Analysis of antimicrobial peptides in venom glands of bumblebees.
Janechová, Daniela ; Cvačka, Josef (advisor) ; Monincová, Lenka (referee)
The growing resistance of bacteria to traditional antibiotics promotes the interest in finding new substances for their production. Antimicrobial peptides have comparable effect to conventional antibiotics, but a different mechanism of action and they do not provoke bacterial resistance. These peptides were characterized in all forms of multicellular organisms. Hymenoptera venom contains many biologically active substances including antimicrobial peptides. For this reason, this thesis focuses on the acquisition of antimicrobial peptide sequences from selected species of bumblebees (Bombus terrestris, B. hortorum, B. hypnorum, B. pratorum, B. lucorum, B. lapidarius, B. humilis and B. bohemicus). The isolation from the venom glands was performed by high performance liquid chromatography with reversed phases. Subsequent analysis was performed using the methods of mass spectrometry, matrix-assisted laser desorption/ionization with time of flight analyzer and electrospray ionization connected with hybrid linear ion trap analyzer with orbitrap. The sequences for the found peptides were determined by tandem mass spectrometry methods "de novo" and Edman degradation. In this work we characterized 17 sequences of peptides extracted from bumblebee venom glands for which antimicrobial activity was determined...

The Influence of British Investments on Economic Development of Argentine, 1880-1916
Švepeš, Václav ; Skřivan, Aleš (advisor) ; Soukup, Jaromír (referee)
The main task of my thesis was to do research work on the British impact on the Argentine economy during the period 1880 to 1916. The research work was focused on the economic aspects and, in particular, on the export policy of the country that, at the given period of time, was one of the most significant centres of primary products manufacturing. The objective of my research work was to discover the reasons for Argentina's economic achievements in the context of British interests in the area of Río de la Plata. For a better understanding of the chosen topic, I subdivided my thesis into eight time-related chapters that besides the country's export and the income from this trade also addressed the urban development, the industrial achievements as well as the country's infrastructure. The work also considers the Britain's dominant impact, not only on Argentina's economy, but also on the country's politics. Great emphasis was also placed on other important events that took place during the defined period that effected Argentina both during the phases of the country's economic prosperity and the phases of recession. Key words: Argentina, economy, Great Britain, 19th century, the First World War

Modern Trends in Sample Preparation of Biological Material for Analysis of Selected Biologically Active Compounds
Sadílek, Petr ; Solich, Petr (advisor) ; Nobilis, Milan (referee) ; Barták, Petr (referee)
Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Analytical Chemistry Candidate: Petr Sadílek, M.Sc. Supervisor: Prof. RNDr. Petr Solich, CSc. Co-supervisor: Doc. RNDr. Dalibor Šatínský, Ph.D. Title of Doctoral Thesis: Modern Trends in Sample Preparation of Biological Material for Analysis of Selected Biologically Active Compounds This doctoral thesis deals with the determination of selected drugs, their metabolites and other biologically active substances in biological materials. This area of instrumental analysis turns out to be very attractive recently. The reason for this is the need to monitor and optimise treatment and the goal is to achieve the most effective therapy with minimum side effects of drugs for every individual; there are also toxicological reasons. The thesis concentrates on the entire analytical phase of laboratory screening, i.e. not just on development and validation of chromatographical methods of analysis, but also (and especially) on preparation of biological samples prior to the actual analysis. This phase is crucial for analytes assessment, since it determines not only correct results of the laboratory examination, but also influences the lifespan of laboratory equipment, especially chromatographic columns. These days mostly traditional...

Austria and the Eastern question 1829-1841
Šedivý, Miroslav ; Skřivan, Aleš (advisor) ; Kovář, Martin (referee) ; Halada, Jan (referee)
Relying on thorough examination of relevant documents housed in the archives in Vienna, Paris, London, Berlin and Munich, this thesis analyzes the attitude of one of the most important personalities of the 19th century, Austrian Chancellor Metternich, towards the Ottoman Empire and his behaviour in the complicated evolution of the Eastern Question from 1829 to 1841, in other words from the signature of the Treaty of Adrianople to the conclusion of the second London Treaty regarding the Straits. Attention is paid particularly to the final phase of the Greeks' struggle for independence and the relating Russian-Ottoman war, the French expedition to Algeria, the two Ottoman-Egyptian crises, the Rhine crisis caused by the complications in the Levant, the Russian-British rivalry in the East and the difficult situation of the Christians in Syria in the spring of 1841. A further reason for writing this thesis is to point out some of Metternich's hitherto unknown characteristics and diplomatic procedures transpiring from his Near Eastern policy. In the whole period covered by this research, Metternich pursued the policy of conservation: he tried to preserve the declining Ottoman Empire and avoid serious complications in the Levant caused by the attitudes of other European Powers or the internal disorder of the...

Care about non-curable and dying patients on the intensive care unit by nursis
Jansová, Renáta ; Mareš, Jiří (advisor) ; Filip, Stanislav (referee) ; Prchalová, Eva (referee)
Thema of my Bachelour's Paper is care about non-curable and dying patients at the Intensive Care Units. The reason is that I meet these patients nearly everyday when I enter at the Intensive Care Units of all departments. We find references about paliative and terminal care in literature first of all in the connection with oncologic patients. But it is very important to realize that patients also die for many types of other diseases which are serious , hurt and destroy. These patients also have the right to receive a good supporting therapy which helps them to live them from the life without distress and with honor. My Bachelour's Paper consists of two main parts - teoretical and empiric. Teoretical part - first of all this part solves the problem of care about non-curable and dying patients who are in the meantime of the Intensive Care Unit. There are sorted the terms as phases of non-curable disease, symptoms which occur the most during these conditions, pain and its therahy, etic aspects in care about these dying patients. The second part - empiric is devoted how to value nurse personal care about non- curable and dying patients at the Intensive Care Unit from the sight of nurse who work at selected Intensive Care Units of Faculty Hospital in Hradec Králové - GMK ICU internal, GMK ICU geriatric,...

Bíbová, Tereza ; Leláková, Lada (advisor) ; Krajíčková, Dagmar (referee) ; Švecová, Dagmar (referee)
The paper deals with a relatively new methode of treatment - Thrombolysis in neurology. It's composed of a theoretical partand a practical one. In the theoretical part there is golbal information about this methode, its indications, its contraindications, atc. Thrombolysis is a methode of treatment used at acute phase of illness - ischeamic stroke. That's why the characteristic of these illness, its diagnostics, its treatment, atc. are included in this part. The practical part is based on quantitative research. Explored group was composed of patients suffering from stroke and hospitalisated on Intensive Care Unit on Neurological Clinic in Fakultní nemocnice v Hradci Králové. In research there was explored how many times the thrombolysis had been aplied, the reason why it hadn't been aplied and the amount. There were explored the average age and the average lenght of hospitalisation too.

Analysis of the means of communication and self-presentation of Czech parliamentary parties in 2004, before and after the Czech Republic joined the European Union
Škývarová, Soňa ; Jirák, Jan (advisor) ; Nečas, Vlastimil (referee)
Since the overall topic of my diploma thesis is political communication, I focused my attention on Czech parliamentary political parties and their internal communication as well as their public communication strategies. To limit the scope of my research to a reasonable size, I selected year 2004 because three elections were held that year: European Parliament elections, Senate elections, and local authority elections. The exact topic of this thesis is "Analysis of the Means of Communication and Self-Presentation of Czech Parliamentary Parties in 2004, Before and After the Czech Republic Joined the European Union". There are three basic components of my thesis: the theoretical part exploring the notion of political communication, the descriptive part dealing with the political system of the Czech Republic in the given year, and finally the analysis of the communication methods of the selected parties. My research was carried out in several phases. I studied thoroughly my subject and formulated definitions and drew graphs to capture the notion of political communication. I defined it as acts carried out by political actors, both inside and outside of the political organization. Powered by TCPDF (www.tcpdf.org)

Model for a Fluid-Particle Breakup in a Turbulent Flow.
Zedníková, Mária ; Vejražka, Jiří ; Stanovský, Petr
A model is developed for predicting the outcome of breakup of a fluid particle (bubble or drop), which is initially deformed (e.g. due to turbulence) and breaks into two daughter particles. An initially dumbbell-shaped deformation of the particle is assumed. The evolution of sizes of particle sub-parts is calculated using Rayleigh-Plesset equations, which consider the inertia of surrounding fluid, capillary action and viscous effects. The redistribution of internal fluid in the particle is calculated using Bernoulli equation. The model computes the sizes of daughter particles after the breakup. By assuming various initial conditions (various initial shapes and initial velocities of deformation), the size distribution of daughter particles is obtained. These size distributions are qualitatively compared with available experimental data and reasonable agreement is observed. Because of strong assumptions, this model cannot be used directly for accurate prediction of size distribution after a breakup. However, it provides an insight in the physics of the breakup, especially on the effect of inner phase properties.
