Růst Mycobacterium smegmatis na agarovém médiu a agarovém médiu pokrytém celofánovou folií - morfologická a proteomová studie

Biofilm formation is one of the most common bacterial survival strategies. Majority of bacterial species are able to form these three-dimensional structures, including pathogens like Mycobacterium tuberculosis. Representatives of Mycobacterium genus widely occur in the nature, although they can cause serious problems when they appear in medical equipment and artificial replacements of the human body. Non-pathogenic Mycobacterium smegmatis mc2 155 was used as a model organism in our experiments. We investigated morphology of the three- and six-day-old colonies (in fact biofilms) on agar and agar covered with cellophane using Stereo microscope and Scanning Electron Microscope. We found that a type of surface as well as a carbon source has a great influence on the morphology of the M. smegmatis colonies. We isolated proteomes from the agar and cellophane cultures and from planktonic culture. Two-dimensional electrophoresis was used as the main proteomic method. Proteomic data were analyzed using PDQuest software. Then the sets of proteins detected by qualitative and quantitative analyses were compared using Venn diagrams. Vladimíra Vroblová, Ph.D. Title of diploma thesis: Comparison of the immunophenotypes of normal and pathological plasma cells - determination of the rate of risk of progression in patients with MGUS Four-color flow cytometry is a necessary part of plasma cell imunophenotypisation today. It allows to define plasma cell count with a high accuracy and to distinguish normal and pathologic cell on the base of surface and cytoplasmatic markers expression. An expression of the following surface markers, CD138, CD38, CD19, CD56, CD45 CD20 and CD27, was monitored in this dissertation. An expression of the cytoplasmatic light chain κ and λ followed which confirmed monoclonality of the plasma cells in patients with multiple myeloma or with monoclonal gammopathy of undetermined significance. Theory and Practice an the Competence of Graduate Nurse. Working in the Surgical Ambulance This bachelor thesis deals with the contemporary situation and new trends in health service - in the branch of nursing. It presents and evaluates theoretical assumptions of further development of a graduated nurse in new conditions. The empirical part is aimed at identification of the conditions for the work of a graduated nurse and to recognize whether the current legislation enables such a nurse to work individually, concretely in surgical ambulance. The thesis concludes that graduated nurses are allowed to work individually and separately conformably to the laws, and patients and insurance companies take concern in their work. On the basis of this fact some questions arise: Why is there a problem to implement it in practice? Do we really take care in graduate nurses?

Utilization of biotechnological method in growing of technical hemp

Hemp (Cannabis sativa L.) is a multi-use crop, able to provide fibre celulose and hurds for industrial treatment biomass for energy conversion and produces secondary metabolites useful for pharmaceutical application. For its resistence to stress and ability to accumulate high concentration of heavy metals it can be potentialy used for phytoremediation. The aim of current research is the development of new strains, with specialy improved qualities and the enhancement of its applicability. Besides traditional breeding methods, genetic manipulation might be the possible tool. What is currently discussed is finding optimal conditions allowing efective in vitro cultivation as a basic assumption of genetic manipulation. Hemp transformation via Agrobacterium tumefaciens co-cultivation might be the appropriate technic.

The Russia policy of Germany 1917-1918

German foreign policy and military activity on the Eastern Front between 1917 and 1918 was primarily determined by effort to make peace and to stabilize the situation there, which would allow the transfer of command of the German troops to the western line. There was necessary to determine the current impasse before landing of the first sections of the United States in the spring of 1918. Their presence would reverse the current ratio of balanced forces of both warring parties against the Germany. This policy has been influenced by several interconnected factors. Besides the already mentioned time limit on troop movements between fronts there was a important relationship with the new Government of St. Petersburg from November 1917. Despite the Germans initially envisaged cooperation in the negotiations on the basis of support for the Bolsheviks on their way to power, but then there was a fundamental contradiction and friction surfaces, which were partly caused by the differing views on the spread of revolution among the Bolsheviks themselves. Vliv kůrovcové kalamity na společenstva epigeických Collembola (Insecta: Entognatha) klimaxových smrčin v Národním parku Šumava, Česká republika

Soil surface activity of epigeic communities of Collembola were studied in undamaged climax spruce forests, in dead spruce forests after bark beetle attack and in clearings in the Šumava National Park, South Bohemia, Czech Republic. Five pitfall traps were exposed at each of the nine study sites. They were exposed for 12 months during summer, fall and winter periods from 1999 to 2000. More than 149 000 specimens of Collembola belonging to 54 species were trapped, determined and evaluated. The highest epigeic activity of Collembola was in the summer period in the undamaged spruce forests (624.2 ind./trap/30 days), lower in the dead forests (604.1 ind./trap/30 days) and lowest in the clearings (549.8 ind./trap/30 days). The undamaged spruce forests had 19, whereas dead forests only 14 and clearings 17 characteristic species with constancy C > 49 %.

Title of Diploma Thesis: Application of the SIA system for aluminium determination This diploma work deals with determination of aluminium ions in samples of surface water in a sequential injection system. For determination of aluminium ions method described in ČSN 83 0520 part 22 B was used. Determination was based on complex formation between aluminium ions and aluminon. The incurred complex is colored and color intensity is directly proportional to aluminium ions concentration in the sample. In this work spectrophotometric detection was used. Primarily, an optimization of reaction conditions in the test tube was performed. The calibration graph was linear in the range 0.02 - 1 mg/l Al with correlation coefficient R = 0.9932. Also detection limit (LOD = 0.0119 mg/l) and quantification limit (LOQ = 0.0390 mg/l) were assumed. Consequently reaction was converted into the SIA system and optimization of reaction conditions in the SIA system was performed. Application of magnetic particles for isolation and purification of DNA

With a development of molecular biology methods it is an increasing interest in new procedures of DNA isolation of high quality. DNA isolation is performed on crude cell lysates by many techniques e.g. phenol extraction, salting out or adsorption on solid phase. Classical DNA isolation, such as phenol extraction is quite complicated and time consuming. New alternative methods of DNA isolation was development using reverse immobilizing DNA to a solid phase. Widespread is the use of the magnetic particles as carriers, which allow the isolation of DNA in high quality directly from crude cells lysates of complex samples. The current method of DNA adsorption onto the surface of magnetic particles does not provided sufficiently pure DNA for analysis of some comlex samples (e.g. food). Some inhibitors of the polymerase chain reaction (PCR) are apparently adsorbed onto the tube wall and the next step of DNA elution leads to their release into the solution and cpnsequent negative effect on quality of DNA (e.g. decreasing of PCR amplification). The principle of the developed procedure is design a device, which utilizes transfer of magnetic particles by paramagnetic newddle from one to another Eppendorf tube, in which further processing of the sample extends. Transfer of magnetic particles with DNA using needle prevents transmission of contaminating impurities. The proposed device allows to realize above-mentioned procedure. The functionality of the device being tested in the isolation of plasmid pUC19 DNA from crude lysates of E. Coli JM 109 (pUC19).

The basic mission o f the state is to create the conditions for the stability and to regulate the social climate, to absorb the social tension and by the development o f social activities to stabilise the situation in the society in order to enable the social subjects fully develop, affect and realise their targets by their own forces. These efforts intend to establish the conditions for the social defensibility of the social reform and/or minimize its impacts. In order to assure this target, the state disposes by the wide system of legal, financial and institutional instruments. The aim o f this thesis is the global evaluation of the actual legislation on the security of the people in socially indisposed situation; this people enters under the application of the Act No 117/1995 Coll., on social state aid, Act No 108/2006, on social services, Act. No 110/2006 Coll. about the existential minimum and Act No 111/2006 Coll. on aid in material necessity. When I evaluated the practical problems with the realisation of these systems I have resulted from the experience o f the employees of the Employment office in Tábor and of the Department o f social affairs and public health o f the Office o f region South Bohemia.
