Methodology of transgenic mouse preparation.

The thesis deals with methods used for preparation of transgenic mice and with comparison of them in terms of efficiency and suitability of applications for research purposes. Technologies for the transfer of gene constructs involve microinjection of DNA into the pronukleus of fertilized oocyte, which seems to be the most reliable one. Another possibility is associated with the use of sperm, which carry the incorporated gene construct into the oocyte during in vitro fertilization, or with the use of retroviral vectors by which the oocytes are transfected. Somatic cell nuclear transfer into enucleated oocyte, the use of embryonic stem cells which are incorporated into strange embryo or transgenesis by liposomes are techniques far less explored and not so often used. The next section of this thesis introduces to the reader individual steps which are necessery for successful transgenesis. It is focused on ovarian stimulation which is necessery to obtain sufficient amount of oocytes, as well as on stimulation of recipient mother in embryo transfer as well as on capacitation of sperm required for in vitro fertilization. The evaluation of energetic expenditure in pregnancy.

Background: Energy expenditure in pregnancy of Czech women is a lowly explorer part of energy metabolism. Objektive: The purpose of this study was to estimate total energy expenditure in Czech pregnant women in gravidity and to compare our values with values presented in foreign literature. Design: Energy expenditure was estimated in 137 Czech women. Total energy expenditure was estimated from filling date each activities, which pregnant women filled to questionnaires and activities missed in guestionnaires wrote to paper. Seven days activities was estimated in a specially designed program GraphPad Prism 4. As the values of total energy expenditure were considerated program values calculated multiplication factors (1,035; 1,062; 1,177 for the first, second and third trimester of pregnancy. There is described also other energy characteristics - i.e.basal metabolit ratein the study Result: In our study there was total energy expenditure 2993 ± 599,6 kcal/day on average.There was seen a gradual increase total energy expenditure 2415 ± 533,9 kcal/day for first trimestr, 2719 ± 457,3 kcal/day, for second trimester and 3223 ± 586,5 kcal/day for third trimester. BMI group <19,9 was 2295 ± 239,3 kcal/day TEE, BMI group 20 - 26 was TEE 2800 ± 383,3 kcal/day and BMI group > 26 was TEE 3554 ± 532,6 kcal/day....

Awareness of the brand Yves Rocher

This bachelor's thesis explores the awareness and perception of the brand Yves Rocher from the customer's perspective. The aim of this thesis is to evaluate awareness of the brand Yves Rocher on the Czech market and also to assess its perception of consumers. The first part of the thesis deals with the theory of brand and marketing research issue. The second, practical part includes the presentation of the company Yves Rocher and analysis of its competitors, an important part is also the questionnaire survey and its subsequent assessment. The conclusion of this thesis contains a summary of the results and some suggestions for improving the awareness of the brand on the Czech market and also for attracting more customers.

Ankylosing Spondylitis: Comparison of Diagnostic Methods and Digital 3D Model Production

Title:Ankylosing spondylitis, comparision of diagnostic methods and production of digital 3D model Aim: The aim of this thesis was to explore the issue of diagnosis of ankylosing spondylitis (AS). Describe the investigative methods used to diagnose and monitor the progress of the disease and try to create 3D geometric model of two vertebrae from magnetic resonance images (MRI) has demonstrated the possibilities of this process and possibly illustrate the pathological processes taking place on the monitored vertebrae. Method: The thesis is elaborated in the form of annotated literature review. They were used the available literature sources from which the information was drawn for determination of pathological processes, clinical symptoms, diagnostic methods and on therapy in AS. The practical part is used for creating 3D model of MR images in the program Scanview and Amira program for creating 3D objects. Software has Amira correction options and adding image data with a lack of meaningful that MR images show. N-gram language model for a Czech spellchecker

The aim of this thesis is to explore the possibilities of using n-gram language models for spellchecking Czech texts and to implement an extension to the spellchecker which would be able to find such misspelled words that are true Czech words. Furthermore, the aim was to implement a simple web application which would present the extended spellchecker. The influence of using lemmatization and morphology analysis of words regarding the hit rate of finding misspelled words was also looked into. The methods of language modelling used in the thesis are described first. What follow, then, is the description of the procedure of the spellchecking program using language models. The next part shows the way of getting the data for language model training. In the following part, the evaluation of the language models created is presented. The final part shows the results achieved for each option of spellchecking.

Analysis of the implementation and use of SaaS IS in the Russian market

This paper deals with the implementation of SaaS in the Russian market, especially in the Ural region of the Russian Federation -- Republic of Bashkortostan. SaaS is usually considered as an alternative way for the dominant model of software installation at the customer office. SaaS has certain key differences related to the use of different types of outsourcing and switching from buying products for permanent use to the system with subscription fee, depending on the specific use of resources. The goal of the theoretical part of the paper is to construct the comprehensive picture of Cloud computing, cloud services especially SaaS, to identify the strengths and weaknesses of SaaS technology, call attention to issues of risk, safety, the process of deployment, focus on the analysis of the status of SaaS information systems in the world and analyze situation on the Russian market. The goal of the theoretical part of the paper is to get acquainted with the companies in the Ural region of the Russian Federation, the Republic of Bashkortostan, to show their attitude to cloud-based technologies, highlight the exploration and use of SaaS in the Republic of Bashkortostan, compare the current situation in Russian enterprises with the status of SaaS in the world. By questionnaire survey I will obtain basic data of the awareness and use of SaaS in this region of Russia. The expected benefits of work may include extending the knowledge of readers about Cloud computing and one of its services - the form of SaaS. Next benefit is that Czech managers and directors can use this paper during the decision on the implementation of this model in their business.

Athanasius Kircher (1602-1680) and his Influence on the Natural Philosophy in the Czech Lands

Resumé: The dissertation deals with various layers of influence that Athanasius Kircher and his work had on the development of natural philosophy in the Czech lands around the half of the seventeenth century. It is possible to observe one such layer of this influence in his correspondence with the Czech lands, which often had patronage seeking character but was also a source of natural philosophic knowledge, observations and interests. Besides the correspondence influence, a diachronic influence and changing evaluation of Athnasius Kircher's work in the course of time is explored in one chapter. Another chapter introduces his own autobiography, which provides evidence of Kircher's conscious effort to shape the opinion of his person and his work to be made by his contemporaries and next generation of scholars. The last layer of influence being in focus of this work regards the influence of ideas based on an analogy between the movement of sea waters and movement of fluids in human body. Kircher's ideas on geocosmos expressed in his Mundus subterraneus (1664-1665) and Iter extaticum II (1657) are compared with the work of his correspondent, physician from Wroclaw and editor of the first medicine journal, Philipp Jacob Sachs von Löwenheim's Oceanus macro-microcosmicus (1664). The Spirit of a Festival

The theme of my thesis is the "A Detection of the Spirit of Pilgrimage". There is a pilgrimage in Europe, 1.500 km in length, which goes from the French town, Le Puy, to the Spanish town, Santiago de Compostela. The pilgrimage is widely known as "Camino de Santiago". My work focuses on exploring and finding the answer: What is the essence of pilgrimage? My work is divided into two parts; the first is theoretical, introducing the world of medieval pilgrimage from the very beginning to the present. The practical section consists of my own experience with the Camino, and the analysis based on the grounded theory. The thesis explains developement of the pilgrimage and the meaning of the Camino for cultural and religious life in Christian Europe, describes my own accomplished pilgrimage from Prague to Santiago de Compostela and deals with formation of a hypothesis based on the grounded theory. Development of the Czech form of the Patterns of Adaptive Learning Scales (PALS)

The undergraduate thesis explores validity of genuine Czech translation of PALS questionnaire in Czech environment. This method is built on Carol Dweck?s goal theory, which is described in theoretical part, also with basic background in the field of educational motivation. Research was conducted on the sample of 549 pupils in 8th and 9th grade and their results were compared with original research of Carol Midgley et al. (2000). Factor analysis and reliability analysis was done on used scales and it is obvious that simplification and language modification of items is needed, but still it appears from the results that this method distinguish goals correctly in Czech environment and it can be put into practice after further research.

English book titles in gerundial form

The category of gerund is a symptomatic component of the grammatical system of present-day English. Numerous studies have dealt with its nature and functions as well as with its delimitation as opposed to related forms of present participle or verbal noun. No matter whether the term gerund is used or not and whether it is theoretically sustainable to differentiate between the related -ing forms, it is obvious that the -ing form that in many contexts clearly displays both nominal and verbal features plays a crucial role within both written and spoken discourse. Nonetheless, apart from the gerund operating within continuous texts, it is revealing to explore its behaviour and functions in the text-frame components, particularly the title, i.e. a text-frame component with a crucial role in the printed publication of a book type. In this context, it is necessary to present 3 basic starting points of the present study: 1. the notion of gerund as a grammatical category is retained in this study (see 2.1.3); 2. a new term - gerund title l - is introduced in order to capture the analyzed structures (see the detailed characterization in 2.3); 3. the analysis is largely based on the functional approach.
