Entrainment of the circadian system in rodents

The circadian clock located within the suprachiasmatic nuclei (SCN) of the hypothalamus responds to changes in the duration of day length, i.e. photoperiod, differently in the separate SCN parts. The aim of the study was i) to compare the effect of a long and a short photoperiod with twilight relative to that with rectangular light-to-dark transition on the daily profiles of clock gene expression and their protein levels within the rostral, middle and caudal regions of the mouse SCN; ii) to elucidate the dynamics of adjustment to a change of a long photoperiod to a short photoperiod of clock gene expression rhythms in the mouse SCN and in the peripheral clock in the liver, as well as of the locomotor activity rhythm; iii) to elucidate whether and how swiftly the immature rat fetal and neonatal molecular SCN clocks can be reset by maternal cues and iv) to reveal when and where within the rat SCN the photic sensitivity of clock gene expression develops during the early postnatal ontogenesis and to compare it with development of cfos photoinduction. Lurcher mutant mice represent a model of olivocerebellar degeneration. They are heterozygots carrying a mutation in the glutamate receptor 52-subunit gene. The receptor malfunction causes excitotoxic apoptosis of Purkinje cells, extinction of which leads to secondary degeneration of cerebellar granule cells and inferior olive neurons. Lurcher mutants suffer from ataxia and deterioration of spatial learning and orientation ability. The aim of the work was to assess the effect of embryonic cerebellar tissue transplantation and enforced physical activity as well as the effect of their combination on spatial orientation ability in Lurcher mutant mice and to observe cerebellar graft survival in adult and young Lurcher and adult wild type mice. For the graft survival analysis adult wild type mice and young and adult Lurcher B6CBA mice were used. To study the effect of the transplantation and enforced activity on spatial orientation adult and young Lurchers of the same strain were used. The graft obtained from mouse embryos without the Lurcher mutation was applied into the cerebellar area of the host as solid pieces. Control mice were treated only with vehicle. The Magdeburg law in the towns of northwest Bohemia in pre-Hussite period

The beginnings and propagation of the Magdeburg Law in the Czech lands in the Middle Ages are in the centre of attention of historical sciences since the second half of the 19th century. The period of great interest in this theme ends in the middle of the last cen tury and there i s a change in the direction of conservation of older knowledge. A detailed look on notorious development trends in connection with use of different types of sources can open new possibilities for efforts concerning deeper cognition of one of the most important components of the so called medieval transformation. The Magdeburg Law is a phenomenon transgressing borders of contemporary as well as medieval states in Centra!- and Eastern Europe, which is the reason to study it in the context of wider regions. Presented study incorporates three themes. The first chapter is dedicated to the analysis of the initial situation that existed in the thirteenth century in one of the most important cities of northwest Bohemia Litoměřice, the later head of a great family of cities of German town law. Effect of carbamazepine on thyroid hormone homeostasis

8. Summary Introduction: While carbamazepine (CBZ) treatment may affect serum thyroid hormone concentrations it rarely leads to clinically important hypothyroidism. This study was aimed to evaluate an early effect of CBZ on thyroid status in hypothyroid patients with thyroid hormone replacement, as compared with patients without a thyroid disorder. Methods: Twenty-nine patients indicated for CBZ treatment were followed prospectively. Their thyrotropin (TSH), total thyroxine (TT4), free thyroxine (FT4) and antibodies against thyreoperoxidase (TPO- ab) and thyreoglobulin (TG-ab) serum levels were assayed before the start of CBZ medication (150 mg/d increasing to 450 mg/d), and then their TSH, TT4 and FT4 serum levels were assayed at week intervals for 7 weeks. Nineteen patients had no thyroid disorder before CBZ treatment (control group A), whereas 10 patients were treated with L-thyroxine (median 100μg/d ) for hypothyroidism and were stable before CBZ treatment (group B). The relationship of pulmonary function tests and exercise tolerance in patients with chronic obstructive pulmonary disease

Výsledky studie přinesly několik zásadních zjištění: 1. Tolerance zátěže je u pacientů s CHOPN významně omezena, přičemž různé typy zátěže jsou tolerovány značně rozdílně. Hlavní limitací obou typů zátěže však zůstává nemožnost zvyšovat VT (a následně VE) v odpovědi na zvýšený ventilační drive doprovázející fyzickou zátěž. 2. Limitace vzestupu VT (a následně VE) je určována stupněm plicní hyperinflace a její změnou v průběhu zátěže samotné, která se dá při absenci náročné přístrojové techniky nepřímo odhadovat podle hodnot IC nebo IVC. 3. 6-MWT je na rozdíl od ergometrie doprovázen rychlejším nárůstem dynamické plicní hyperinflace, která je odpovědná za významnou poruchu oxygenace. Proto lépe, než jiné testy tolerance fyzické zátěže odráží schopnost běžných denních aktivit.
Early diagnostic of mnestic disorders in neurodegenerative diseases

The ageing of the world's population means that the number of people with dementia, especially Alzheimer's disease (AD), will dramatically increase. That's why there is now a great effort to detect the presence of AD in its earliest predementia stages, in the stage of mild cognitive impairment (MCI), and particularly to identify individuals with amnestic syndrome of the hippocampal type (Ha-MCI - preclinical AD). The aim of our studies was to reveal whether spatial navigation testing could serve as an early biomarker of AD - whether spatial navigation is impaired early in patients with MCI, especially in Ha-MCI patients. We used the human analogue of the Morris water maze, the Hidden Goal Task (HGT), which is designed t! o separate two different modes of navigation, egocentric (body- centred, hippocampus independent) and allocentric (world-centred, hippocampus dependent), using a real space navigation setting called the Blue Velvet Arena (BVA), fully enclosed cylindrical arena, as well as a computer version of the BVA. Our results suggest that spatial navigation is impaired already in patients with amnestic MCI, who are more likely to progress to AD, especially in those with amnestic syndrome of the hippocampal type. The bachelor thesis focuses on a survey of concepts of human nature at the end of 20th century, specifically in the works of Francis Fukuyama. The introductory part descibes general contemporary situation and background. The main part deals with the formation of different natural aspects of human behavior from the perspective of sociobiology such as cooperation, intelligernce, language and morality the way F. Fukuyama interpretes them. It tries to expound how these aspects were formed during the long evolutionary development. Understanding these mechanisms should help us comprehend why there are natural tendencies to bahave the way we do. It also searches boundaries between human nature and cultural environment.
