HLATKÁ, Tereza
The aim of the thesis was to analyze the influences affecting the fertility of sows (number of piglets/1 litter) in the monitored breeding from the period 2017 to 2023. The highest number of piglets was in 2023, with a total of 15.89 born, out of which 13.53 were born alive (a difference of 2.36 piglets). The lowest number of piglets was in 2017, with 13.78 born in total, out of which 12.34 were born alive. Within the genotype, the highest number of piglets was recorded in the landrace breed sows, with a total of 15.05 born and 13.52 born alive. Conversely, in the duroc breed, the number of born piglets was 10.88 in total, with 9.35 born alive. There was a consistent difference of 4.17 piglets (P<0.05) between the two breeds in both total and alive births. For hybrid sows of genotype YL and (YL)D, there was a difference of 0.88 piglets (P<0.05) in total born and 0.67 (P<0.05) in alive born. The lowest number of total born piglets (14.04) and alive born piglets (12.81) were born to sows in their 1st and 2nd litter. Sows in their 3rd-5th litter showed the most total born piglets (15.25) and alive born piglets (13.48). From the 6th litter onwards, there was a downward trend in the number of piglets, with a significant drop observed in alive born, by 0.55 piglets (P<0.05). Sows inseminated at 250 days old had a litter frequency of 13.12 total born piglets and 12.09 alive born piglets. Sows inseminated at 251 days old had a litter frequency of 14.37 total born piglets and 13.13 alive born piglets, with a difference of 1.25 total born and 1.04 alive born. With the prolongation of sow gestation length, there was a slight decrease in both total and alive born piglets. Sows with a farrowing interval 147 days had a higher number of total born piglets (15.34). With an interval 148 days, the number of total born piglets was 0.48 lower (P<0.05). The same trend was observed for alive born piglets, with a difference of 0.37 (P<0.05) between sow groups. Sows with an interval from weaning to insemination 4 days achieved a higher number of total born piglets (15.31) compared to sows with an interval 5 days (14.88), with a difference of 0.43 piglets. The same trend was observed for alive born piglets, with a difference of 0.34 piglets.
Thermoelectric Power Generation System
Brázdil, Marian ; Nerud, Pavel (referee) ; Pospíšil, Jiří (advisor)
In the last decade there is a rising interest in thermoelectric applications. Thermoelectric generators enabling the direct conversion of the heat into the electricity become attractive. This fact is caused by the demands of environmental operation and saving primary resources. Scientists intensively investigate and develop new materials and structures suitable for these applications. The efficiency of the thermoelectric conversion progressively increases. Unfortunately we have no available materials with sufficient thermoelectric properties which could provide cost-competitive price. Thermoelectric generators seem to be useable devices. For example, in case of the unused waste heat using of the thermoelectric generator can increase the overall effectiveness of the unit despite the low efficiency of the generator. This master thesis deals with the issue of the Peltier modules representing the main part of the thermoelectric generators. The physical principles and structures of the thermoelectric modules and the possibility of thermoelectric power production are described here. In the practical part of this thesis the design of the low power generator utilizing waste heat from biomass boiler Verner A 251.1 is proposed.

Hrudíková, Radka ; Šeda,, Ondřej (referee) ; Bobek,, Jan (referee) ; Velebný, Vladimír (advisor)
Hyaluronic acid (HA) is an important substance, which is mostly used in pharmaceutical and cosmetic industry. This substance is commonly found in the human body. HA is one of the factors contributing to virulence of microorganisms. Some bacterial strains produce hyaluronic acid in the form of a mucoid capsule that encapsulates the cell to protect bacteria against the immune system of the host organism. One of the main producers is the bacterial strain Streptococcus equi subsp. zooepidemicus. Contipro a.s. uses the strain CO4A to produce hyaluronic acid in large scale. The production strain was obtained by random mutagenesis by UV light. The aim of the work was to study changes in the genome, which led to a significant increase in hyaluronic acid production, using DNA microarray and real-time PCR (qPCR). The genome of the strain CO4A was sequenced and compared to reference ATCC35246 [1]. The size of the genome is 2,167,251 bp and 83 relevant variants (59 SNV and 34 indels) have been identified. Variants in coding regions were annotated and amino acid sequence changes were determined. In SNV mutations there was a change in the amino acid sequence in 45 cases. The change was identified in every case of indel mutations. The expression level of selected groups of genes was monitored in both strains by the method of DNA microarrays. A cascade of increased expression level of amino sugar metabolism genes leading to the synthesis of UDP-N-acetyl glucosamine was observed in strain CO4A (the increase in expression level of these genes compared to ATCC35246 was on average 28 %). Subsequently, the expression of selected genes was verified by qPCR. There was no significant difference in the expression level of the has operon genes of both strains. The effect of supplementation of the culture medium with N-acetylglucosamine (GlcNAc), which is one of the precursors of HA synthesis, was also studied by qPCR. A positive effect of the supplementation of the culture medium with external GlcNAc in the CO4A strain has been recorded. Also, the supplementation has positive effect on the yield of HA from the medium (increase in yield was on average by 17 %). GlcNAc has been shown to have a positive effect on the yield of HA in ATCC35246 strain as well (increase in yield was 9 % on average), but no significant changes in the expression levels were found in selected groups of genes in ATCC35246.

Dynamic metabolomic prediction based on genetic variation for Hordeum vulgare
Nemčeková, P. ; Schwarzerová, J.
Hordeum vulgare, like many other crops, suffers from the reduction of genetic diversity caused by climate changes. Therefore, it is necessary to improve the performance of its breeding. Nowadays, the area of interest in current research focuses on indirect selection methods based on computational prediction modeling. This study deals with dynamic metabolomic prediction based on genomic data consisting of 33,005 single nucleotide polymorphisms. Metabolomic data include 128 metabolites belonging to 25 Halle exotic barley families. The main goal of this study is creating dynamic metabolomic predictions using different approaches chosen upon various publications. Our created models will be helpful for the prediction of phenotype or for revealing important traits of Hordeum vulgare.
