Jelínková, Iva ; Chocholouš, Petr (advisor) ; Vlčková, Hana (referee)
Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Analytical chemistry Candidate: Iva Jelínková Supervisor: PharmDr. Petr Chocholouš, Ph.D. Title of diploma thesis: Solid-phase extraction and its miniaturization by method Lab-On- Valve for determination of pharmaceutical substances This thesis deals with the development of the solid-phase extraction (SPE) method with the use of microextraction by packed sorbent (MEPS) for the determination of vitamins A, D, E and the possibility to automate this method. The determination was based on absorption of vitamins on microcolumn MEPS, interfering components were removed by washing solution (dilute acetic acid of pH 3) and extraction was performed by eluent solution (100% ACN). Detection was made by UV spectrophotometer at wave lengths of absorbing maximum for vitamin A - 325 nm, for vitamin D - 265 nm, for vitamin E - 295 nm. The method was optimized. There was developed a program, where concentrations of individual vitamins, dosing volumes, flow speed and composition solvents were tested. After the optimization method there was performed the extraction and the subsequent determination of blood plasma fortified with vitamins A, D, E by the sequential injection chromatography method. Recovery of the method for vitamin A was...

Autoevaluation of secondary school
Dostál, Filip ; Vališová, Alena (referee) ; Koťa, Jaroslav (advisor)
The diploma work deals with the introduction of grammar schools auto-evaluation process together with the curriculum reform in progress. Analysis of school legislation in force and of binding curriculum documents led to the conclusion that schools autoevaluation is the main and almost the sole mechanism of control and quality and efficiency improvement of grammar school education. This fact contrasts markedly with the marginal position of auto-evaluation in the legislation in force and curriculum documents. Some theoretical basis for grammar schools auto-evaluation were defined and critically reviewed through compilation of literature; they were interpreted in the light of post-modern, positivist and pragmatist philosophical concepts; constituent review was based on neo-tomist positions. The focus of the work lies in analysis of auto-evaluation phases recommended in literature. What is new is a concept of "pre-evaluation" phase which assumes a metaevaluation character although preceding the evaluation itself. A two-phase questionnaire survey was devised and applied at the curriculum reform pilot grammar school for this phase. Finally, two tools for auto-evaluation of study results of grammar school students were devised - a statistical analysis of grammar school graduates results of admission to...

Lawfull use of force
Lipert, Jan ; Vokoun, Rudolf (advisor) ; Herczeg, Jiří (referee)
of my Master's degree thesis in English Lawful use of firearms The purpose of my paper is to draw attention to the lawful use of firearms in self- defence. The relation between the lawful use of firearms and self-defence is not explained sufficiently today. For example, it is not clear in what situations police officers have authority to act in self-defence and what their liability would be should they exceed the regulation of lawful use of firearms. The author offers possible solutions to this issue and describes their consequences. The paper is composed of three chapters. Introduction brings the reader into the issue. First chapter serves as an extension to the introductory part and defines basic terminology used in the paper such as defences, necessity, self-defence, dangerous offender, lawful use of firearms. Chapter Two deals with relevant international, constitutional and statutory legislation with a focus on the lawful use of firearms and its regulation. Chapter Three introduces basic knowledge from practice split into rulings and case studies. Author of this Master's degree thesis searched and chose the most relevant rulings with relation to the lawful use of firearms. Case studies include two cases of lawful use of firearms by the Police to which the author had access. Chapter Four looks...

Working Motivation of Nurses
FLORIÁNOVÁ, Michaela
Current status: The word motivation means to move. A motive is a factor that forces us to move ahead and meet our goals. The American Professor Frederick Herzberg dealt with motivation and compiled the motivation-hygiene theory of motivation. In this theory he focused on the satisfaction and dissatisfaction of respondents with work and the circumstances that led to those feelings. (2, 3). Subject: The aim of the research was to explore the extent to which hygiene (maintenance, external) and motivation (internal) factors affect job satisfaction of nurses. Methods: To obtain the necessary data for processing the thesis, quantitative research was used and it was conducted using a non-standardized questionnaire. Nurses and ward nurses in hospital wards in České Budějovice were involved. The theme of the questionnaire concerned the motivation and external factors according to Herzberg. The questionnaires were processed by SPSS, a non-parametric correlation test, and for evaluating the response rate a descriptive statistical function was applied. For statistical confirmation of the hypotheses was chosen 5% level of significance. Participants: In the study were included shift nurses and ward nurses in hospital wards in České Budějovice. A total of 250 questionnaires (100%) were distributed and 143 questionnaires were returned (57.2%). Nurses were contacted at the Department of Dermatology, ENT, ophthalmology, internal, surgical, rehabilitation, aftercare, ARO, children and urology wards. Results: The evaluation of the external factors of work motivation were observed in the following areas: policy and administration, relations with superiors, relationships with workers of the same level, salary, job security, personal life, working conditions and status. The results show that nurses are not satisfied with the wages (74.2%) and in their opinion, wages do not correspond with the work done (78.4%). Respondents pointed out that there are frequent conflicts within the team (52.5%) and lack of involvement in decision-making (57.4%). On the contrary, most nurses (92.3%) feel good among colleagues and say there is also a good atmosphere in the workplace (89.5%). Interpersonal relationships at work are also evaluated positively (88.8%) and the majority of nurses (93%) get along well with colleagues. The following areas were monitored within the evaluation of the motivational factors: recognition, promotion, work itself, the opportunity for personal growth and responsibility. Evaluation of the motivating factors revealed five negative areas. One of them is the stipulation of benefits given by the employer for work done above the norm; nurses responded that they do not receive these advantages (83.9%). According to the respondents, nurses do not have the opportunity for professional growth (51.1%) and promotion (74.2%). At the same time, in their opinion the occupation of a nurse is not considered prestigious (76.3%).They do not get recognition from superiors for successful or quality work (56.7%). On the contrary, a positive finding is that nurses have the option of continuing education (88.1%) and find self-fulfilment in their work (76.9%). Nurses get recognition from patients (62.2%). Conclusions and recommendations for practice: The results of the survey show that most nurses surprisingly answered both the motivation and external factors positively. The greater presence of external and motivation factors increased job satisfaction of the nurses. Their biggest problem was considered wages, which in the opinion of nurses does not correspond to the workload. The motivating factors fared best were training opportunities, good acceptation to the team and that the nurses enjoy the performed work and it suits them.

Horyna, Roman ; Finková, Dana (advisor) ; Gnad, Tomáš (referee)
Title of the thesis: Current concepts of double poling by classic technique in cross-country skiing Object of the thesis: The spatial-temporal analysis of node points and kinetic phases of the kick double poling and double poling of selected competitors at the FIS Nordic World Ski Championships in Liberec 2009. Method: A case study in which we used the kinematographic method for the analysis of the kick double poling and double poling of selected competitors. We analyzed the video of the 15-km classic in the FIS Nordic World Ski Championships in Liberec 2009. For the analysis we used the software APAS. Results: We performed spatial-temporal description of node points and kinetic phases of the kick double poling and double poling of selected competitors. We characterized current concepts of kick double poling technique, because selected competitors were relatively homogeneous set. By double poling we found out flexion-extension pattern in elbow, hip joints of slipping lower limb and partly in knee and ankle joints of slipping lower limb which results in generation of great propulsive force. Double poling becomes more significant driving element than push-off. Keywords: cross-country skiing, kick double poling, double poling, kinematic analysis, top competitors

Legal aspects in fulfilling of Framework educational programme for pre-school education
FAROVÁ, Lenka
Framework educational programme for pre-school education has binding force for all pedagogical workers in all types of pre-school institutions. Even all legal regulations has also binding force for these workers, and they are obliged to adide them. The framework programmes are made by people with pedagogical experience, whereas laws are made and approved by lawmakers. Both them and school authorities to whom some considerable competences were given, are people from different branches and most of them, unfortunately, do not have any school experience. The theoretical part of my bachelor's thesis deals with a general theory of law, some laws related to the pre-school education and a system of educational programmes. In the practical ( research ) part, I have mapped some contradictions between legal regulations and a current school life. This bachelor's thesis includes my own suggestion how to deal with and solve some concrete contradictions between legislation and the framework educational programme for pre-school education.

Liability in labour law
Dáňová, Lucie ; Hůrka, Petr (advisor) ; Štefko, Martin (referee)
Liability in Labour Law Summary Liability represents one of basic legal institutes which applies in most branches of law, including labour law. However, employment relations have several distinctive features which are also reflected in the conception of lability. Since the consequences of violation of law can substantially affect the position of violator, it is necessary to be aware of one's duties and a possible occurrence of lability. The aim of my thesis is to analyze liability in labour law according to the legislation in force. Liability in labour law is not only a theoretical concept but it also has practical impacts on both subjects of employment relation, therefore it is desirable to have knowledge of this institute. Since liability denotes one of the most complicated institutes in law it is, at first, essential to explain the concept of liability in general. Legal theory construes liability in several different ways and it is interesting to mention opinions of various theorists. The next chapter characterises the concept of liability in labour law and describes its main features. The following chapter deals with division of liability in labour law into several kinds from different points of view. The thesis focuses on liability for damage because of its importance in labour law itself. Chapter five...
