Analysis of plant extract of Orthosiphon stamineus by capillary zone electrophoresis

Capillary zone electrophoresis with UV detection at 200 nm was developed for the determination of four phenolic acids and four flavonoids in plant material. The electrophoretic behavior of phenolic compounds was investigated to optimize their separation as a function of buffer pH, buffer concentration. A 50m mM borate buffer, pH 9 was found most favorable. The separation was obtained with fused-silica capillary column (50 cm x 50 μm I.D.) an applied voltage of 25 kV and a temperature of 25řC. For increase injection volume and decrease the detection limits was performed by applying Large-volume sample stacking with polarity switching. The limits of detection were between 0,006 and 0,014 μg/ml. The method was used to determine these compounds in methanolic extract of Orthosiphon stamineus leaves that contained 710,9 μg g-1 pulverized drug of rosmarinic acid and 38,2 μg g-1 of caffeic acid. About a 60- fold enhancement in senzitivity, relative to a conventional injection, was demonstrated in a real sample.

The diploma paper is devoted to verification the application of SPC methods and performs evaluation of statistical stability and process eligibility of steel stampings in the real production process. In the second part is the author of the paper trying to design the optimal form of SPC methods for its use in a specified manufacturing process.

This thesis handles the issue of Relaunch of the Nestlé Ricoré brand on the Czech and Slovak market. The aim of this thesis was to describe the process of brandbuilding of the real fast mooving consumer good. In the theoretical part of the thesis I focused on the analysis and description of current situation of Nestlé Ricoré brand. In the practical part I proposed optimal strategy for relaunch of this brand.

Motivational system of a retailing company as a way of builing employees´ loyalty

The goal of this thesis is to give answer to basic questions about employees' motivation, motivational tools that the employer can use and optimalization of employees' loyalty. To be more focused and precise, we limit our research to the field of retailing companies. The goal of this thesis is thus to analyse the motivational system of a retailing company and its impact on the loyalty of the employees. The thesis is divided into a theoretical and a practical part. In the theoretical part, we will get to know the basic motivational theories and tools. Besides this, we will focus on current trends in the field of employees'recognition. In the practical part, we will apply the theoretical methods to a real company Notes, a.s. We will create, based on a survey reflecting employees' satisfaction, a picture of motivational sytem of this company and we will try to identify its strenghts and weakneasses We should use all the knowledge from this thesis at the end when suggesting improvements of the motivational system for Notes, a.s.

Optimization of the company tax liability

This thesis introduces the tax system of the Czech Republic. The corporate income tax in particular is specified in bigger detail. Its basic structural elements are described with the focus on the transformation of the accountable profit to the tax base. The practical part is divided into chapters, each of which deals with the optimization of the amount of the real tax liability. The content, instruments mentioned and methods for decreasing company tax liability applied suggest this work more likely addresses smaller and medium-sized companies. These companies need what is referred to as tax planning. This thesis analyzes the possibilities of the use of tax-deductible items provided by law, tax abatements; further it specifies cost and revenue conception of tax optimalization or more precisely the amount of the accountable profit or tax base. The aim of the practical part is to identify tax optimization possibilities and to compare on concrete cases what tax saving or other advantages to the taxpayer each method brings.

Decision of a Steel Company Trading with Emissions

We formulate a Mean-CVaR decision problem of a production company obliged to cover its CO2 emissions by allowances. Certain amount of the allowances is given to the company for free, the missing/redundant ones have to be bought/sold on a market. To manage their risk, the company can use derivatives on emissions allowances (in particular futures and options), in addition to spot values of allowances. We solve the decision problem for the case of an real-life Czech steel company for different levels of risk aversion and different scenarios of the demand. We show that the necessity of emissions trading generally, and the risk caused by the trading in particular, can influence the production significantly even when the risk is decreased by means of derivatives. The results of the study show that even for low levels of the risk aversion, futures on allowances are optimal to use in order to reduce the risk caused by the emissions trading.

The thesis is focused on finding optimal solution of stock company Dencop Lighting spol. s.r.o. At the very beginning of the thesis there is a brief characteristic of Dencop Lighting, their market aims and goals. After this view we are going to introduce you warehouse systems. There are analysis of products, their selection, flows and potentials for company. After completing product analysis we are going to focus on key factors of optimalization. There are mentioned many ways of optimalization with their advantages and disadvantages. Deterministic and stochastic methods are included as well. For better illusion in real live you can find added CD at the bottom of the thesis. Using guide of this software with detailed information about functions is in last pages of the thesis.

Laboratory diagnostics of micrometastases in breast cancer patients

Introduction: The presence of circulating tumor cells (CTC) in the peripheral blood has been associated with worse prognosis and early relapse in breast cancer patients. CTC determination in the peripheral blood has been considered as a liquid biopsy. The aim of this project was to analyze the presence of CTC followed by their molecular characterization with the potential use not only as a new biomarker for real-time monitoring of therapy efficacy but also as a suitable tool for patient's stratification and individualization of treatment for breast cancer. Methods: A total of 54 patients with diagnosed early breast cancer were enrolled into a prospective study. Ten millilitres of peripheral blood were sequentially collected to test for the presence and characterization of CTC during the follow-up of patients. CTC isolation and detection was performed by AdnaTest BreastCancer™ (AdnaGen AG, Germany), which is based on the detection of EpCAM, HER2 and MUC1 specific transcripts in enriched CTC- lysates. cDNA from isolated CTC has been further used for newly optimized qPCR assays for breast tumor and therapy resistance associated genes: TOP1, TOP2A, CSTD, ST6GAL, KRT19 and reference gene actin. qPCR results have been analyzed by Genex software (MultiD Analysis). Optimization model of production in food industry

The thesis deals with planning and scheduling of production in the food industry. The theoretical part is concerned with the formulation and structure of the task scheduling and analyses the flow shop in the detail, for which a real application for an unnamed company has been created. It also briefly describes the technical specifications of production, which are crucial for the practical part. The practical part deals with the formulation of a mathematical model. The optimal solution is obtained using the optimization program MPL for Windows. Mathematical model includes variables that solves serial and parallel processors and try to capture as most exact as possible manufacturing processes with the technical specifications for a particular enterprise. The criterion for optimization is to minimize the cost in terms of the selected technology of production.

Mathematical Programs for Dynamic Pricing - Demand Based Management

Tato disertační práce se zabývá vývojem, modelováním a analýzou poptávkově orientovaných úloh, které zahrnují marketingová, operační a logistická rozhodnutí. Úlohy jsou zvoleny tak, aby mohly být dále rozšířeny o koncept tzv. dynamického oceňování a jiných dynamických marketingových rozhodnutí. V práci jsou využity dvě základní poptávkově orientované úlohy: a) úloha kolportéra novin, která je zvolena pro její jednoduchou formu a která tak slouží jako nástroj pro ilustrativní ukázky rozhodovacích procesů v podobných typech úloh, a b) úloha návrhu dopravní sítě, kde jsou využity některé výsledky a znalosti získané při řešení úlohy kolportéra novin. Kolportér (či obecně maloobchodník) čelí náhodné poptávce, která může být postupně ovlivněna oceňováním, marketingovými (tj. reklamními) rozhodnutími a nakonec jejich kombinací. Poptávka obsahuje tedy náhodnou složku, která je pomocí přístupů stochastické optimalizace modelována ve specifickém tvaru (tj. aditivní či multiplikativní tvar). Závislost cena-poptávka je zachycena pomocí nelineární klesající poptávkové funkce, zatímco (vhodná) reklama vede ke zvýšení poptávky (běžně rostoucí s-křivka či konkávní funkce). Výsledky získané při řešení úlohy kolportéra novin s oceňováním jsou následně využity v úloze návrhu dopravní sítě. Tato stochastická úloha je modelována (reformulována) pomocí dvou přístupů stochastické optimalizace: wait-and-see přístup a here-and-now přístup. Jelikož tato implementace vede na lineární či nelineární celočíselnou (navíc scénářovou) úlohu, jsou v práci zmíněny taky výpočetní nástroje. Autor pro řešení používá (původní) tzv. hybridní algoritmus, což je kombinace heuristického (genetického) algoritmu a nástroje optimalizačního softwaru. Potenciální aplikace sestavených modelů, obzvláště v oblasti odpadového hospodářství, jsou diskutovány v závěrečné části disertační práce.
