PharmDr. Petr Pávek, PhD Title of diploma thesis: Cellular differentiation analysis using SERCA3 protein expression in precursor B-cell acute lymphoblastic leukaemia In this work our objective was to investigate an interconnection between calcium homeostasis and process of cellular differentiation in precursor B-cell acute lymphoblastic leukaemia cell lines. After using phorbol-myristate-acetate (PMA), a known cell differentiation inducer we studied the expression levels of Sarco-Endoplasmic Reticulum Calcium ATPases (SERCA proteins), calcium pumps which transport calcium from cytosol to endoplasmic reticulum. We used two cancer cell lines RCH-ACV and KASUMI-2 for our experiments. After the induction of cellular differentiation we determined SERCA expression levels by Western blot using PLIM430 and IID8 as anti-SERCA monoclonal antibodies that recognize SERCA2 and SERCA3, two SERCA isoforms. Our results showed that induction of cellular differentiation led only to an increase of SERCA3 expression levels, whereas SERCA2 remained mostly unchanged. Effective Strategies of Forming Legal Consciousness of Upper-Secondary School Students

The presented PhD. thesis deals with effective strategies of forming legal consciousness of upper-secondary school students, and with methods that can be used for developing it. So far, legal scholars have paid only insufficient attention to this topic, even though many law experts have repeatedly pointed out its importance. Moreover, they stress the fact that a more detailed understanding of legal consciousness of citizens and of ways of influencing it can help us better understand how law works within the society. The thesis opens with the theoretical part, which defines the concept of legal consciousness, describes its level captured by the available empirical research and identifies effective strategies, which possess the potential to cultivate legal consciousness of the youth. In the following practical part, the author seeks to verify effectiveness of these strategies by means of student questionnaires, interviews with teachers and observations of their lessons. The theoretical part firstly introduces legal consciousness as the basic concept of the whole thesis. Synthesis of chiral secondary amines useful as organocatalysts

This bachelor thesis deals with the preparation of chiral secondary amines, which are used as catalysts in many chemical transformations leading to enantimerically pure compounds. The emphasis of this work is placed on the comparison of the effectiveness of published syntheses those organocatalysts. The first part focuses on the comparison of efficiency and optimization of two methods of preparation Jørgensen's catalyst, the first approach is based on the Z-protected (S)-proline, and in the second approach as starting material was used. The second part deals with the preparation of other Jørgensen's and Hayashi's types of catalysts, via introduction of various silyl functional groups.

Vaclav Havel and His Philosophical Talent

The thesis deals with the talent for philosophy of a playwright, politician and thinker Václav Havel (born 1936). It examines in what ways is Havel's talent for philosophy active, where it finds its fulfillment and to what degree it has been meaningfully put in practice. In the first chapter the author briefly characterizes the concept of a philosopher and discusses some deceptive notions which are commonly associated with philosophers. The following three chapters reveal Havel's world of ideas and the way Havel works with philosophical topics. The author does so by presenting the themes of responsibility, of the relation between the being and consciousness, and on the problems of the natural world. Havel's approach is always compared with that of other philosophers. In the concluding chapter, Havel's main thinking qualities as well as their limits are described.

Pharmacovigilance in the Toxicological Information Centre

1 ABSTRACT Background The annual drug overdose rates have been increasing exponentially since the 90's worldwide. Toxicological information centre (TIC) represents a valuable source of information for evaluating the trends in the drug poisonings in Czech Republic. Aim of the study The purpose was to analyze the number and trends in the calls concerning poisonings due to central nervous system (CNS) affecting drugs, identify the reasons of medication errors caused by laymen, frequency and consequences of these errors across all age groups and also to analyze the numbers, causes, symptoms and severity of the paracetamol intoxications. Methods During the reference period the data from the enquiries were extracted from the TIC electronic database, discharge reports from the hospital were studied or phone call follow-ups with the patients were carried out to be able to evaluate the outcome of the poisonings. Results In the years 1997-2002 the number of calls caused by poisoning with tricyclic antidepressants and barbiturates decreased (by 366.7 % and 340,0 %, respectively) whereas the calls due to selective serotonin reuptake inhibitors and benzodiazepines overdose increased (by 1347.4 % and 359.8 %). Internet as a source of marketing information

This thesis describes various ways of obtaining information using the Internet in business environment. The first part defines the term information and then continues with its division from different perspectives - the type of media the information is available on, originality, expertise, price and accessibility. The second chapter analyzes the global Internet communications network while focusing on available sources of information - from companies, public registries, state portal, statistical and legal information, business and export support, customer information and last but not least the information from news servers. It also deals with ways of evaluating information sources and obtaining data for Internet marketing research. Third chapter uses the Internet to show examples of obtaining specific information needed when managing a business - legislative information, e-shop statistics, market price evaluation and advertising systems data.
