Barbora Vaňásková Supervisor: Prof. PharmDr. Martin Doležal, Ph.D. Title of Doctoral Thesis: Derivatives of pyrazinecarboxylic acid as potential antituberculotics (synthesis and biological evaluation) This doctoral thesis deals with searching for potential antituberculotic drugs derived from pyrazinecarboxylic acid. Thesis contains theoretical part, in which problematics of tuberculosis, factors hindering the effectiveness of treatment (development of resistance and HIV coinfection) and current therapeutic practice are outlined. An individual chapter is devoted to the composition of the mycobacterial cell wall. A brief overview of first-line and second-line antituberculars as well as drugs newly introduced into the clinical practice and promising derivatives in various phases of preclinical and clinical trials is further stated. Special attention is dedicated to pyrazinamide, current theories dealing with mechanism of action of this first-line antituberculotic drug and to ribosomal protein S1, a specific target of pyrazinecarboxylic acid. Vývoj bankovního systému Ruské federace

V poslední době došlo k posílení pozitivních tendencí v rozvoj bankovního sektoru. Tempy růstu většiny jeho ukazatelů jsou nejvyšší za celou dobu vývoje a zároveň roste jejich podíl k hrubému domácímu produktu. Roste význam bankovního sektoru pro ekonomiku země, roste důvěra k bankám ze strany vkladatelů a věřitelů. Současně růst bankovního podnikání a komplikace jeho povahy, rozšíření nabízených bankovních produktů a služeb, včetně v oblasti drobného úvěrování, je doprovázen akumulaci rizik. Proto přednostním úkolem je posílení odolnosti úvěrových institucí, bankovních skupin a celého bankovního sektoru cestou zlepšení systém řízení rizik a vnitřní kontroly, rozvoje manažerského vedení a transparentnosti. Takže existuje prostor pro zlepšení kvality regulace bankovní činnosti a systému bankovního dohledu.

Tvorba obchodně úspěšného webu

Tato práce popisuje postup při tvorbě webu, který by měl být obchodně úspěšný. Cílem je shromáždit a popsat všechny potřebné kroky pro vytvoření takového webu. Nejedná se však o přesnou metodiku, ale spíše o volný návod. Při konkrétním webovém projektu by měly být vybrány kroky a způsob jejich realizace, které jsou pro daný projekt relevantní a jejichž provádění přinese patřičný efekt. V této práci popisuji své znalosti, zkušenosti a postřehy, které jsem získal během působení ve firmě H1.cz. Popisovaný postup je zároveň postupem, který se i v této firmě pro projekty nového webu používá. Popisovaný postup i celá práce je rozdělena na části: Vstupní studie, Analýza, Návrh, Implementace, Kontrola a Provoz webu. Na začátku práce se navíc zabývám tím, co je vlastně obchodně úspěšný web, ke kterému má celý postup směřovat. Obsahem Vstupní studie je seznámení se s oborem, ve kterém bude web působit, a odhad velikosti poptávky a předpokládaných nákladů. Analýza se zabývá cíli webu, cílovou skupinou, konkurencí a již existujícím webem. V Návrhu jsou popsány stanovení strategie webu a návrh struktury, navigace a jednotlivých stránek a sekcí webu. V části Implementace a Kontrola jsou popsány možnosti kontroly kvality při implementaci webu. V části Provoz webu se nacházejí vybrané poznatky vztahující se k využívání webu, jeho správě, propagaci a řízení pomocí KPI.

Práce se zabývá výkonností, efektivností FreeNetů (resp. PilsFree). Občanské sdružení PilsFree je komunitní počítačová síť působící v Plzni a jejím okolí poskytující Internet. Práce porovnává kvality Internetu poskytovaný komerčními poskytovateli a PilsFree pomocí metod nákladově užitkové analýzy. Dále se věnuje výkonnosti Internetu poskytovaný Prahou 5. V poslední části porovnávám nákladovost na zřízení koncového bodu v závislosti na technologii.

Adolf Liebscher 1857-1919

Adolf Liebscher belongs among the artists known as the "National Theatre Generation." Like ist foremost representatiwes, F. Ženíšek, M. Aleš and V. Hynais, Liebscher also produced an oeuvre of great diversity - he created decorative paintings and historical compositions, was interested in folklore painting, his drawings were published by the major journals of that time as Zlatá Praha or Světozor and rus illustrations appeared in many albums, books of poems and novels. Liebscher was bom on March 11, 1857, into a family of a Provincial Committee engineer Josef Liebscher. His older brother Karel (1851 - 1906) later became an appreciated landscape painter, with whom Adolf often cooperated. In 1875 - 1878, Adolf Liebscher studied at the School of Applied Arts in Vienna, in the three-year course for hand-drawing teachers under F. Laufberger. After obtaining a diploma, he entered the studio of E. A. Donadini for half-a-year, where he prepared his proposals for the competition for the National Theatre. In 1879, he was awarded in the competition and was commissioned to carry out some of his proposals in the passage of the theatre. Here he created eight lunette allegories of Dramatic arts.

Targetting prostate tumor cells by polyomavirus virus-like particles

The aim of this thesis is to investigate the targeting potential of mouse polyomavirus (MPyV) based virus-like particles (VLPs) as vectors for directed cell delivery of therapeutic or diagnostic compounds. Major capsid protein VP1 of MPyV is able to selfassemble into the noninfectious VLPs. Our main goal is to retarget these VLPs from its native receptor to the prostatic cancer cells by changing the receptor binding site in the surface-exposed loop of VP1. We introduced a peptide ligand CTITSKRTC, which binds prostate-specific membrane antigen (PSMA), by insertion or substitution into BC loop of VP1. These modifications did not change the stability of the particles and genetic substitution prevented the native receptor binding. PSMA-specific binding of modified VLPs was tested by pull-down assay and surface plasmon resonance. In order to further utilize these VLPs, we tested several approaches for preparation of VLPs as vehicles for compounds delivery into eukaryotic cells. Although the method for encapsidation of the DNA into the VLPs in cellular nuclear extracts, which mimic the in vivo conditions, did not enabled us to produce pseudocapsids, we successfully optimized procedure for dissassembly and reassembly of purified particles. Physical Activity during Hemodialysis and its Impact on Physical Fitness in Patiens with End-stage Renal Disease (ESRD)

Title Physical activity during hemodialysis and its impact on physical performance at patients with end-stage renal disease (ESRD). Objective: To verify efficiency of exercise training during hemodialysis with emphasis on improvement or preservation ofphysical performance in patients with ESRD. Methods: 13 patients with ESRD on hemodialysis participated in exercise training programme during hemodialysis. Senior Fitness Test (SFT), dynamometry of hands and multifrequency bioimpedance analysis were used to evaluate physical performance before and after 12 weeks of exercise training programme during hemodialysis. 1Onon-exercising patients with ESRD served as a comparsion group. Results: Physical performance in patients participating in exercise programme increased significantly (p<0,05) for SFT (by 16% for chair stand test (p<O,Ol), by 53% for 2-minute step test (p<0,002), by ll % for up-go (p<0,04)) and by 6 % for dynamometry test (p<0,04). No changes in body composition were noted in both groups after 12 weeks. Conclusions: Twelwe weeks of exercise training programme during hemodialysis can improve physical performance at patients with ESRD. Prologue

In film, the term “prologue” nowadays denotes almost everything that appears before the opening credits. The meaning of this word is not fixed. This thesis does not define the “prologue” phenomenon and does not wish to describe it in the historical context of art spheres. It deals with the opening scenes at the beginning of a film before the onset of the actual composition of the main story and addresses their functions in the entire dramaturgic structure of the work. To facilitate the apprehension and description of “prologue”, the thesis attempts to generalise the term and classify it into several groups according to defined criteria. It does not set itself the objective of forming a definition but of analysing the dramaturgic structure of a conventionally understood term “prologue” and initiating a discussion on all fronts of the film-making profession about this tool that can do more than just touching up the first few minutes of a film.
