Optimisation of SIA decarbazine determination

Dacarbazine was determinated by sequential injection analysis with chemiluminiscence detection during reaction of sample and KMnO4 in sulfuric acid. Reagent volumes, their concentrations, flow rate and structure of cycle were optimised. Also the influence of solvents (water, 60% ethanol, 60% methanol) and enhancers of CL reaction (sodium polyphosphate, formic acid, glutaraldehyde and formaldehyde) was tested. Final optimal conditions are: 40 μl 1 M H2SO4, 3 μl 10 mM KMnO4, 30 μl 10 mM solution of dacarbazine, 40 μl 1 M H2SO4, eventually aspiration of 50 μl enhancer. Aspiration of the solutions proceeded in flow rate 100 μl/s, mixing and flow rate to the detector was 40 μl/s. Washing with oxalic acid was used after each measurement cycle to prevent KMnO4 settling on tubes. Voltage at the photomultiplier of the detector was 420 V. The calibration relations of CL intensity to concentration of the samples were nonlinear with possibility of selection of linear areas. The linear area for the solution of dacarbazine without enhancers wasn't found with correlation coefficient high enough, for the solution of dacarbazine with 60% methanol was 0.5 - 10 mmol/l, for the solution of dacarbazine with 1.39 M formic acid 4 - 10 mmol/l. Limit of detection for the solution of dacarbazine without enhancers was 0.1474...

Crohn's disease and ulcerative colitis are both entities of inflammatory bowel disease (IBD). The nuclear transcription factor kappaB (NFκB) family consists of five different members, which are namely p65 (RelA), c-Rel, RelB, p50 and p52. NFκB has ability to promote the expression of various proinflammatory genes, to influence the course of mucosal inflammation and to mediate different cell-type specific effects. Bilirubin is an endogenous substance with anti-oxidative and anti-inflammatory effects, which influences the process of inflammation and has protective effect. This diploma thesis was aimed at conduction of methods of NFκB detection on paraffine tissue slices and observation of NFκB expression in large intestine walls of normobilirubin and hyperbilirubin rats (Gunn's variety) after chronic imunohistochemically- induced inflammation. Eight Gunn's variety rats (G+ ) and eight Wistar's variety rats (K+ ) were used in this study, all after long time administration of dextrane-sulfate (DSS). For seven following days, DSS (in concentration of 2%w/v) was added into drinkable water for these rats, followed by 14 days of clear drinkable water (without DSS) administered to rats. Determining the compression behaviour of microcrystalline cellulose and aluminium stearate

This graduation theses delas with determination of compactibility of microcrystalline cellulose and aluminium stearate and their viscoelastic properties during the tablet formation. The force-distance record was used. The theoretical part concerns with characterization of microcrystalline cellulose and aluminium stearate and describes individual evaluation procedures and the influence of single materials to elastic recovery. The aim of experimental part was the determination of compaction pressure to compaction energy, compaction pressure to tablet relaxation and aluminium stearate concentration to compaction energy. Tablets were compacted in the T1 FRO machine, the preload was 2N, speed of preload 2mm/s and cycle rate 2mm/s.

This thesis aims to serve as a suitable material for future entrepreneurs who want to orientate quickly and reliably in the issue of starting a business in the legislative environment of the Czech Republic before making own decision to establish a company. The work is divided into two parts - first, theoretical and methodological, will explain all theoretical aspects regarding establishment of businesses in our country. The second part, analytical, will transfer these aspects into practice. On example of a fictional company the reader will be step by step acquainted with it's whole life cycle.

The diploma thesis is itself aimed at using the computers in the mixed class of kindergarten. This diploma thesis shows the steps, which the teacher has take do if she decides to use the computer in the class. In the scientific part I have concentrated on the integration of the computer in education at program, the other possibility to use the computer in education at process is by meeting the conditions of using the computers. In the next part I have rated the computer programs for children. The final topic is the opinion and attitude of the parents.

Comparison of the effectiveness of government measures at the time of major depression

The goal of my work is to compare the Great Depression in the USA to the current depression. I would like to compare the causes, development and government actions to fight both these depressions. My work concentrates at the tools which were used to ''save the economy'' in the USA after the economic breakdown in 1929 and also at the results of using these tools. According to the analysis of individual steps of the New Deal I will try to estimate the effectivity of proposed similar measures supposed to fight the current financial crisis in the USA as well as in the European Union and in the Czech Republic.

This study was focused on implementation of experimental techniques for purification of human liver microsomal fraction. There were chosen three basic steps of purification process. As a first step, ion-exchange chromatography was chosen followed by two purification steps on the base of gel filtration. Before purification the microsomal fraction had to be solubilized to release proteins from membrane bounds and conditions had to be optimized. Single fractions were subsequently incubated with specific substrate of reductases oracin and the quantity of metabolite 11-dihydroracin was assessed by achiral HPLC analysis. For the most active fractions the SDS-PAGE analysis was done to set size of unknown enzyme. By use of antibody against 11beta-HSD1 was axcleded the presence of this reductase in all analyses fractions. By use of described purification techniques we were successful to concentrate fraction with unknown human liver reductase.

Evaluation of effectiveness of the EU's regional policy

Regional Policy of the European Union should lead to decrease deepening of regional disparities, which is present due to ongoing integration and enlargement of the EU. A lot of regional disparities is caused by so called agglomeration forces, which lead to concentration of economic activity in strong centers (agglomeration poles), and which are described by New Economic Geography. This thesis deals with the question if today's Regional Policy of the EU leads to reduce regional disparities. Random walk and game theory

The results od random walk analysis have been applied to computer science, economics, biology tec. This thesis is concentrated on connection between random walk and games theory especially with thinglering simulation. The application part of this thesis is focused on game strategies, that determine the amount of bet in next game.
