Determination of caseins in cow milk

The diploma thesis is dealing with design of analytical method for determination of caseins in cow‘s milk. Capillary electrophoresis was used as separative method. The developed procedure was verified on real samples. Theoretical part gives basic information about caseins. Their chemical and physical properties, structure, way of manufacturing, usage in food industry and non-food applications are described. There are further some nutritional factors mentioned, whose application in cattle nutrition can affect milk composition, especially milk protein concentration. Literary part contains summary of capillary electrophoresis methods used for casein determination. Experimental part describes used separative system, procedure of casein isolation from milk and casein content in individual milk samples. Separation of milk caseins was performed in an untreated fused-silica capillary with dimensions of 50 um i.d. x 96 cm total length (71 cm effective length). Phosphate buffer (10 mM, pH 2,5) served as the background electrolyte. Casein from milk was obtained by isoelectric precipitation at pH 4,6 after adding 10% acetic acid. Product was washed with acetone to remove fat and dried. Total casein yield was the highest in bio-milk (27,04 g/l), the lowest in skim long-life milk (22,04 g/l). The diploma thesis has arisen on the basis of cooperation of Institute of Food Science and Biotechnology, Faculty of Chemistry, Brno University of Technology and Research Institute for Cattle Breeding, Ltd., Department of Animal Nutrition Physiology in Pohořelice.
Separation and determination of isoelectric points of GnRH peptides by capillary electrophoresis in conventional and carrier ampholytes-based background electrolytes

Two mixtures of human and salmon gonadotropin releasing hormone (GnRH) derived peptides were successfully separated by capillary zone electrophoresis (CZE) in six classical buffer-based BGEs and by carrier ampholytes-based capillary electrophoresis (CABCE) in the same number carrier ampholytes-based isoelectric BGEs of the same or close pH values. In addition, from the pH dependence of peptide electrophoretic mobilities, the isolectric points of separated peptides were determined.
Chiral analysis of beta-alanyl-D,L-tyrosine and its derivatives by capillary electrophoresis with 2-hydroxypropyl-beta-cyclodextrin stereoselector

A new capillary electrophoretic method was developed for separation of enantiomers of antimicrobial dipeptide betha-Ala-D,L-Tyr and its derivatives using 50 mM Tris-phosphate, pH 2.50, as background electrolyte and 2-hydroxypropyl-betha-cyclodextrin at concentrations 20 – 60 mg/mL as chiral selector. In addition, association constants of complexes of the enatiomers of dipeptide betha-Ala-D,L-Tyr and its derivatives with 2-hydroxypropyl-betha-cyclodextrin chiral selector were determined.
Application of capillary electrophoresis in forensic science: Analysis of fireworks

In all samples of fireworks, K+, Na+, Ca2+ and Mg2+ ions were proved and quantified by the method of capillary zone electrophoresis using water solution of 10 mM 4-aminopyridine, titrated with hydroxyisobutyric acid to the pH 4.3, as background electrolyte and indirect UV-absorption detection at 200 nm. Pb2+ or Ba2+ ions were proved in few samples. The other selected inorganic cations were not proved in any sample of selected fireworks. NH4+ ions were not detected in any pre-blast samples, but they were established in some post-blast (glazed tiles) samples.
Peptide profilig of tryptic digests of water soluble proteins from Bacillus thuringiensis - transgenic and non-transgenic maize species by capillary zone electrophoresis

The tryptic digests of water-soluble proteins from transgenic Aristis-Bt and non-transgenic Aristis and Coventry maize varieties were analyzed by capillary zone electrophoresis (CZE) with UV-absorption detection at 206 nm in several acidic background electrolytes. Resolved tryptic peptides were characterized by effective electrophoretic mobilities and corrected peak areas. Some significant qualitative and quantitative differences in CZE-UV profiling of tryptic peptides were found between Aristis Bt and Aristis or between Aristis and Coventry varieties.
Extraction of water-soluble proteins from Bt-transgenic and non-transgenic maize species and tryptic digests peptide profiling by capillary electrophoresis

Water-soluble proteins extracted from Bt-transgenic and non-transgenic maize varieties were digested by trypsin and analyzed by capillary zone electrophoresis (CZE) in several acidic background electrolytes. The found qualitative and quantitative differences among CZE profiles of tryptic peptide maps can be potentially used to differentiate transgenic and non-transgenic maize species.
Separation and characterization of GnRHs by capillary electrophoresis in classical and isoelectric buffers

Capillary zone electrophoresis (CZE) in classical buffer-based background electrolytes and carrier ampholytes-based capillary electrophoresis (CABCE) using isoelectric BGEs (narrow pH fractions of carrier ampholytes used in isoelectric focusing) have been applied to separation and characterization of synthetic human gonadotropin-releasing hormone (hGnRH) and its fragments. From the measured effective mobilities of these peptides within a broad pH range (3.0-10.0), their isoelectric points (pI) were determined.
