Style analysis of editorials in contemporary journals

Při zkoumání korpusu editorialů jsme mohli potvrdit naši hypotézu, že editorialy lifestylových časopisů Marianne a Maxim tvoří ideologickou bránu do prostoru časopisu a svou povahou se liší od ostatních článků uvnitř čísla. Editorialy mají dvě základní funkce, které se v menší míře objevují i v ostatních článcích. Jedná se o funkci identifikační a atrakční (blízkou funkci reklamní). Jazyk editorialu, téma a ideologické poselství slouží ke ztotožnění se (identifikaci) čtenáře jednak s autorem textu, jednak s ostatními čtenáři periodika. Po přečtení editorialu čtenář ví, zda celý časopis je skutečně "pro něj", nebo má sáhnout po jiném. Tato komunikace probíhá ve dvou směrech: cílová skupina čtenářů ovlivňuje podobu časopisu a zároveň je autory časopisu ovlivňována (a to zejména prostřednictvím editorialu). Postoj ke čtenářům se liší podle toho, zda se jedná o časopis určený ženám, nebo mužům: autorka editorialu ženského časopisu přes veškeré zdání rovnoprávnosti, které je čtenáři primárně předkládáno, vystupuje jako autoritativní rádkyně, zatímco autor editorialu v pánském časopise se staví zcela naroveň čtenáři, dokonce podceňuje své schopnosti, což můžeme hodnotit jako projev v pánském časopise všudypřítomného zlehčování a nadhledu. Transformation of Second Homes in The Kokořínsko Protected Landscape Area

The main content of this Thesis is the second home as a part of tourism and recreation - very popular form for leisure time. The aim of the Diploma Thesis is to monitor changes in the second home in a protected landscape area Kokořínsko. The comparison of the second home in Sudetenland and in the Protectorate of Bohemia and Moravia is emphasised. Although this district did not belong to the important areas of the second home from the very beginning, after the Second World War Kokořínsko became more developed, mainly in the terms of cottage building. The Thesis is divided into several parts. Individual parts are important for understanding the second home in this district. The Thesis deals with a brief history of the second home in Kokořínsko and characterises the Sudetenland with the whole area from the point of view of development and localization. Concerning research methods, the Thesis uses mainly a field research method, a questionnaire survey and a statistic data research. AJAX technology

This paper's goal is to provide a complex description of the AJAX technology. The paper provides detailed technical description of the technology and its parts; names the technology's positives and negatives resulting from its usage and also addresses its most commonly used alternatives (hidden frames, Flash, Java, ASP, NET, XUL, XAML). Further on, the paper addresses interesting JavaScript frameworks facilitating application development (Prototype, script.aculo.us, dojo, qooxdoo, YUI) or compatibility of the most commonly used browsers (DOM, CSS, ECMAScript). The paper's conclusion is dedicated to security of applications based on JavaScript, their optimalization and performance. Several recommendations for optimizing the JavaScript are tested on the most commonly used browsers and a wide selection of browsers - both Windows and Linux based - is subject to complex performance tests.

Práce se zaměřuje na úpravu odpovědného rozpočtování v jednotlivých zemích Commonwealthu a v Latinské Americe, kde tyto zákony byly přijaty. Zatímco v anglosaských zemích (zejména v USA) mají tyto zákony poměrně dlouhou tradici, v Latinské Americe byly obvykle přijímány spíše v nedávné době, a to většinou v souvislosti s problémy veřejných rozpočtů. Po analýze a komparaci úpravy odpovědného rozpočtování v jednotlivých zemích dochází práce k závěru, že latinskoamerické zákony jsou obvykle podrobnější, konkrétněji specifikují cíle a povinnosti vlády a většinou zahrnují i zřízení a financování určitého „stabilizačního“ fondu, jehož prostředky mají pomáhat v době ekonomického poklesu. Z analýzy dále vyplynulo, že ve federacích je odpovědné rozpočtování na federální úrovni upraveno obvykle spíše v obecné rovině, popř. federální legislativa upravuje především transparentnost informování o stanovených cílech a jejich plnění vládami. Konkrétnější úprava je pak ponechána v rukou jednotlivých států federace. Některé federace byly ovšem nuceny přistoupit k podrobnější federální úpravě, aby deficity jednotlivých států federace příliš nezatěžovaly centrální rozpočet. Obecně platí, že zákony o odpovědném rozpočtování obvykle stanoví určité cíle a limity, které musí exekutiva plnit, a pokud je v daném roce neplní, musí to napravit v letech následujících. Rozpočtová omezení mohou postihovat celou zemi, sankce typu ztráty části mzdy nebo zákazu výkonu politické funkce obvykle postihují konkrétně osobu (ministra, guvernéra), která svůj úřad zadlužila. Kromě analýzy přijaté legislativní úpravy práce také hodnotí efektivnost přijatých opatření, a to jednak na základě odborných studií, jednak podle empirických výsledků snižování veřejného zadlužení. Za rámec běžného plánování podníkání: budování internetového startupu v rychlé ekonomice

First part of the thesis compares available approaches on business planning, namely the traditional and newly emerged and proposes a Business Planning Approach Selection Framework that should help (newbie) entrepreneurs with orientation in the chaotic business planning world and choosing an approach suiting their specific needs. Conclusion of the theoretical part is that both the traditional and modern business plans have their role in todays business environment. While the old are great for external presentation and funds securing, the new enable to create more complex process for planning successful business from the original idea to and beyond their launch, building on customer feedback and gradual iterations. The practical part then builds on the theoretical research and aims to check feasibility of an actual business project, a multisided platform mediating sales of automotive financial products. It describes the model refinement and product creation process as well as eventually proves viability and feasibility of the whole business.

Mutational analysis of Potato virus X (PVX) coat protein

The thesis deals with the mutational analysis of conserved amino acids of Potato virus X coat protein (PVX CP). The importance of selected amino acids for the spread of the virus in the plant should be determined. Nicotiana benthamiana was selected as an experimental plant. Mutations of the PVX CP were based on the comparison of PVX CP amino acid sequence with the sequence of Papaya mosaic virus coat protein (PapMV CP), the only representative of potexvirus, which includes PVX, with the described crystal structure of the CP. The importance of certain amino acids for interaction of coat protein subunits PapMV CP,CP and PapMV CP,RNA and thus for virus particles formation was experimentally determined. The available amino acid sequences of isolates and strains of PVX CP were obtained from the National Center for Biotechnology Information (NCBI) database, compared with each other and alsowith the sequence of PVX CP used in the Laboratory of Virology, Institute of Experimental Botany of the CAS. Codons encoding conserved phenylalanine and lysine at positions 33 and 118 in the PVX CP amino acid sequence were mutated using methods of molecular biology. Five constructs of PVX CP mutants were prepared, Two deletion mutants of the PVX CP N-terminus which were created in the vector derived from PVX (pGR106) in which the cDNA sequence of PVX is integrated, the remaining 3 point mutants were prepared only as a product of SOE PCR. The reporter gene GFP for monitoring of infection and spread of mutants in plant tissue was cloned into pGR106 carrying the deletion mutants of the N-terminus of PVX CP (deletion of 2.-32. or 2.-33. amino acid). Both mutants are able to move only in a short distance from infected cells to adjacent cells within the inoculated N. benthamiana leaf. These two deletion mutants showed difference in the speed and in the extent of the GFP signal spread. Deletion mutant still possessing the codon for F33 showed faster onset of the GFP signal and was able to spread more rapidly to surrounding cells in comparison with deletion mutant, where the codon for F33 was removed. Other mutants carrying the point mutations were also prepared: the deletion mutant of the codon for F33 (deletion F33), the substitution mutant, in which the codon for phenylalanine at position 33 was replaced by the codon for alanine (F33A) and substitution mutant, where the codon for lysine at position 118 was replaced by the codon for alanine (K118A). Unfortunately, all three point mutants could not be cloned into the vector pGR106, therefore, the evaluation of their spread in N. benthamiana plants was not possible. Based on obtained results it is possible to conclude that the amino acid F33 is important for the intercellular movement within the neighboring cells. To assess the importance of the amino acids F33 and K118 in the systemic infection, it would be necessary to evaluate also the point mutants deletion F33, F33A, and K118.

Changes of enzyme activities in fruits during long-term storage

This study is focused on study of changes of enzyme and low-molecular weight antioxidants in different fruits during long-term storage. In theoretical part individual low-molecular weight antioxidants and enzymes are described. The main causes of fruit decay and some possibilities of fruit preservation and storage are summarized. As biological material some common fruits were chosen - green and red apples, peaches, plums and white grapes. The fruits were stored in laboratory, cellar, in refrigerator and in freezer. In freezing experiments some ways of fruit preparation and processing were tested and their influence on fruit antioxidant status was compared. Shortened storage experiment was applied on blueberries, cranberries, raspberries and strawberries too. In fruits some group parameters – total antioxidant status, dry mass content, ascorbate level, total flavonoids and total phenolics were analyzed spectrophotometrically. Individual flavonoids and phenolics were determined by RP-HPLC/UV-VIS and on-line LC/PDA/ESI-MS. Antioxidant enzyme activities (superoxide dismutase SOD, catalase CAT, polyphenol oxidase PPO and lipoxygenase LOX) were measured by spectrophotometry. The surface microscopy and cultivation of moulds from fruit surface were performed too. Influence of storage conditions on biological activities is dependent on fruit sort. Freezing is the most suitable procedure for long-term storage without significant changes of active substance content. Long-term storage in controlled temperature conditions and/or atmosphere is usable for fruits with longer storage period. In these fruits stabile levels of antioxidant enzymes are stored for relatively long time. Some of enzymes act synergistically. Enzyme activities differed according to storage phase; at the beginning mainly high SOD and LOX activities were observed. CAT and PPO are probably activated as defence systems in rippened and/or damaged fruits. Levels of total as well as individual low molecular weight antioxidants varied during storage in all sorts, generally, increased course with longer storage period can be observed.

Czech Centres

This Bachelor thesis focuses on the Czech Centres (Česká centra), a semi-budgetary organisation funded by the Ministry of Foreign Affairs of the Czech Republic. The main aim of this organisation is to promote Czech culture abroad, but its work is based on four pillars: culture, education, trade and tourism. Although other transnational political organisations also focus on three of these pillars, the promotion of Czech culture is a prerogative of Czech Centres. This thesis contains a comprehensive description of the functioning of the Czech Centres both from legal and economic point of view, its integration to the state administration, territorial allocation, history etc. The second part of this thesis compares the Czech Centres with two other foreign cultural centres: the Goethe Institute and the Polish Institute.  A separate chapter is devoted to dramaturgy of the Czech Centres and to a trip of one of the theatres from Prague (Dejvické divadlo) to Buenos Aires.

Residential real estate market during the financial crisis : empirical evidence from the CEE region

1 Abstract This thesis investigates the housing price determinants and possibilities of housing price bubbles in the residential real estate markets of Central and Eastern Europe before and during the economic crisis of 2007-2009. Using data from international institutions, national central banks and national statistical offices three quantitative methods are applied. Price-to-income ratios suggest housing price bubbles that were eliminated during the crisis in three out of five countries covered. Second approach of simple panel data models sheds additional light on housing price bubbles and indicates GDP growth, unemployment and average real wage as the main determinants of housing prices in the region. First indication of severe housing price persistence in CEE is demonstrated by the results of the models as well. More reliable results for housing price determinants are obtained from variance decomposition and impulse response functions of vector autoregression models. Each country is modeled separately and substantial differences exist between the countries. Poland is the only country that does not exhibit housing price persistence and dynamics in Austria are less volatile as compared to the new EU members in the sample. Czechoslovak CIAM Chapter after the Second World War

Swiftly describing Czechoslovak participation in the thirties the stress of this dissertation on the history of the Czechoslovak CIAM chapter after the second war. It describes the success of the Czechoslovak presentation on the congress in Bridgwater (national and regional plannig), the cooperation on the development of the CIAM Grid with Le Corbusier, the missed opportunity to hold the congress in Prague, the impact of centralization on the Czechoslovak CIAM group, the influence of socialist realism on East European countries, the unabled activities on the following three congresses and finaly the renewed participation at the last congress in Dubrovnik. While doing so it concentrates itself on four themes: regional planning, synthesis of the arts, historical centres and habitat which were all discussed at post-war CIAM.
