Characterization of adsorption properties of probiotic bacteria

The aim of the diploma thesis is to characterize the adsorption properties of probiotic bacteria. Characterization was performed using of designed and optimized spectrophotometric method of determination of adhesion of probiotic bacteria to mucin as the main viscoelastic component of mucus. In order to provide a more detailed description of the adhesion interactions involved in the adhesion of probiotic bacteria to mucin, the viability of selected probiotic bacterial strains Lactobacillus rhamnosus CCM 1825, Lactobacillus plantarum CCM 7039, Lactobacillus delbrueckii subsp. bulgaricus CCM 7190, Lactobacillus acidophilus CCM 4833, Lactobacillus casei CCM 4798, Bifidobacterium breve CCM 7825, Bacillus coagulans CCM 2658 and a potential probiotic strain of Lactobacillus zeae CCM 7069. Moreover, bacterial adhesion to carbohydrates was also determined for all the bacteria tested. The last proposed and optimized technique was dynamic and electrophoretic light scattering characterizing surface properties such as -potential, average size distribution and isoelectric point. From this method, information was obtained on aggregation of bacterial cells and on electrostatic interactions. The combination of these methods was used as a complex tool to characterize adhesion of the tested bacterial cultures as a very specific, sensitive and key parameter of a successful probiotic microorganism that is influence by the multiple effects.
Study on protective effect of 3-hydroxybutyrate and its derivatives

The aim of this thesis is the study of chemical chaperones, as a preservative enzymes before denaturing processes. In the experimental part of the study we investigated the protective effects of five selected potential protective compounds – derivatives of 3-hydroxybutyrate – two model enzymes: lipase and lysozyme. Protective effects of potential chemical chaperones were investigated by dynamic light scattering (DLS), which was determined by the size distribution of the aggregates lipase produced depending on the temperature at the heat-induced denaturation. Further we have been used differential scanning calorimetry (DSC), which was determined by the temperature of the denaturation of lysozyme in the presence or absence of potentially protective substances. The last parameter was to determine the effect of potentially protective agents at different concentrations on the catalytic activity of lipase and determining the residual lipase activity after application of denaturing two factors – the high temperature and freezing. Of all tested structural analogues of 3-hydroxybutyrate was the highest protective effect observed (stabilization of enzyme molecules) with the succinate, which has two carboxyl groups. Conversely, 1,3-butanediol showed virtually no protective activity, indicating that the presence of carboxyl groups on the protective effect of essential. The low protective efficacy butyrate further indicates that it is essential that an effective protectant in addition to the carboxyl group contained as a further functional group – either carboxyl or hydroxyl. More significant protective effect was observed in 3-hydroxybutyrate than in 2-hydroxybutyrate. Interestingly, the effective protectant while at higher concentrations strongly inhibit the enzyme activity of the model enzyme, which is probably related to the solvation and conformations of the protein in the presence of protectant and the availability of the active site for the substrate.
Inflow and Influence: Literature in Times of Bookstagram and Book Influencers

The thesis mostly focuses on Czech Instagram culture of book influencers. It is formed by characteristics of Instagram as a medium and by analysis of the currect situation on the book market. Instagram is shaping a space for bookstagram, bookstagram therefore has to be shaped by Instagram. The text surveys, how bookstagram influences literature, how it affects current reader, where does it come from and what is the nature of its influence. The text also overlaps in the field of book market crisis, erudition and fast reading, book as a photographed object and the aesthetics of reading. Further the thesis is focused on specific profiles of book influencers, it characterizes their rhetoric and aesthetics and how they treat literature. The main aiming of the thesis is created by a question after the influence of mentioned phenomena, what changes happen in literature thank to or because of them and what does the influencer represent for literature and reading culture. The thesis is based mainly on historical overviews of history of reading. In the conclusion a few other related themes are outlined, one of them is, if and how it is possible to do the popularization of literature well.
Possibilities of cognitive development in preschool children

This thesis aims to create an elementary task situation for preschool children, which would allow the development of cognitive functions and facilitate the child's understanding of relationships and connections between objects and phenomena. Task situation also reflects the wider social and cultural context where the process of learning and cognition takes place. Preschool age is a period when shaping the personality of the child and his cognitive apparatus necessary for preparation for school and future life.
Motivation to study french language at the high school

This diploma thesis focuses on the motivation of students at high school to study french language. The aim is to discover how is possible to motivate students in french language teaching. The thesis consists of the theoretical and the practical part. The theoretical part describes motivation to study french from the point of view of her three main components: learning, social environment of school and character of french language. The practical part has research form and explores what motivate students at high school to study french, compares motivation for this language at elementary school contrary to high school and compares english to french at high school. The main explored aspects of motivation in research part were: the reasons to study, measure of motivation, teaching environment, surrounding environment of french language, forms of teaching, social influence and barriers in studying french language. I collected the knowledge for the topic by questionnaire survey at Grammar school in Ústí nad labem, at Grammar school in Děčín, at Elementary school school Nativity in Děčín. "You will know the truth, and the truth will set you free" Freedom in the gospel of John

This thesis deals with the question of the concept of freedom in the Gospel of John and the relationship between freedom and truth. The first part of this thesis is focused on the research of concepts of freedom and truth from the Old Testament to the New Testament, it is a description of historical background in which both terms were formed and transformed. This thesis also includes a detailed exegesis of the text of J 8.31-36, which served as the starting point. Due the fact that notion of freedom is most often found in the New Testament in the Gospel of John and Paul's Epistles, I insert a chapter charting the occurrence and use of the concept of freedom in Paul's epistles. In the main part of the thesis I compare the use of the concept of freedom in the Gospel of John and in Paul's letters.

The bachelor thesis deals with bilateral investment treaties, which create investment environment and ensure investment protection between states. Protection is based on adherence to accepted rules. In the event of a breach of terms of the agreed rules, there are investment disputes which this thesis also discusses. The aim is to analyze and identify bilateral investment agreements and the problems associated with them.

This thesis deals with the issues of trading and the characteristics of the business environment in predominantly Arab countries of the Middle East and North Africa. The aim of this thesis is to provide a comprehensive overview of the information needed for doing business, investing and trading in these countries in successful way, and within the content it focuses on individual aspects of economic, geopolitical, legislative, social and cultural specifics. The attention is also dedicated to supporting factors in deciding to enter this market, meaning the level of infrastructure and human capital. In the end of the thesis, there is an evaluation of the current status of individual countries in the areas of trade liberalization, investment attractiveness and the overall business environment and finally, possible prospects for the successful future development of the region are outlined.
