Influence of Explosive Hardening on the Structure and Characteristic of Hadfield Steel in Terms of use in the Railway Transport

The high alloyed austenitic manganese steel, the Hadfield steel, thanks to its good wear resistance of the work surface and maintaining the high toughness of the internal material in the same time, is successfully applied for casted crossings. The crossings are the most dynamic stressed components in the railway turnouts. Thanks to low surface and sub-surface hardness the occurrence of plastic deformation and the progressive wear of the crossing running surfaces can be found since the initial stages of the crossing operational life. One possibility how to increase the surface and the sub-surface hardness and this way improve the dimensional stability and the crossing lifetime as well is to apply the explosive hardening of the crossing running surfaces. The technology mentioned above means the application of the explosives in the imminent nearness to the running surface of the crossing, when the high pressure wave acting within the extremely short period actuates the plastic deformation of the material structure. The work deals with analysis of influence of explosive hardening on the structure and characteristic of Hadfield steel. The explosive hardened samples have the surface and the sub-surface hardness checked and the microscopic analysis and X – ray diffraction is applied, including the TEM analysis as well carried out by the transmission electronic microscope. The characteristics experimental testing including outcomes from assessment of the contact – fatigue load of the explosive hardened Hadfield steel samples are the part of the work as well. As an experiment final part the outcomes of the long –term validation, the surface hardness and wear of the crossing running surfaces of the explosive hardened crossing already installed into the Czech Republic railway track are introduced as well.

The thesis is focused on the identification of service and administrative processes and sub processes inside the chosen company and proposal of their possible separation and outsourcing using the Business Process Management methods. Potential changes should be aimed to release capacity of the monitored team of the company and optimize their processes and related capacity during busy season and also from the long term perspective. The company set the conditions for potential changes by keeping current standards about quality of products and services and keeping current rules for client communication. Additionally company expects decreasing of personal costs related to inquired services.

Implementation of Total Quality Management System in Small or Medium Service Enterprise

Úspěch společnosti poskytující služby je závislý na efektivním využití kvalifikovaných lidských zdrojů. Na rozdíl od výrobní společnosti u firmy poskytující služby není vždy proveditelné zcela standardizovat výstupy, proto efektivita společnosti je závislá na optimalizaci vnitrofiremních procesů a přesném vymezení rolí u jednotlivých pozic. Principy Total Quality Managementu aplikované na společnost poskytující služby zajistí standardizaci fungování a vnitrofiremních procesů společnosti při zachování potřebné flexibility, což z dlouhodobého hlediska společnosti poskytující služby přinese výhody oproti konkurenci, zefektivnění řízení organizace a zvýšení návratnosti investice pro majitele/investora. Součástí práce je návrh implementace TQM připravený pro SME zabývající se poskytováním IT služeb.

Management and production of recording of folklore ensemble

Main objective of this thesis is to create a sort of guide for a wide range of educated public, which would ensure covering of the financial aspects of projects focused on preserving our intangible cultural heritage. Funding such projects primarily uses subsidies such as grants, whose system is quite administratively demanding, which can be considered a negative, in various cases even demotivating factor in provision of these financial resources. The thesis should therefore provide a detailed description of practical actions set up on theoretical bases of this issue, based on the current state of our traditional culture. Due to the practical output and capabilities of individual methods of processing applications for grants is work on real project essential for model demonstration of how to ensure mentioned financial resources. In this case of the recording of Prácheň folklore music is preservation of intangible cultural heritage and its preservation in subsequent form of music media the key issue and challenge for future direction of both, regional and national culture.

Changes of enzyme activities in fruits during long-term storage

This study is focused on study of changes of enzyme and low-molecular weight antioxidants in different fruits during long-term storage. In theoretical part individual low-molecular weight antioxidants and enzymes are described. The main causes of fruit decay and some possibilities of fruit preservation and storage are summarized. As biological material some common fruits were chosen - green and red apples, peaches, plums and white grapes. The fruits were stored in laboratory, cellar, in refrigerator and in freezer. In freezing experiments some ways of fruit preparation and processing were tested and their influence on fruit antioxidant status was compared. Shortened storage experiment was applied on blueberries, cranberries, raspberries and strawberries too. In fruits some group parameters – total antioxidant status, dry mass content, ascorbate level, total flavonoids and total phenolics were analyzed spectrophotometrically. Individual flavonoids and phenolics were determined by RP-HPLC/UV-VIS and on-line LC/PDA/ESI-MS. Antioxidant enzyme activities (superoxide dismutase SOD, catalase CAT, polyphenol oxidase PPO and lipoxygenase LOX) were measured by spectrophotometry. The surface microscopy and cultivation of moulds from fruit surface were performed too. Influence of storage conditions on biological activities is dependent on fruit sort. Freezing is the most suitable procedure for long-term storage without significant changes of active substance content. Long-term storage in controlled temperature conditions and/or atmosphere is usable for fruits with longer storage period. In these fruits stabile levels of antioxidant enzymes are stored for relatively long time. Some of enzymes act synergistically. Enzyme activities differed according to storage phase; at the beginning mainly high SOD and LOX activities were observed. CAT and PPO are probably activated as defence systems in rippened and/or damaged fruits. Levels of total as well as individual low molecular weight antioxidants varied during storage in all sorts, generally, increased course with longer storage period can be observed.

Opportunities to Develop Tourism in Protected Areas

The thesis is devoted to the issue of tourism development in areas with specific protection. Due to the massive development of tourism, it is necessary to focus on its negative effects, which should be avoided. The performance is therefore sustainable tourism development, which proclaims the idea of preserving existing tourism attractions to future generations. Based on field research, structured interviews and questionnaires with tourism stakeholders identified the status of tourism in protected natural areas in the Czech Republic. Subsequently, the proposed options for tourism development in these areas. A case study is focused on the tourist neglected protected landscape area of Iron Mountain. Protection of cetaceans in international law

In four chapters that this thesis consists of is presented the protection provided to cetaceans by international law. The first chapter gives insight into the biology of cetaceans, names facts relevant to their conservation and its legal basis. The second chapter deals with legal regulation of whaling. It analyzes the history and current situation in the International Whaling Commission as the only global organization to manage the exploitation of large cetaceans, particularly in terms of adopting protective measures for the recovery and conservation of whale stocks. The third chapter focuses on the species approach in protection of cetaceans as endangered animals. The fourth chapter addresses the protection of cetaceans' environment. First it deals with the protection of marine environment in general and then with the protection of cetacean habitats.

Dental pulp stem cells

The aim of this dissertation study was to optimize the isolation and long term cultivation protocols for human dental pulp stem cells. The protocols which showed best results were used for cultivation of dental pulp stem cell isolated from exfoliated teeth (SHED). Additional aims were to characterize DPSC and SHED and prove their ability to proliferate over Hayflick's limit and differentiate into mature cell lines (osteoblasts, chondroblasts and adipocytes). In order to find optimal protocols for isolation of dental pulp from tooth, we tried three different approaches. During optimization of cultivation protocol we focused on decreasing amount of fetal calf serum (FCS) from 10 % FCS in cultivation media (most often used in literature) into 2 % and thus get closer to cultivation conditions suitable for clinical usage. We compared DPSC cultivated in three different media (medium with 10 % FCS, 2 % FCS supplemented with growth factors and media with 2 % FCS supplemented with ITS and growth factors). For characterization of DPSC and SHED we used basic biological properties (proliferation activity, viability, morphology), their phenotype and karyotype. Práce přináší výsledky inventarizačního průzkumu motýlů s denní aktivitou (Lepidoptera, Zygaenoidea, Hesperioidea, Papilionoidea) navrhované přírodní památky Střelnice u Semanína. Průzkum byl proveden v roce 2007 a byl zde prokázán výskyt celkem 47 druhů motýlů. V práci jsou komentovány nejvýznamnější nálezy a je doporučen vhodný management pro zachování a posílení populací motýlů.

The essays of Marina Tsvetaeva in Czech translations

Czech translations of six essays by Marina Tsvetaeva that focus on poetry writing and on the work of fellow-poets were examined, following a detailed analysis of Tsvetaeva's style and the historical and biographical context of her prose. The evaluation criteria for the translations were: accuracy in reproducing the author's meaning, the ability to distinguish the characteristic features of Tsveateva's style from the systemic features of Russian and the translators' creative input in reproducing especially the lexical and euphonic intricacies of the original. In addition to providing a general overview of translation problems posed by the author's style the work attempted to trace differences between the approaches of two translators of Tsvetaeva's princi pal essays on art: Jana Štroblová (Art in the Light oj Conscience) and Jan Zábrana (Poet and Time, The Epic and Lyric ofContemporary Russia). All Czech translations were found seriously lacking in accuracy; the errors in the reproduction of meaning are partly due to the idiosyncrasies of Tsvetaeva's style (e.g. punctuation), but sometimes to sheer carelessness.
