This bachelor´s thesis deals with design documentation of the family house with an establishment. Family house is designed for a four person family living. The house is situated in the Nitra region, in the territory of the municipality Belince, located on the parcel 248/19. The building is designed as a brick construction with ISOVER insulation system. The vertical and horizontal load bearing constructions are designed of system YTONG. Family house is covered with a gable roof with inclination of 25°. The construction of the sloping roof is made of wooden components forming the construction framework. The building has two floors. In the first floor there is situated the day part of family house and in extented part, on the front side of the house, is part of establishment.. In the second floor there is the night part of family house – the rooms for the privateuse of the owners.

This Master’s thesis´ topic is “Business Plan Proposal: Photovoltaic Power Plant” and it has been assigned at Brno University of Technology, Faculty of Business and Management. The target is to create business plan proposal based on real business case that would be actually used not only as summary of all the necessary information for the project owner but also as information source for potential investors and other stakeholders of the project. Except the above mentioned, thesis demonstrates the knowledge of the energetic sector’s environment, ability to implement the legislation, use particular managerial tools and to prepare the technical and economical valuation to make decision making easier and to catch the investors´ eyes. The project concerns construction and operation of the photovoltaic power plant in the Czech Republic.
Manufacture of Chandelier Hinge

The master’s thesis presents a design for the technology of manufacture of a chandelier hinge from the material ČSN 42 3005 (Cu99.5) with a sheet thickness of 0.5 mm. Due to the spherical shape of the part and the series 40 000 parts per year, the technology of deep drawing was chosen for two drawing operations, while the redrawing is performed by reverse deep drawing. Due to the nature of the component, the additional technology is shearing. The manufacturability of the part was verified using numerical simulation in the PAM-STAMP software. The forming process is performed using three forming tools on three different presses. For the first, combined tool, an LE 160C eccentric press is used. A hydraulic press ZHO100 is applied in the second tool for the reverse drawing, and finally, an eccentric press LEK160 is applied for the shearing in the third operation. With the selected profit value of 25 %, the final price for the component was set at CZK 104.17. The turning point occurs after reaching 16 248 parts.
Production and characteritzation of extracellular hydrolases from selected moulds

This diploma thesis is focused on study of potential production of extracellular hydrolytic enzymes. The theoretical part deals with characterization of selected hydrolytic enzymes, their catalytic properties, the possibility of extracellular hydrolase production by fungi and their applications. In experimental part production strains Aureobasidium pullulans, Fusarium solani and Phanerochaete chrysosporium were used. Productions of cellulase, amylase, xylanase, lipase, protease and lignin-degraded enzymes (laccase, manganese- dependent peroxidase, lignin peroxidase) were observed. Cultivations were carried out in submersed mode in mineral medium supplemented by waste co-substrates such as wheat bran, corn bran, rice bran and oat bran, sawdust, rice, apple fiber, egg pasta and egg-free pasta. Production of enzymes depended on the substrate type and time of cultivation. The highest cellulase, xylanase and amylase activities were measured in the first period of cultivation (3 to 7 day). Lignin-degraded enzymes and proteases were produced at the end of cultivation (7 to 10 days). Lipolytic activity was detected only in A. pullulans, where the activity increased with time of cultivation. The highest value was determined during cultivation on wheat bran (3.6 nmol/ml.min). The highest xylanase and celulase activity (170.3 nmol/ml.min, 248.0 nmol/ml.min) were determined during cultivation of F. solani on corn bran. The highest amylase activity (111.8 nmol/ml.min) was reported in P. chrysosporium during the cultivation on rice. The highest protease activity (68.0 nmol/ml.min) was determined in F. solani grown on wheat bran. The best producer of laccase was A. pullulans, the highest production was recorded for egg-free pasta (27.0 nmol/ml.min). The maximum lignin peroxidase activity (12.5 nmol/ml.min) was measured during the cultivation of F. solani on egg pasta, while the highest yield of Mn-dependent peroxidase (7.7 nmol/ml.min) was achieved during the cultivation of A. pullulans on wheat bran. Lignin-degraded enzymes behaved as inductive, while the other enzymes were produced in mineral medium too. Activity of cellulase in the mineral medium was in A. pullulans strain higher than in media with waste substrates. Enzymes produced into A. pullulans medium were purified by ultrafiltration, ion exchange chromatography and gel filtration.

The subject of my thesis is a project documentation of kindergarten in the land of the municipality Litovel. It is a two - storey detached house which disposal solution meets the conditions of operation of the building – the education of children aged from three to six years. There are classes for children aged three to four years and technical support for the school in the first floor. In the second floor there are two classes, one class for children aged four to five and second class for pre - school children and children with deferred schooling. Each department has a separate entrance, which is located on the north side. The constructive design of the system is designed from the POROTHERM system. The vertical rim of the structure of the first and second floor are bricked up wiith POROTHERM 44 T Profi dimension of 248 x 440 x 249 mm. Inside walls with POROTHERM 30 Profi, a wall between the rooms, and the food servery is of POROTHERM 30 Aku Profi dimension of 247 x 300 x 249 mm and internal masonry POROTHERM 11, 5 Profi dimension 497 x 115 x 249 mm. In the object there are used SDK pre-walls and ceilings Rigips 150 mm thickness. The ceiling is ceramo-concrete POROTHERM system (carrier beams Pot and cartridge Miako) 290 mm thickness. The vertical structure are obtained from internal and external parties plaster. The roof is designed as a green roof. The windows and the front door are designed in plastic with mahogany colour. Floor coverings are designed with regard to the use or operating rooms - the ceramic floor tiles or vinyl flooring. The operation of the school on the 1 st and 2. floor is linked with monolithic staircase. The outer surface layers are equipped with mineral coloured paint. The Impact of Metabolic Syndrome on the Expression of Inflammation and Cellular Damage Parameters in Relation to the Presence of Psoriasis

Metabolic syndrome is a chronic metabolic and systemic inflammatory disease that is associated with high morbidity and mortality. The increasing prevalence of metabolic syndrome highlights the need for a deeper understanding of the pathophysiological mechanisms and their interrelationship with common comorbidities, including psoriasis. By analysing changes in the levels of selected parameters of inflammation and cellular damage, the knowledge of the pathogenesis of the inflammatory process of both diseases and their combination can be expanded. The results may contribute to effective secondary prevention and primary care interventions and facilitate modification of therapies. Data for the doctoral thesis were obtained from three separate studies, for which a set of parameters reflecting metabolic status, inflammatory processes, and cell damage (oxidative DNA/RNA damage, calprotectin, angiopoietin-like protein 8 /ANGPTL8/, clusterin, elafin, chromosomal aberrations, CRP, and vitamin D) were selected. The studies included groups of people with metabolic syndrome, persons with psoriasis, persons with psoriasis and metabolic syndrome, and healthy subjects. Uvedený příspěvek uvádí historická fakta, minulé a současné aktivity a též hlavní priority činnosti České společnosti biomedicínského inženýrství a lékařské informatiky ČLS JEP v budoucím období. Jedná se o odbornou společnost, která je součástí České lékařské společnosti a zastupuje vůči orgánům státu, a to zejména MZ ČR regulované profese biomedicínský technik, biomedicínský inženýr a klinický inženýr ve zdravotnictví. Za dobu své existence prokázala důležitou úlohu, a to nejenom v zastupování uvedených profesí, ale i v realizaci evropského a světového kongresu, ve vydávání vlastního časopisu a též ve vytvoření procesů, které slouží a budou i nadále sloužit k podpoře uvedených profesí.
Diffusion Behavior of Analyte Molecules in a Nanoporous Matrix Created from Polystyrene Nanoparticles

This study investigates the diffusion behavior ofanalyte molecules in a nanoporous matrix of polystyrene particles.Two approaches, steady-state current and potential stepchronoamperometry were employed to calculate diffusioncoefficients and compare the effect of nanoporous matrices withdifferent diameter of pores and different method ofimmobilization of enzyme. Pores were created with close packingof polystyrene nanoparticles. The covalent and cross-linkingimmobilization were used to immobilize the enzyme.Chronoamperometric measurement was applied to detect analyte.The study found that the presence of nanoporous matricesimmobilized on p-lysine affects diffusion. The potential stepchronoamperometry method showed the improvement ofdiffusion. This study provides insight into the diffusion behaviorof analyte molecules in nanoporous matrices, which can be usefulin optimizing the design of amperometric biosensors.
Breast cancer in women.

This bachelor thesis deals with the topic of breast cancer in women. The work consists of theoretical and practical part. The theoretical part of the thesis first describes the anatomy of the whole breast, symptoms and causes of breast cancer. Not only the diagnosis of the tumor itself, but also the treatment of breast cancer is described in detail. An important point of this part is the education of women midwives about breast cancer, especially in the prevention of this disease. The practical part describes a quantitative research survey. The aim of the work was to find out the awareness of women about breast cancer. Another goal was to find out from what sources women find information about breast cancer and the prevention of this disease. Two hypotheses were established for these purposes. The first hypothesis was: Older women have more knowledge about breast cancer than younger women. This hypothesis has not been confirmed. The second hypothesis was established: Women with higher education perform breast self-examination compared to women with lower education. This hypothesis was also not confirmed. Data collection took place using an online questionnaire, which was completely anonymous. The questionnaire consisted of open, closed and semi-open questions. The research group consisted of 300 women active on the social network Facebook and other Internet servers aged 15 - 75 years. Most women responded between 15 and 24 years. The data were processed together with graphical and tabular evaluation thanks to Microsoft Excel 2007. Furthermore, the data were statistically evaluated using the chi square test and Student's T-test. Graphs and tables were prepared from the resulting answers, which show the answers of the respondents. The research found that women are informed about breast cancer. The most common source of information about breast cancer was the Internet, which was mentioned 163 times. The respondents were divided into two groups according to their age, the younger and older group, where the younger group consisted of women aged 15-44 and the older group consisted of women aged 45-75. Although there were a total of 248 younger women (82.7%) and 52 older women (17.3%), age did not appear to affect knowledge about the disease, so this hypothesis 1 was not confirmed by a statistical test. The research also showed that 196 (67.4%) respondents perform breast self-examination. Of these, 84 are women aged 15-24. On the other hand, 95 (32.6%) respondents do not perform breast self-examination, of which 56 women are also aged 15-24. The concept of breast self-examination is known to 291 (97%) women out of 300 (100%) women and once a month 89 (45%) women perform this self-examination of the breast. A total of 180 (91.84%) women have the impression that they perform this examination correctly. For example, 291 (97%) women knew that mammography was an X-ray method designed to show breasts, and 213 (71%) women knew that it was examined by an insurance company from the age of 45. It will be possible to publish the results of the bachelor's thesis at professional lectures or seminars.
The impact of the activities of LAG Aktivios for the development of the region

The cooperation of the population, non-profit organizations, the private business sphere and the public administration is called the local action group. This is a group that helps from the idea, through its financing to the actual realization. It supports the development of business, takes care of the landscape and develops education. All this contributes to the overall development of the region, which is the main goal of this work. The theoretical part contains a basic definition of concepts related to the development of the region with the help of local action groups of the Czech Republic. Subsequently, the connections with regional development, cooperation of municipalities, the LEADER method or Community-led local development and its strategy are defined. Furthermore, this part provides a comprehensive view of the issue of the influence of the local action group in the development of the region. An integral part describes the influence of LAGs on the development of regional planning and association into the national network and regional associations of LAGs within the country. The practical part brings findings from the questionnaire survey and direct analysis and synthesis of the evaluation of the activities of Aktivios within its contribution to the development of the region. The evaluation of the final outputs is further based on a structured interview provided to me by the chairman of the Regional Association of the National Network LAGs of the Czech republic of the Pilsen Region. The results of the thesis bring proposals for possible changes in the management of LAG Aktivios, Especially in evaluating the results of the position of the LAG for the past programming period.
