Souhrn Metabolický syndrom je chronické metabolické a systémové zánětlivé onemocnění, které je spojené s vysokou morbiditou a mortalitou. Rostoucí prevalence metabolického syndromu akcentuje nutnost hlubšího poznání patofyziologických mechanismů a jejich provázanosti s častými komorbiditami, mezi které je řazena i psoriáza. Cestou analýzy změn hladin vybraných parametrů zánětu a buněčného poškození lze rozšířit znalosti patogeneze zánětlivého procesu obou onemocnění i jejich kombinace. Výsledky mohou přispět k účinným intervencím sekundární prevence a primární péče a usnadňovat modifikaci terapií. Data pro disertační práci byla získána ze tří samostatných studií, pro jejichž cíle byl vybrán soubor parametrů odrážejících metabolický stav, zánětlivé procesy a buněčné poškození organismu (oxidační poškození DNA/RNA, kalprotektin, angiopoietin-like protein 8 /ANGPTL8/, clusterin, elafin, chromozomální aberace, CRP a vitamín D). Do studií byly zařazeny skupiny osob s metabolickým syndromem, osoby s psoriázou, osoby s psoriázou a metabolickým syndromem a osoby zdravé. Celkově se výzkumu účastnilo 248 osob ve třech studiích (122, 85 a 41 osob). Laboratorní stanovení vybraných parametrů bylo prováděno metodami ELISA, EIA, nefelometrií a mikroskopickou analýzou (testem chromozomálních aberací). Následně bylo... Metabolic syndrome is a chronic metabolic and systemic inflammatory disease that is associated with high morbidity and mortality. The increasing prevalence of metabolic syndrome highlights the need for a deeper understanding of the pathophysiological mechanisms and their interrelationship with common comorbidities, including psoriasis. By analysing changes in the levels of selected parameters of inflammation and cellular damage, the knowledge of the pathogenesis of the inflammatory process of both diseases and their combination can be expanded. The results may contribute to effective secondary prevention and primary care interventions and facilitate modification of therapies. Data for the doctoral thesis were obtained from three separate studies, for which a set of parameters reflecting metabolic status, inflammatory processes, and cell damage (oxidative DNA/RNA damage, calprotectin, angiopoietin-like protein 8 /ANGPTL8/, clusterin, elafin, chromosomal aberrations, CRP, and vitamin D) were selected. The studies included groups of people with metabolic syndrome, persons with psoriasis, persons with psoriasis and metabolic syndrome, and healthy subjects.
