Czech television "Silvester" - changes in 1978-1998

This thesis The Czech Television "Silvester"(New Year's Eve)- Changes in 1978-1998 is mainly concerned with the Czech television entertainment. In this work has been studied by one of its products- the TV New Year's Eve (the "Silvester"). Using archival materials were the selected programs described and analyzed. Furthermore, the selected years were supplemented by reviews from newspapers. The first part deals with the development of television entertainment, especially in the Czech environment. However, there is described the term "fun" or insight into the beginnings of television in the context of television entertainment. This work also deals with the concept of "cult television New Year's Eve" and the way to the emergence of the traditional New Year's shows. There is also a more thorough look at the important personality of the Czech television New Year's Eve, Vladimir Menšík. The Years 1978, 1983, 1988 and 1993 are described only in terms of Česká and Československá televize, in the last analysis in the 1998 is extended on New Year's analysis of the TV Nova and Prima TV program. This Bachelor thesis focuses on a life story of Albína Palkosková-Wiesenbergerová (1908-2002), newspaperwoman, writer and a political prisoner of the 1950's. The thesis describes Albína's life based on her written memoirs and confronts these with the archived documents to create an appropriate view of her life. One part of the work focuses on the life style of female political prisoners in communist labor camps.
Nesting behaviour and population genetics of solitary bees (Hymenoptera: Anthophila)

My thesis is focused on nesting behaviour and population genetics of solitary bees. These two topics, although seemingly unrelated, bring much new information and thus contribute to the better understanding of solitary bee biology that is still insufficiently known. Although the solitary behaviour represents the necessary original state for the evolution of higher sociality or obligate cleptoparasitism, its role is largely unappreciated. Furthermore, intraspecific cleptoparasitism, which is an alternative and facultative nesting strategy in bees, is a probable antecedent state of obligate cleptoparasitism. Although the obligate cleptoparasitism is a very common strategy in solitary bees, the information about the frequency and the occurrence of intraspecific cleptoparasitism in solitary bees is rare. We studied the nesting behaviour of solitary bees to detect different behavioural patterns that could serve as preadaptations to sociality or cleptoparasitic behaviour and we also focused on the detection and description of intraspecific cleptoparasitism in solitary bees. Intimacy and technology: Smartphone as a best friend?

This paper focuses on intimacy and technology in the context of the modern society. A relationship of these two aspects of society is introduced as mutually influencing, since intimacy structures technology and on the other hand technology has wider impact on intimacy. Because technology is narrowly connected with commerce, it allows commerce to access the intimate and highly personal relationships. Discussing relevant authors, a picture of contemporary relationships is drawn, being presented as contradictory and full of paradoxes, and consequently more difficult to deal with. Tendencies towards relationships, which are sustained by technology, are presented as ideal and in the text are posted as an opposite to the real ones. An analysis of intimacy dimensions demonstrates that the contradictions continue to exist in the modern relationships even though they are experienced through technology. Further on discussions are interconnected in the example of smartphone, which commerce presents as the best friend of the human. In the end the role of commerce is highlighted, since it introduces the relationships being sustained by technology as the natural ones, and by this activity an effort is made to gain better control in intimate relationships. The effect of gut microbiota composition on condition in birds

The aim of this thesis is to summarize present knowledge concerning the effects of gut microbiota composition on condition in birds. The effect of gut microbiota on various aspects of animal physiology is currently intensively studied and the understanding of its biomedical importance is a topical theme. However, the majority of the current research is focused on humans and other mammals, and in birds on agriculturally significant species. Therefore, this thesis focuses also on the applicability of results of agriculturally orientated research on free-living birds' study. The results of agricultural research show a high interspecies and intraspecies variability in the effects of certain bacteria on the condition of the host. That should be taken into account when designing experiments in wild-living species. This thesis summarizes current knowledge concerning the effects of bacteria on condition of birds that was gained in correlational and manipulative studies. Manipulative studies include those in which probiotics or antibiotics were given to birds. The main modes of action of probiotic bacteria are also summarized here. Barriers to injecting drug users related to access social services

The diploma thesis deals with the social stigma of injecting users of illegal addictive substances, specifically it deals with the social stigmas that may occur in connection with the use of addictive substances. Social stigma can result in barriers to the use of social services by injecting drug users. The diploma thesis deals with the obstacles that injecting users of illegal addictive substances encounter when using social services. The work tries to identify, map, and describe these obstacles. The theoretical part of the thesis describes the topic of the social environment, addictive substances, addiction, methods of working with users of addictive substances and obstacles associated with the use of social services and other services. The main chapters deal with theoretical topics related to the topic and title of the diploma thesis. The subchapters then elaborate the main topics in more detail. The empirical part of the work was realized through a qualitative survey, specifically through semi-structured interviews. The research sample was a set of contact center clients in the Ústí Region. 1 Operational and Investigative actions Abstract Presented thesis deals with the topic of operational investigative actions specified by the Criminal Procedure Code, to which they have been incorporated with effect from 2002. The main aim of this thesis is comprehensive description of the essence and of the domestic applicable legislation of these actions, and also the analysis of possible problematic points and deficiencies. Currently, the mentioned Code in its title nine called procedure prior to initiation a criminal prosecution defines types of these actions in an enumerative way. Those types are pretended transfer, surveillance of the individuals and objects and usage of agent. The thesis itself is divided into six chapters. First chapter is focused on definition of these actions, putting them into context with so called supporting operational investigative actions defined by the Police of the Czech Republic Code, and also on the brief history of their usage in the territory of the Czech Republic. In the second chapter there is a summarization of the general conditions concerning usage of these actions, which apply to all operational investigative actions with few exceptions. DEVELOPMENT OF A UHPLC-MS/MS METHOD FOR NILOTINIB DETERMINATION IN RAT SERUM

This bachelor thesis aimed to develop a UHPLC-MS/MS method for the determination of nilotinib in rat serum. The developed UHPLC-MS/MS method was used to monitor the pharmacokinetic release of the active substance in a rat model organism in a project focused on the formulation of a tablet containing nilotinib with a slower release than before. The optimal conditions of the method were as follows. Chromatographic column Acquity UPLC BEH PHENYL 100x2.1 mm, 1.7 μm from Waters. The mobile phase consisted of methanol and distilled water, both with the addition of 0.1% formic acid using gradient elution. The flow rate of the mobile phase was 0.3 mL/min, the temperature in an autosampler 15 řC, the column temperature 40 řC, the analysis time 6.5 minutes, and the injection volume 2 μl. The MRM transition monitored for nilotinib was: 530.2 → 289.10 (Q1 = -26 V; CE = -31 V; Q3 = -20 V) and for nilotinib D6: 536.2 → 295.15 (Q1 = -26 V; CE = -31 V; Q3 = -14 V). The setting of the ion source was as follows: nebulizing gas flow 3 L/min; drying gas flow 10 L/min; source temperature 300 řC; desolvation capillary temperature 250 řC. Alkaloids of Amaryllidaceae family: isolation, structural identification, biological activity. IV

Perennial plants from the Amaryllidaceae family are generally known for their beauty but also like herbs which contain wide range of alkaloids. To these days more than 500 alkaloids have been isolated. Amaryllidaceae alkaloids (AmA) are derivatived from aminoacid tyrosine and divided into nine basic groups. Biological activity of these substances icludes antitumor, antibacterial, antifungal, antiviral, antimalarial activity and some of them are used for treatment Alzheimer's disease (AD). Narcissus cv. PROFESSOR EINSTEIN have been choosen thanks to previous research of summary extract. Twelve alkaloids have been detected by GC-MS and ten of them have been identified (e.g.: lykoramine, pluviine, haemanthamine, pancracine, homolycorine). Due to this diversity of alkaloids and the fact that summary extract has relatively high inhibitory activity (IC50 = 49,99 ± 5,38 μg/mL) against HuBuChE Narcissus have been appropriate for isolation of the alkaloids and for further study of their biological activity. Summary ethanolic extract for gain pure compounds was prepared from 34.3 kg fresh bulbs. Separation was initiated by column chromatography and extract was divided into almost 500 fractions.
