The thesis focuses on the development of business activities of the company TV PROBBE s.r.o on a foreign market. The bussiness activities of the company include production, wholesale and retail of pet food and pet-supplies. The theoretical part summarizes the findings which serve as a base for the other parts of the thesis. The analytical part evaluates the current situation of the company's external and internal environment. The last part contains recommendations, the implementation of which should lead to the development of the company's business activities.

This dissertation Barrier-free Work Environment examines and emphasizes some elements, which are projected and will be projected even more into architectural production – architect’s work. Buildings designed specially to create a working environment accessible for disabled people are of a current interest and they will become a very prominent subject of social development in the future, because it will be necessary to react sensitively to the disproportions in human abilities and in the society’s capability to create new workplaces, implement progressive technologies, computer systems, and last but not least, to react to new possibilities of people’s creative invention. This thesis is based on the key definitions for this topic – the terms health and handicap. It is aimed at the ethics of communication in a society and its perception by a person with some kind of disability – handicap. This thesis also analyzes the limits of human possibilities in the context of contemporary and future progressive technologies. The topic of accessible workplaces necessarily derives from the historical development of workplaces, and it indicates the future trends of development. An important part of this thesis is concerned with the conditions for creating a friendly working environment, not only from the aspect of the construction itself, but mainly by the interior design, its flexible furniture, and special work tools not only for the handicapped. This dissertation and its contribution to the scholarly research are based on evaluation of an opinion survey, whose respondents were companies employing handicapped people. The analysis and consequent synthesis of the answers given by the employers of the disadvantaged people brought pieces of information, which are summarized in conclusions and recommendations.

Next generation sequencing in clinical virology: method optimization and it's use for samples with unknown infectious agent

The use of the MinION sequencer (Oxford Nanopore) was tested on samples prepared to simulate infectious samples. The tested procedure is to simulate work with a sample with an unknown pathogen. Therefore, a metagenomic approach was chosen. Three kits were tested: Rapid Barcoding Sequencing, PCR Barcoding and Premium whole genome amplification. Each kit differed in duration, difficulty to prepare and in amplification of nucleic acids. In total it was chosen eight viruses with different genome lengths and with varying types of the genome (5,6 - 152 kb, ss/ds RNA, dsDNA). Ten samples were prepared to simulate different types of infection (respiratory, gastrointestinal tract and urine), and one sample contained pure water as a negative control. Before preparation of the library with Oxford Nanopore's kits, DNase/RNase treatment was used. The viral RNA was transcribed into DNA and in chosen samples were amplificated to reach a higher concentration of nucleic acids. Rapid barcoding sequencing kit detected all selected viruses with the highest number of viral reads (4403) with a length between 100 and 250 nt and quality coverage of viral genomes. Nikolaj Vasilijevič Gogol: myths, phantasy, visions

Fenomén Petrohradu je natolik silný, že dokáže prostřednictvím literárních textů lákat a přitahovat i člověka, který se s ním ve svém životě nikdy přímo nesetkal. Díky své dvoutvářnosti, temnosti střídající se s okázalou krásou, leskem a jakousi reprezentativní lehkostí, si ponechává roušku vzrušujícího tajemství, kterou pouze někdy, velmi zřídka, jakoby koketně poodhaluje, dovoluje nahlédnout do svého tajemného světa a umocňuje tak pouto přitažlivosti. Petrohrad je nepochybně jedním z největších fenoménů ruské literatury. Toto město si dokázalo takřka vzápětí po svém vybudování získat pozornost bezpočtu lidí několika generací. Získalo na jedné straně své obdivovatele a na straně druhé pravděpodobně mnohem početnější skupinu odpůrců. Nezaměnitelným a neopakovatelným způsobem však zasahovalo do života obou. Pro jedny byl Petrohrad svůdnou múzou, nevysychajícím pramenem inspirace, pro druhé byl plíživým, tíživým, temným, neustále pronásledujícím stínem vyvolávajícím neutěšené představy, vize a pocity, peklem, městem samotného ďábla. Do které skupiny však zařadit Nikolaje Vasiljeviče Gogola? Petrohrad pro něho pravděpodobně nebyl múzou v povznášejícím slova smyslu. Nezasvětil mu celý svůj život a už vůbec ho oslavně neopěvoval. Dalo by se uvažovat o určitém druhu inspirace, ale byl jejím původcem doopravdy...
