Phosphorylation of the adaptor protein PSTPIP2 and its role in neutrophilic granulocytes

PSTPIP2 is an adaptor protein of the F-BAR family, which is an important regulator controlling the effector mechanisms of innate immune cells. The regulatory functions of this protein were discovered thanks to the CMO mouse strain, which lost the expression of this protein. As a result of PSTPIP2 deficiency, mice of the CMO strain develop an autoinflammatory disease affecting bone tissue and skin. The main mechanism that drives its pathology is the loss of regulation of the neutrophil granulocyte activity. These cells then produce excessive amounts of the pro-inflammatory cytokine IL-1β and reactive oxygen species. However, the exact molecular mechanism of action of the PSTPIP2 protein is unknown. When the PSTPIP2 protein is activated, it is phosphorylated and interacts with other proteins, which mediate its regulatory function. Interaction partners described so far in neutrophil granulocytes include phosphatases of the PEST family, the lipid phosphatase SHIP1 and the non-receptor tyrosine kinase CSK. In this thesis, we identified kinases from the SRC family as kinases that phosphorylate PSTPIP2. Furthermore, we found that the main phosphorylation sites of PSTPIP2 are tyrosines at positions 323 and 329. Guest House "Na Vyhlídce"

This thesis deals with the project documentation of the accommodation facility. Specifically, it is a guest house situated at the outskirts of Blansko – Ceskovice. Guest house kapacity is 22 beds and 50 seated restaurant. The object is designed as a three-floor with a partial basement. Basement floor will consist of storage, utility room and squash court. In the first floor is located main entrance, reception desk and restaurant with facilities. In the second floor are rooms for guests. The building is walled with saddle roof.

The aim of this Bachelor thesis is to make the orifice flowmeter accessible for laboratory tuition. The introduction of this Bachelor thesis explains and describes the issues of measuring of liquid flow rate. The individual measurement methods and the types of flowmeter sensors are introduced. The practical section of the Bachelor thesis consists of a proposal and an actual realisation of a model of an orifice flowmeter with a computer supported evaluation.

Compensation of geometric distortion of electrophoretic gel image

This master thesis is engaged in problematics of creation and compensation of geometric distortions in 1D agarose electrophoresis. This master thesis analyze the problematics of cause of these distortions and summarize the theory needed for compensation of these distortions. Based on acquired theory and created electrophoretic phantoms, the master thesis contains several suggestions for compensation of incurred distortions. These suggestions are recreated into functions, which are connected into a functional user interface for gel image analysis and geometric distortions compensation.

The thesis pursue to describe each step taken while investigating a traffic accident, as well as provides an overview of processing the facts collected and the measurements taken at the scene. Furthermore, a summary of each type of clue and damages occurring in the causality with traffic accidents is provided.
