Micková, Zuzana ; Jarošová, Eva (advisor)
This diploma thesis is analysing the conditions of leadership development for potential leaders in Hilti spol.s r.o.. In this intention the theory of leadership is elaborated, further existing processes inside the company are described and finally data derived from Hilti internal research are evaluated. Information is supplemented by the findings derived from an e-questionnaire created for the purpose of clarifying uncertainties from the previous research data. To identify the objectives the thesis aligns existing processes with the theory of leadership and with the empirical researches. Exploiting these data within the company it is possible to identify strengths and weaknesses of leadership development processes for young leaders as well as propose areas which would be capable of implementing new trends with focus on higher effectiveness of these processes.

Colloid properties of hyaluronane sols
Krouská, Jitka ; Mašek, David (referee) ; Pekař, Miloslav (advisor)
Předmětem této práce bylo popsat agregační vlastnosti kyseliny hyaluronové (HA) a jejích hydrofobně modifikovaných derivátů. Za tímto účelem byly připraveny vodné roztoky kyseliny a dvou derivátů, lišící se stupněm substituce a druhem anorganické soli jako prostředím. Koncentrační řady čisté kyseliny hyaluronové a jejích derivátů byly studovány dvěma metodami, tensiometricky a spektrometricky.

The thesis is focused on the main aspects of market risk measurement and hedging and emphasizes the potencial problems of financial derivatives arranging by small and medium enterprises for this purpose. The teoretical part describes general methods of risk quantification and points out their main adventages and imperfections, then it introduces the hedging financial instruments which are offered on the market. In the practical part, according to the case study, my aim was to highlight possible ineffectivness and counterproductivity of hedging by complicated derivative instruments.

Synthesis of xanthone derivatives by microwave-assisted methods
Vrbata, Petr ; Macháček, Miloš (advisor) ; Vinšová, Jarmila (referee)
Synthesis of Xanthone Derivatives by Microwave-assisted Methods Petr Vrbata This diploma thesis deals with preparation of xanthone derivatives as inhibitors of growth of human cancer cell lines. A new methodology for synthesis of dihydropyranoxanthones 1 and 2, based on the usage of heterogeneous catalysis combined with microwave irradiation, was optimized. This allowed decreasing significantly the reaction time and improving yields. Along with this the Grover, Shah and Shah method (J. Chem. Soc. 1955, 3982) for synthesis of xanthone derivatives was improved due to usage of microwave heating. Using this method a new xanthone derivate 5,8,10-trihydroxy-7H-benzo[c]xanthen-7-on (3) was prepared. Its structure was verified by IR, 1 H NMR and 13 C NMR spectra. Synthesis of NK1 antagonists.
Puchnerová, Lucie ; Doležal, Martin (advisor) ; Zitko, Jan (referee)
Synthesis of NK-1 antagonists Lucie Puchnerová Department of Organic and Pharmaceutical Chemistry, Faculty of Pharmacy University of Seville NK-1 receptor antagonists are represented so far by only one drug, Aprepitant, which is available on the market for prevention of chemotherapy-induced nausea and vomiting. Nevertheless, it is assumed that NK-1 antagonists will be able to participate in treatment of migraine, rheumatoid arthritis, asthma, pain, inflammatory bowel disease, Parkinson's disease, anxiety and depression in future. This thesis is concerned with asymmetric synthesis of derivatives of 2-amino-4H-pyran, which thanks their substitutions are in accordance with structure of pharmacophore of NK-1 antagonists. As a starting material for their preparation has been used commercial methyl p-tolyl sulfone and (R)-methyl p-tolyl sulfoxide, which was synthesized from menthyl-(S)-p- toluenesulfinate by nucleophilic substitution. These compounds were subjected to reaction with ethyl 2-picolinate. From prepared β- ketosulfoxide and β-ketosulfone were obtained 2-amino-4H-pyrans by Michael addition. The derivatives proceeding from β-ketosulfoxide were subjected to reduction of the sulfoxide and trifluoroacetylation of the amine group on the carbon 2 of the pyran ring. Prepared derivatives had agonistic activity...

"European Identity" of a Teacher
Gröschlová, Andrea ; Walterová, Eliška (advisor) ; Chvál, Martin (referee) ; Janík, Tomáš (referee)
This work attempts to identify and analyse factors that have an effect on the identification of teachers with Europe: "European identity" of a teacher is being argued. We widely discuss actual meaning of the term "Europe" and seek an empirical approach to the "European identity" research. Correctness of the "European identity" of a teacher is not put to test; actual issue of the teacher's profession is being discussed instead. The structure of this work consists of three chapters. First of all some basic assumptions for the work need to be posted: we discuss such terms as "globalisation", "knowledge society", "educational aims" and context of "European dimension of education". The second chapter presents three interrelated parts. They are focused on the meaning of theme "teacher as a subject of research", "identity as a subject of research" and "Europe as a subject of research" . We conclude with the own meaning of the "European identity" of a teacher in the end of the second chapter. Empirical approach throughout the rest of the work is based on this conclusion. The methodological approach is derived from the character of the postulated hypothesis. We compare two independent samples. Two samples being a) teachers (teachers of basic and secondary schools in Prague) and b) random population. Based on the...

Park and Ride System (P+R) in Prague
Fenclová, Veronika ; Chmelík, Jakub (referee) ; Vondráčková, Petra (advisor)
The thesis concerns the traffic in Prague. It targets the potentialities of short-term parking on the city outskirts, on metro and train stations, and summarizes the conditions for people who combine personal and public transport in their commuting. The central part of the thesis focuses on the park and ride (P+R) scheme, and their location and availability. The bike and ride (B+R) and kiss and ride (K+R) systems are regarded as well. A questionnaire, carried out with respondents who regularly commute to Prague for employment or education, is evaluated. It comes out clearly that awareness of parking possibilities on the outskirts of Prague is low. And even if a respondent is aware of the possibilities, then the system is usually not used, for reasons that are summarized in the evaluation of the questionnaire. The goal of the thesis is to inform about P+R system and to highlight its shortcomings, which have been either deduced from the questionnaire or derived from P+R usage data. Keywords: system P+R, parking, commuting by car, Prague

Preparation and characterization of LmbX protein involved in lincomycin biosynthesis
Jiráčková, Petra ; Janata, Jiří (advisor) ; Janderová, Blanka (referee)
Lincomycin is an antibiotic used in clinical praxis. It is produced by Streptomyces lincolnensis. Lincomycin is composed of an amino-sugar and an amino-acid moiety linked by an amide bond. The amino-acid precursor is propylproline (PPL), whose biosynthesis undergoes the pathway derived from tyrosine. The modified PPL biosynthesis pathway was also discovered in pyrrolobenzodiazepines (PBD) and hormaomycin. In the biosynthesis of PBD the PPL precursor is further modified by reactions catalysed by specific enzymes missing in the biosynthesis of lincomycin. The genes encoding these enzymes could be transferred to the lincomycin biosynthetic gene cluster. In this way we could get producers of hybrid antibiotics with better properties and even antimalaric effects. Six enzymes participate in PPL biosynthesis, which are encoded in the lincomycin biosynthetic gene cluster. The first two reactions of PPL biosynthesis pathway are proven, therefore, this work focuses on the third reaction that is supposed to be catalysed by protein LmbX according to literature. The proposed function of LmbX is a hydrolysis of C-C bond. However, LmbX belongs to the protein family of isomerases by sequence homology. The protein LmbX was overproduced in this work and its activity was tested in the presence of the expected...

HPLC analysis of dexrazoxane and its metabolites
Dlouhý, Lukáš ; Kovaříková, Petra (advisor) ; Kučera, Radim (referee)
High performance liquid chromatography (HPLC) is one of the most used separating methods of instrumental analysis enabling both qualitative and quantitative analysis of substances. Anthracyclines are cytotoxic drugs widely used in clinical practice for treatment of hematological malignancies (leucaemias, lymphoms) and solid tumours (stomach, breast, ovarian cancer). Treatment by these drugs has a serious limiting factor, however. This factor is an anthracycline induced cardiotoxicity, which is caused by hydroxyl radicals and other reactive oxygen forms. The main role in their formation play the complexes with iron. Dexrazoxane is a derivate of bisdioxopiperazine, which is used to reduce the cardiotoxic effect of anthracyclines. It is supposed that it takes effect by the means of its active metabolite ADR-925, which works as a substance chelating iron. This mechanism hinders the iron dependant formation of oxygen radicals. This thesis deals with analysis options of dexrazoxane and its hydrolysis products/metabolites on the stationary phase Ascentis HILIC in conditions compatible with mass spectrometric detection (MS). The best results were achieved in conditions of isocratic elution using mobile phases containing 5 mM ammonium formiate with pH 5 or 7 and acetonitrile in ratio 22:78 (v/v). Mobile...
