Title: System to support decission making in dental implantology - quality of life of patients with dental implants Author: MUDr. Pavel Kříž Department: Paediatric Stomatology of 2nd Medical School and Faculty Hospital Motol, V Úvalu 84, 150 06 Prague 5 Supervisor: Prof. MUDr. Taťjana Dostálová, DrSc., MBA Supervisor's e-mail: tatjana.dostalova@fnmotol.cz Dental implants are the method of choice in the treatment of missing tooth/teeth replacement. Implant therapy must be preceded by a detailed examination and the overall treatment plan. As an aid for decision-making for dentists was created decision-making scheme, which gradually, logically and schematically guides the dentists in this particular situation. Health is closely related to the quality of life. Our work evaluates the oral health-related quality of life (OHRQoL) of patients with dental implants. The aim of our study was to determine whether treatment with a dental implant(s) improve(s) OHRQoL. We created a questionnaire to determine the quality of life before and after implantation. In our study, we evaluated only patients who were treated by the only one implantological system to eliminate the influence of other systems on the quality of the results of the study; we evaluated a total of 297 implants. It was assessed a total of 97...

Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Mgr. Rudolf Andrýs Supervisor: Prof. Ing. Vladimír Wsól, Ph.D Title of dissertation thesis: DEVELOPMENT AND APPLICATION OF AFFINITY CARRIER FOR ISOLATION OF HUMAN CARBONYL-REDUCING ENZYMES For several millennia the human medicine is based on application of small bioactive molecules that are administered in the form of plant extracts or synthetic compounds. However, their use in modern medicine is not possible without a detailed understanding of their biochemical effects and identification of their molecular targets. Chemical proteomics based on the specific recognition between the bioactive molecule and the target molecule is currently the most widely used techniques for identification of molecular targets of small molecules. Compared to conventional biochemical methods (e.g. 2D electrophoresis), chemical proteomics represents particularly sensitive and very selective technique that enable successful identification of biomolecules from complex biological samples that are naturally presented in very small concentrations. Carbonyl- reducing enzymes, which play an important role in physiology due to their involvement in metabolism of various endogenous (e.g. prostaglandins, steroid...

Charles University in Prague Faculty of Science Department of Biochemistry Studies of NK cell receptors and other proteins using recombinant expressions and mass spectrometry Summary of Ph. D. Thesis Daniel Kavan Supervisor: Prof. RNDr. Karel Bezouška, DSc. Prague 2010 Daniel Kavan Introduction Introduction NK cells and CD69 as one of their surface receptors Natural killer cells (NK cells) are the subpopulation of large granular lymfocytes, which lacks the surface receptors typical for B cells or T cells. They are characterized by the presence of NKp46 and NKp30 [Moretta L. et. al. 2002], however. They were named natural killers according to their function in the organism, as they do not need any activation and nevertheless they are able to eliminate abnormal (i. e. infected or transformed) cells from the tissue [Kiessling R. et. al. 1975]. This function is dependent on scanning the major histocompatibility complex (MHC) class I molecules of ambient cells. The resulting action (killing or not killing the target cell) is dependent on the balance of activating and inhibiting signals mediated by the NK cell surface receptors and forwarded to the specific signaling pathway [Raulet D. H. et. al. 2001]. Specificity of NK cells is not based only on one type of antigen receptor as it is in case of T and B cells,...

Monitoring of selected markers of damage to the colon after induction of chrohnic colotis Pavla Římalová 2010 Charles University in Prague, Faculty of Pharmacy in Hradec Králové Department of Pharmacology and Toxicology Crohn disease and Colitis belong to the Infammatory Bowel Diseases. AHR is Arylhydrocarbon receptor, which was suggested to affect the activity of immune system. Bilirubin is endogenous substance with significant antioxidative and anti-inflammatory effects. In this pilot thesis, we wanted to study the expression of AHR and large intestine wall after the induction of chronic colitis in normobilirubenimic (Wistar) and hyperbilirubinemic (Gunn) rats. Sixteen rats for Wistar and sixteen rats for Gunn were used in the experiment. Eight rats from each strain were administered by dextran sulphate (DSS) in drinking water for 7 days. All animals were on drinking water for another 14 days. This cycle was repeated three times. Immunohistochemical staining revealed the expression of AHR muscular layer and muscularis mucosa in all experimental groups. Administration of DSS resulted in increased expression of AHR in epithelial cells in Wistar rats. AHR expression was higher in non-treated Gunn rats when compared with non-treated Wistar. DSS treatment resulted in decreased expression of AHR in...

Establishment and phenotyping of three mouse congenic strains.
Flachs, Petr ; Trachtulec, Zdeněk (advisor) ; Vopálenský, Václav (referee)
Mouse congenic strains are widely used as an important genetic tool for various purposes of research. Congenic strains are special inbred strains in which part of the genome of one mouse is transferred to another by backcrossing the donor mouse to the recipient strain with appropriate selection. The classical ten-generation breeding protocol is commonly utilized and adapted for different experimental needs. Our department is focused on the study of hybrid sterility between two related mouse strains M. m. musculus and M. m. domesticus. The first aim of this work was the utilization of congenics to stabilize three transgenic lines, which previously helped to identify first mammalian hybrid sterility gene Hst1 as Prdm9 (Mihola et al, 2009). Bacterial artificial chromosomes (BACs) present in transgenes encompassed several genes including Prdm9. The second goal was to look for some new phenotypes of our congenic strains, which may be causally associated with genes encoded by the BACs, as well as an additional research on the hybrid sterility phenomena. We established three new congenic strains on C57BL/6J (B6) background and found significant differences between transgenics versus their littermate controls in weights of testis, gonadal fat, body, liver, and heart.
