Selective isolation of of the genus Lactobacillus bacteria from foods

Probiotic lactic acid bacteria of genus Lactobacillus play an important role in the digestive tract of human. They are used in food processing and they are the part of food supplements. Lactic acid bacteria of the genus Lactobacillus can be identificated by polymerase chain reaction (PCR). Bacterial DNA was isolated from cell lysates of 4 synbiotic food suplements by magnetic particles P(HEMA-co-GMA). Isolated DNA was amplified by genus-specific and species-specific primers. Magnetic particles with immobilized antibodies against Lactobacillus bacteria were used in the next part of thesis. These particles were used for isolation target cells from products with their identification by genus specific PCR.
Identification of selected genes in lactic acid bacteria

Lactic acid bacteria are natural habitants of human gastrointectinal tract. Among the most important are bacteria of genus Lactobacillus and genus Bifidobacterium that contain a lot of probiotic species. Probiotic species are used as food supplements. This work was focused on DNA separation from crude cell lysates of 4 food supplements using magnetic carrier P(HEMA-co-GMA) covered by carboxyl groups. DNA was reversible adsorbed to the carriers in the presence of PEG 6000 (16%) and NaCl (2 M) (final concentrations) and eluted into TE buffer. Lysis of cells from food supplements was performed by lysozyme, SDS and proteinase K. The amount of lysozyme was optimalized. Concentration of separated DNA was measured by spectrophotometric method. The amount of isolated DNA was suitable for PCR. Isolated DNA was used for PCR with universal primers, PCR specific for genus Lactobacillus and genus Bifidobacterium and for 9 different species-specific PCRs: Lactobacillus rhamnosus, Lactobacillus acidophilus, Lactobacillus plantarum, Lactobacillus fermentum, Lactobacillus casei/paracasei, Streptococcus thermophilus, Bifidobacterium longum, Bifidobacterium bifidum and Bifidobacterium infantis. Amplicons were detected by agarose gel electrophoresis (1,8%). It was shown that DNA amplification methods are quick and precise for identification of studied species. The results of bacteria identification were compared with data provided by the manufacturer. In all food supplements, bacteria of genus Lactobacillus and Bifidobacterium were detected. However, only some species provided by manufacturer were identified by PCR in each tablet.
Determination of carnitine in food supplements

This work deals with the determination of L-carnitine in food supplements. L-carnitine is a substance naturally occurring in organism, essential for metabolism of fatty acids. In food supplements is used especially for reducing body weight as a "fat burner". L-carnitine is popular with athletes for improve athletic performance. While data are not available to support these positive effects of carnitine, the positive results of carnitine supplementation in the medicine were found, mainly on cardiovascular system. The literary part of this study describes the properties and the use of carnitine in the diet of human. Furthermore, a review of methods used for determination of L-carnitine in variety of samples is mentioned. In the experimental section the efficiency of SPE method for preparation matrix of real samples was investigated. A cation exchange solid phase extraction seems to be the most effective. The optimal conditions for isolation of L-carnitine by SPE extraction were not developed. L-carnitine was determined in six various food supplements by reverse phase chromatography with UV/VIS detection.
In vitro study of antioxidant activity of food supplements

Bachelor thesis deals with the determination of antioxidant activity of food supplements in vitro. The theoretical part focuses on legislative requirements of food supplements, their characteristic and classification. Further, the attention is also devoted to antioxidants, their properties, effects, importance and utilization. Separate chapter describes herbs, which are an essential component of researched supplements, their effects and properties. For experimental part several food supplements in various forms (capsule, tea, juice, dried fruits) were chosen and total content of polyphenols was determined using the UV-VIS spectroscopy. Further, the total antioxidant activity is characterized by electron paramagnetic resonance involving DPPH method.
Demarcation between the categories of products with physiological function on human body in EU law

The thesis is focused on theoretical principles of demarcation of products with health effects in the EU law and on the reflection of these issues in the application and administrative practice in commercial law. The thesis developed yet been brought judicial interpretation with the scientific interpretation of certain key and decisive problems that separate from each other law-regimes of regulation: medicinal product for human use, foods, cosmetics products, medical devices and biocides. Correct law-regime for a particular product is determinating of the general legal basis in the EU internal market. It is therefore a important problem not only in EU law, but also in EU economic. Thesis results are highly critical of current practice in the local markets of EU member states, especially Czech Republic, and in many instances documented sub-optimal application of this law in general practice.
The market of food supplements and their trustworthiness

The bachelor thesis deals with the market of food supplements and their trustworthiness. The aim of this thesis is to inform about the current situation on the market of food supplements in the Czech Republic and abroad, to highlight the importance of consumption of food supplements and to present conclusions obtained during a personal survey. The work could be divided into two parts, the first part is theoretical and the second one practical. The purpose of the first part is primarily to introduce the topic of food supplements and to provide information about the market of the food supplements as well as about the food supplements themselves. The practical part provides information about shopping behavior of customer and about his general knowledge in this area.

The Goal of the Master's thesis is to create a marketing plan for a specific company. The first part contains chapters that capture the theoretical foundations needed to build a marketing plan. The practical part deals with the Phyto CZ, Ltd. Company which operates in the production and sale of food supplements. It contains the steps needed to develop a marketing plan. First, the company's marketing mix a selected elements of situational analysis are analysed, with the results of a questionnaire survey added. Subsequently, the company is recommended a set of objectives for the future, including a proposal of specific marketing procedures which could lead to their achievement.
Legal and ethical rules on advertising in healthcare

The aim of this thesis is to compare food supplements used on the activity of endurance character and their influence on the cardiovascular system. The teoretical part is pursued to the basic food components and their metabolism and to kinetic activities splitting. The practical part gives information about splitting of sports supplements used on the activity of endurance character, their usage and effects. It deals with the question of human organism behaviour during the strain of endurance character and how the food supplements help to regenerate the human body. Further, it presents a food supplements overview according to their focus and their effect on the human organism during the strain of endurance character. Finally, the summary states which of the specific food supplements are the most optimal for the strain of endurance character.
Pharmaceuticals, food supplement and medical device advertising

The thesis brings a complex insight into the legal environment of the pharmaceutical and food supplement advertising. It focuses on the legal regulations in the branch with emphasis on decision making processes of the supervising and other relevant state bodies. Special attention is paid to the ability of the above mentioned authorities to enforce the powers given to them by the law and to their cooperation. The main goal of the thesis is to assess whether the contemporary adjudication on the two markets (pharmaceuticals and food supplements market) is transparent and close enough to ensure effective protection of the public and consumers. A very important aspect of the whole problem is also to find out whether the jurisdiction allocation among the authorities and the way the make use of their powers leads towards sufficient assurance of legal certainty and public priorities, that is to say consumer protection and public health.
