Optimalizace GC/MS metody stanovení silyl derivátů lignanů
BEDNÁŘ, Jan
The main goal of this work was optimization of GC-MS method for the analysis of lignans trimethylsilyl (TMSi-) derivatives of two model compounds hydroxymatairesinol(HMR) and enterolactone (ENTL). It included validation of GC-MS metod for TMSiHMR and TMSi-ENTL, comparison of chosen derivatization reagents and verification of time stability for TMSi-HMR. It was shown that TMSi-HMR and TMSi-ENTL provide a nonlinear calibration curve. Second order polynomial curve gave the best fitting and was used for method validation. N-Methyl-N-(trimethylsilyl)trifluoroacetamide activated I and II (Sigma Aldrcih) were selected as the best choice for simultaneous derivatization for HMR and ENTL mixture. Room temperature was confirmed to be appropriat for 3 days storage of TMSi-HMR. Also it was shown that storage at -80 °C preserve the lignan samples for at least 90 days storage.

Mimicking of thiram metabolism using electrochemistry hyphenated with mass spectrometry
Jaklová Dytrtová, Jana ; Jakl, M.
On-line hyphenation of a home-made electrochemical cell with an electrospray ionization mass spectrometer (EC-ESI-MS) was investigated to be applicable in mimicking of thiram (T) metabolism. The EC-ESI-MS instrumental approach confirms the creation of the first metabolic product of T (Tm). Tm is created via the decay of the disulfide bond. The catalytic effect of copper originated from the copper working electrode to this reaction is discussed.

The practical part was complemented by a survey of 160 university students, with the focus on the frequency of use for the loyalty program and the willingness of a individual to provide personal data.

The importance of food in disease celiac sprue
Matlášková, Jana ; Faměra, Oldřich (advisor) ; Capouchová, Ivana (referee)
The celiac sprue is an autoimmune disease that affects 1% of the population. The disease is dependent on several factors. One of this factors is a genetic predisposition, which is related to the genome HLA-DQ2 and DQ8. It also depends on the environment - the period of breastfeeding, the period of inclusion the gluten into the diet, and also taking antibiotics. These factors include gluten itself, which works as a trigger of the celiac sprue disease. The gluten is composed of two protein components of prolamin and glutelin. It is contained in wheat, barley, rye, oat and in their hybrids. It appears at creation of paste. If a patient with the celiac sprue disease eats gluten, his autoimmune system doesn´t respond correctly and it starts attacking its own cells, because it´s unable to recognize the difference between its own and foreign cells. The symptoms of the celiac sprue disease include intestinal problems (diarrhea, abdominal pain), osteoporosis, anemia etc. It is also related to some of the autoimmune diseases such as diabetes mellitus I or dermatitis herpetiformis. Some patients are difficult to diagnose because they don´t have any of these symptoms. Celiac disease leads to the villi in the small intestine damage. The villi may completely disappear when the disease is not long-term treated. A gluten-free diet is the only way how to treat this disease. It means that a person with the celiac sprue diagnosis has to eliminate from the diet all foodstuffs that contain risk proteins. First of all it is wheat, rye, barely and oats. Gluten may be hidden in some foods and therefore each package must contain allergens used in the product. The product can be labeled as gluten-free only if the amount of gluten is to 20 mg/kg of food. These products are symbolized by a crossed-out grain. People with celiac disease most use the products made by rice, maize, millet and pseudocereals, because they´re gluten-free. This diet is lifelong and needed, because each offense may affect the health of the patient.

Simulation of Accelerator Driven Nuclear Reactor for Spent Nuclear Fuel Transmutation
Jarchovský, Petr ; Ing. Antonín Krása, Ph.D., SCK.CEN Mol (referee) ; Katovský, Karel (advisor)
This master’s thesis deals with usage of burn-up (spent) nuclear fuel in nuclear power plants of next generation – accelerator driven transmutation plants which is produced in current nuclear power plants. This system could significantly reduce the volume of dangerous long-lived radioisotopes and moreover they would be able to take advantage of its great energy potential due to fast neutron spectrum. In the introduction are listed basic knowledge and aspects of spent nuclear fuel along with its reprocessing and the possibility of further use while minimizing environmental impact. As another point detailed description of accelerator driven systems is described together with its basic components. In addition this search is followed by individual chronological enumeration of projects of global significance concerning their current development. Emphasis is placed on SAD and MYRRHA projects, which are used like base for calculations. This last, computational part, deals with the creation of the geometry of subcritical transmutation reactor driven by accelerator and subsequent evaluation which assembly is the most effective for transmutation and energy purposes along with changing of target, nuclear fuel and coolant/moderator.

Antiradical activity of herbal drugs Epilobii herba and Ononidis radix extracts and flavonoids.
Dvořáková, Monika ; Spilková, Jiřina (advisor) ; Martin, Jan (referee)
1 ABSTRAKT Mgr. Monika Dvořáková Reactive oxygen species play a key role in the patophysiology of renal diseases. The kidney is an organ highly vulnerable to damage caused by reactive oxygen species. Oxidative stress mediates a wide range of renal impairments, from acute renal failure, rhambdomyolysis, glomerular damage, obstructive nephropathy, hyperlipidemia to chronic renal failure and hemodialysis. This study examinated flavonoids contained in extracts of Epilobii herba and Ononidis radix as scavengers of superoxide radical. Extract of Epilobii herba is used in the treatment of the first symptoms of benign prostatic hyperpasia. It inhibits the intracellular prostatic enzymes 5 - α - reductase, which converts testosterone to dihydrotestosterone. It has anti - inflammatory effect too. Ononidis radix is component of a lot of diuretic and urologic tea mictures. This drug facilitates urine secretion, lowers risk of creation gravel and stones renal in kidney and in bladder. The aqueous extracts of Epilobii herba and Ononidis radix and the aqueous extracts of selected flavonoids contained in diuretic acting drugs were tested. Superoxide radical was generated by the NADH/PMS system in phosphate 19 mM buffer, pH 7,4. Flavonoids were quantified by spectrophotometry and HPLC. Epilobii herba contains 0,4827 ±...

Education and gender - causes, relationship and possible solutions to the issue of gender inequality
Kubištová, Markéta ; Sycha, Martin (referee) ; Beneš, Milan (advisor)
The subject matter of this paper is searching for relations established between one's biological sex, gender (the social construction of sex) and position in the society. It discovers the causes and correlations of unequal attitude towards men and women, as well as it attempts at finding possible solutions of the issue by means of education. The first chapter explains the basic terms and relations between them, the creation of human biological sex and establishment of gender. The term is explained as socially conditioned cultural differences between men and women. The chapter also states certain theoretical standpoints clarifying gender construction, in particular the process of socialization of an individual, during which the person's gender is created. The aim of the second chapter is to describe the role of educational institutions and the system of education in establishing and maintaining social inequalities. It explains the mechanisms of schools contributing to socialization of a person, with a special emphasis on creating, or supporting gender identity of a child. The work of teachers and their attitude to pupils is an important aspect supporting the traditional perception of gender roles. The purpose of the third chapter is to define the scope of education of adults, its basic principles, goals and...

Žáková, Alena ; Hazuková, Helena (advisor) ; Hušková, Julie (referee)
Anotation: Žáková, A.: Expression of the spatiality in areal art creation of children in a period before their compulsory education. /A diploma work/ Prague 2006 - Charles University, Pedagogical Fakulty, the Art Department, 153 p. + riders (text ones: no.: 1 . - 5 . , CD presentation: no.: 6. - 69 of kindergarten children's art work). Subject: A diploma work theme is elaborated in a form of the qualitative pedagogical research and it is divided into two parts - theoretical and empirical one. Developmental psychology knowledge and children's art expression ontogenesis, with a view to a spatial relationship presentation as well as the problems of space perception, are compiled and arranged in the theoretical part. The main aim of this work was to elaborate and verify the examination methology for a representative sample of child population. On the basis of children's work analysis results then to bring the actual information about the standard of the third dimension in art expression of preschool children that could become a way out for pedagogical diagnostics. The part of an empirical study is focused on the examination of teachers' professional competency standards in kindergarten schools. The research proved that the competency standards as well as the teachers' motivation can influence the form and the...
