The Netherlands: migration trends and policy

Dutch society have gone through many changes over 400 years, the biggest one came during the 20th century, when the Netherlands became a multicultural country. The Netherlands is well-known for its extreme tolerance for minorities and the government tends to integrate these people into Dutch society through immigration and integration policy. Country does not struggle only with the influx of immigrants, the renewed problem is the leaving their own people. Initially, in a relatively peaceful coexistence of immigrants with residents over time, there have been first manifestations of racial hate, religious wars and conflicts due to cultural differences. As a respond to this situation is emigration of the local population. The government still has to adapt its policy because of the changes in structure of migration to suit both parties.

Role of endocytosis and endosomal acidification in TRAIL-induced apoptosis

TRAIL (TNF-related apoptosis inducing ligand) became known for its ability to selectively eliminate cancer cells. This ligand is a member of the TNF (tumor necrosis factor) ligands family and triggers extrinsic apoptotic pathway by binding of its death receptor 4 or 5 (DR4/5), and subsequent formation of death-inducing signalling complex (DISC). This signalling complex is required for successful transmission of apoptotic signal and activation of proximal caspases. However, regulation of the initial steps leading to activation of caspases is still not fully understood. Endocytosis of a TRAIL- DR4/5-DISC complex can be one of modulators of the initiation of extrinsic apoptotic pathway. Recent studies show controversial data documenting that endocytosis of TRAIL receptosomes can in cell type specific manner either positively or negatively influence TRAIL-induced apoptotic signalling. In this study, we focus on the analysis of a role of endocytosis and acidification of endosomal compartments during TRAIL-induced apoptosis in human colorectal cancer cell lines. Our results support the view that both clathrin-dependent endocytosis of TRAIL receptosome and endosomal acidification positively affect activation of caspases during the early stages of TRAIL-induced apoptosis. Title: Vessel segmentation Author: Ján Dupej Department / Institute: Department of Software and Computer Science Education Supervisor of the master thesis: RNDr. Josef Pelikán, KSVI Abstract: In this thesis we researched some of the blood vessed segmentation and visualization techniques currently available for angiography on CT data. We then designed, implemented and tested a system that allows both semi-automatic and automatic vessel segmentation and visualization. For vessel segmantation and tracking we used a region-growing algorithm that we overhauled with several heuristics and combined with centerline detection. We then automated this algorithm by automatic seed generation. The visualization part is accomplished with an adaptation of the well-known straightened CPR method that we enhanced so that it visualizes the whole cross-section of the blood vessel, instead of just one line of it. Role of protein kinase C isoforms in cardioprotective mechanism of chronic hypoxia

Cardiovascular diseases, particularly acute myocardial infarction, are one of the leading causes of death in developed countries. It is well known that adaptation to chronic intermittent hypobaric hypoxia (IHH) confers long-lasting cardiac protection against acute ischemia/reperfusion injury. Protein kinase C (PKC) appears to play a role in its cardioprotective mechanism since the administration of general PKC inhibitor completely abolished the improvement of ischemic tolerance in IHH hearts. However, the involvement of individual PKC isoforms remains unclear. Therefore, the primary aim of this study was to investigate the potential involvement of PKCδ and PKCε, the most prevalent PKC isoforms in rat heart, in the mechanism of IHH-induced cardioprotection. We showed that IHH up- regulated PKCδ protein in left ventricle, enhanced its phosphorylation on Ser643 and increased its co-localization with markers of mitochondrial and sarcolemmal membranes. PKCδ subcellular redistribution induced by IHH as well as the infarct size-limiting effect of IHH was reversed by acute treatment with PKCδ inhibitor rottlerin. The assesment of trombocyte function by optical and impendance agregometry

Introduction: Platelets have an important role in the process of haemostasis in which they form blood clot generation. In some cases it is necessary to suppress activity of platelets by antiplatelet drugs to prevent life threatening states. Acetylsalicylic acid (ASA) and clopidogrel can influence the mechanism of primary haemostasis by inhibition of platelet function. This process has to be controlled. Clopidogrel is used for prevention of thrombotic states. The active form of clopidogrel is formed by cytochrome P450 3A4. Clopidogrel binds to the receptor P2Y12 instead of ADP, so platelets cannot be activated. The effect of clopidogrel is very variable. It is known that there is unresponsiveness to clopidogrel in 5-30 % patients. Acetylsalicylic acid acetylates cyclooxygenase and blocks the conversion of arachidonic acid to tromboxan A2, so platelets cannot be activated. ASA treatment is effective, but in some patients can appear so called aspirin resistance (AR). Possible unresponsiveness to the therapy is the reason why it is suitable to monitor antiplatelet therapy, which may be provided by various methods. Index Suitable for Similar Search in High-dimensional Spaces

In this paper, we focus on indexing and searching in high-dimensional data. To achieve the target we implemented the Metric Index, a model of the similarity search based on the metric spaces, that employs many of known principles of partitioning and filtering. The metric space is a general model of similarity, which enables the usage of implemented index for various data. With this index, stored data could be searched effectively. The internal structure of data is hidden, we just require an implementation of the function for feature extraction, which produces a vector representing data, and the metric function applicable to the given data. The Metric Index was implemented as a data cartridge, the mechanism for extending the capabilities of the Oracle server. This data cartridge enables indexing of large unstructured data in the Oracle server known as LOBs.

Molecular genetics of celiac disease

Celiac disease is an organ-specific autoimmune disease that arises as a consequence of hypersensitivity to the grain gluten in genetically susceptible individuals. Genetic predisposition are HLA-DQ2 and HLA-DQ8 genes, which are necessary but not sufficient for the emergence of celiac disease; it is involved in approximately 40% of the inheritance. In the course of the time, other genes that might contribute to the pathogenesis of celiac disease are being discovered. Among these so-called candidate genes, which are sought on the basis of known knowledge of molecular mechanisms of innate and adaptive immune responses, are for example: MIC, TNF, CTLA-4, CD28, ICOS, MYO9B, MMP, TLR and PTPN22. Immune response triggered by gluten peptide penetration into the lamina propria leads to mucosal damage. Different gluten peptides are involved in the pathology of celiac disease in different ways, some peptides trigger an adaptive immune response, while others, such as peptide p31- 43, triggers an innate immune response.

Cis and trans elements in plant response to drought (with a particular focus on photosynthetic genes)

Abiotic stresses, such as drought, high salinity and cold can strongly affect plant development and growth. Cis and trans elements are one of the options how plants regulate their own metabolism in those cases. That's the way how the expression of many target genes is induced. The products of these genes function not only in stress tolerance but also in general stress response. Many transcription factors and regulatory proteins (trans elements) are involved in these adaptions; those often interact with specific sequences in gene promoters (cis elements). Recenly, a progress has been made in analyzing the signal paths and complex cascades of gene expression regulation, although a little is still known about this regulation during drought conditions. Some of these target genes code products participating in photosyntesis and the regulation of their expression can significantly affect this process. This essay briefly describes main cis and trans elements of plant response to drought (with a particular focus on the regulation of photosyntetic genes). The main topic of this bachelor theses is intercultural communication and its use in the field of international human resources management. Emphasis is placed on employees, who must adapt to a different culture for effective performance abroad. The theses briefly introduces cultural standards and barriers hampering the process of intercultural communication. The aim is to clarify the importance of intercultural communication training in the management of expatriates and application of knowledge about the specifics of international human resources management into practical development of intercultural competence.

Already the stan- dard back-propagation training algorithm used for their adjustment provides often applicable results. However, efficient solutions to complex tasks cur- rently dealt with require a quick convergence and a transparent network structure. This supports both an improved generalization capability of the formed networks and an easier interpretation of their function later on. Var- ious techniques used to optimize the structure of the networks like learning with hints; pruning and sensitivity analysis are expected to impact a bet- ter generalization, too. One of the fast learning algorithms is the conjugate gradient method. In this thesis, we discuss, test and analyze the above-mentioned methods.
