variable technical solutions and the selection of the most suitable technical solution regarding the purpose of the building.
Analysis of C. necator genome changes after evolutionary adaptation

The p53 protein is a transcription factor that belongs to the group of the tumour suppressor proteins. p53 functions include cell cycle regulation, interaction with DNA, and responses to damaged DNA that may lead to apoptosis or even senescence may occur. The theoretical part summarizes the latest information about the chemical structure and functions of the protein p53 and allosteric modifications that the p53 protein may possess in cells. It also describes selected methods of isolation and characterizes proteins. The aim of the experimental part was to isolate and characterize -isoforms of the p53 protein, which were isolated from the microorganism E.coli. Protein production was preceded by preparation and transformation of plasmids into production cells. From the production cells, 4 -isoforms of p53 were isolated by affinity chromatography, due to the polyhistidine anchor that the proteins contain. The isolated proteins were subsequently characterized by SDS-PAGE and western blotting. Moreover, the transactivation potential in the yeast system was monitored by luciferase assay under conditions involving the use of various culture media, as well as expressed or coexpressed under the inducible GAL1 promoter and the constitutive GPD promoter.
