Long-term Analysis of Ultrasound Video Sequences Using Interest Point Detectors

This doctoral thesis deals with the analysis of ultrasound (US) video sequences. It specifically focuses on long-term tracking of the common carotid artery (CCA) in transversal section and measurement of its geometric parameters in a sequence of US images. The design and implementation of a system for automatic tracking of the artery is described in this thesis. The proposed system utilizes Viola-Jones detector and Hough transform to localize the artery in the image. Interest points are detected in the area of the artery wall. These points are then tracked using optical flow. The proposed system comprises a number of innovative methods which allow to perform accurate long-term measurement of parameters of CCA and store the results. A novel mathematical model describing the movement of CCA in transversal section during a cardiac cycle is defined afterwards taking the influence of breathing into consideration. A number of artificial sequences of US images based on this model have been created. These sequences were consequently used to evaluate the accuracy of the proposed system in terms of measuring the parameters of CCA. The sequences are unique because of their length which makes them suitable for evaluation of tracking accuracy even in long video sequences.

Detection of Signal Electrons in High Pressure Conditions in Environmental Scanning Electron Microscopy

The thesis deals with the study of properties of a new system for detection of true secondary and backscattered electrons in high pressure conditions of the specimen chamber of a newly built environmental scanning electron microscope AQUASEM II. Detection system contains three detectors. For the first time is introduced and analyzed the working principle of ionisation detector with electrostatic separator, which is in many experiments compared with ionisation detector of secondary electrons. Experimentally demonstrated are unique properties of this detection system, especially the ability of energy separation of detected signal electrons. For the various working conditions are also analyzed signal levels detected by the BSE YAG detector, which is designed as a part of the new detection system and which worked together with both ionisation detectors.

Survey Report: Detection of the extent of timber damage and traceological analysis of craftsman working Čistá u Litomyšle no. 97 - barn

The surveys, whose combined output is this survey report, covered the detection the extent of timber damage and traceological analysis of the original craftsman’s woodworking of selected elements of a dismantled polygonal barn, formerly belonging to house no. 97 in Čistá u Litomyšle. The impulse for the report was an order by the Wallachian Open Air Museum with headquarters in Rožnov pod Radhoštěm, which included the request for the diagnosis of selected timber elements using NDT devices and their traceological analysis. The knowledge gained can contribute to a qualified assessment and a proposal of gentle transfer of this unique log building.

This thesis focuses on the detective genre and creating of tension in three Petr Schulhoff?s detective films. It tries to describe rules of a good detective story construction and directorial and screenwriting techniques for tension creating in films. Petr Schulhoff?s films are exceptional by their visual authenticity, compliance with the rules of detective genre and especially by the tension which creates a unique atmosphere.

Processing of iris images for biometric applications

Biometrics is a method of recognizing the identity of a person based on unique biological characteristics that are unique to each person. The methods of biometric identification is currently becoming increasingly widespread in various sectors. This work is focused on the identification of a person by iris images. The introductory section describes the principles of the well-known methods for biometric applications and the next part describes the design method and its implementation in Matlab. In the practical part, fast radial symmetry method is used for detection of pupil, from which it derives further image processing. Two dimensional discrete welvet transform is used here. The proposed algorithm is tested on databases CASIA-Iris- Interval and database IITD.

Extraction of arteries and veins from fundus image of human retina.

This thesis deals with processing of retinal fundus images. Vision is the most important human sense and its injury has very serious consequences for humans. Automatic processing of retinal images increases the efficiency of medical examination and accelerates diagnoses of deseases. Retina exhibits unique characteristics for each person and thus can also be used to identify people. In this task is briefly discussed the structure and properties of each parts of the eye, particularly the retina, and their possible diseases such as diabetic retinopathy, glaucoma and age related macular degeneration. Subsequently, the task describes the representation and characteristics of the digital image. Also is devoted to selected image segmentation methods namely thresholding, edge detection and segmentation techniques based on the matched filter. The outcome of this task is the application in which several segmentation methods are implemented for the blood vessels extraction. For each of these methods it is possible to set the parameters of the segmentation to ensure high quality blood vessels extraction in images of different quality.

Novel protease in the reduced mitochondria of Giardia intestinalis.

The proteom of Giardia mitosomes is represented by twenty proteins, which are imported from the cytoplasm, as mitochondrial genome is missing. The N-terminal sequences, which target proteins to mitosomes are removed upon translocation and the remaining protein is folded into its native comformation. Out of three types of mitochondrial processing peptidases only functional half of dominant matrix peptidase is present in the mitosomes of G. intestinalis - feature unique among all eukaryotes. Based on the proteomic analyses, we identified novel putative mitochondrial peptidase (peptidase X) in G. intestinalis mitosomes. Overexpressing the protein with hemagglutinin tag we confirmed its mitosomal localization. Using specific polyclonal antibody combined with carbonate extraction and protease protection assay we have specified the distribution of the protein in the mitosomal subcompartments. Based on these results, peptidase X is anchored in the inner mitosomal mambrane via its C-terminus facing the mitosomal matrix. Peptidase X has mitochondrial targeting sekvence on the N-terminus, which is also capable of targeting protein into yeast mitochondria. The detective genre originated in the first half of the 19th century and it does not seem to be loosing its pace. On the contrary, new authors have been occurring, bringing new rules and attitudes to the classical formo Detective novel is usually c10sely connected to the settings it describes. The aim ofthis thesis is to find out specific features ofthe Sicilian detective novel. We have chosen two authors and their works for a detailed analysis: Leonardo Sciascia and his novels II giorno della civefta and A ciascuno il suo and Andrea Cami11eri and his La forma dell' acqua and II cane di terracofta. The thesis has two parts. Part one has three chapters: in first chapter we deal with the history of detective novel, the European detective style and the Anglo - Saxon style, or the classical and American detective stories. ln chapter two, we focus on the isle of Sicily, its history and the unique character of its inhabitants and their culture. ln the final chapter, we pay attention to the works of the most famous Sicilian writer Luigi Pirandello e. g.Ilfu Maftia Pascal or Cosi e (se vi pare) , his search for reality, human identity and the relativity ofthe truth, all ofwhich occur in Sicilian detective stories.

The marketing analysis of guest loyalty program implementation at Grand Hotel Zvon

The diploma thesis is dealing with a topic of hospitality, a natural part of the services sector. Under the conditions of a constant development and ever-increasing competition there is necessary for hospitality industry enterprises to impress upon their customers their unique features and quality. Customers always seek a higher quality of services provided, individualization of services, logically they always look for the highest return of their investments. One of partial aims of this diploma thesis was an analysis of preferences of guests in a context of concept and implementation of a loyalty program. Other partial aim was to analyze indoor and outdoor attributes of the hotel. Marketing and management methods of SWOT analysis, questionnaire etc. have been used to achieve these aims. The main aim of this diploma thesis was to propose an optimal structure of the loyalty program for guests. The program must respect guests{\crq} wishes and needs while remaining financially acceptable for the hotel. A questionnaire has been used to detect the preferences of guests which then formed the basis of proposed loyalty program. Setting the conditions and principles of loyalty program membership was the next step to achieve the main aim. Finally, a detailed calculation of every item and benefit used in the loyalty program was created.

History of vegetational development in the Cheb basin from sedimentary record of the SOOS National Nature Reserve

Vegetation development in the Cheb basin ( W Czech Republic) recorded in the sediments of the SOOS ational ature Reserve This thesis focuses on palaeoecological research of quaternary sediments in the Soos National Nature Reserve (Cheb basin, NW Czech Republic). Quaternary history of this locality is poorly known, despite it is a valuable and protected area with high natural qualities. The Soos basin is also known for its unique geological record, which is expected to cover information on landscape dynamics at least during the Holocene. A lake existed in the Soos basin enabling accumulation of diatomite and peat sediments of a broad palaeoecological potential. Records were analysed with method of pollen analysis and were dated using 14C. The results of this study bring information about vegetation development in the area of Cheb basin as well as development of lake ecosystem. It also gives a new insight on lake's origin and its age. The accumulation of peat sediments started in the Preboreal period and was continuous at least until the Atlantic following with a hiatus, therefore further detection of vegetation development is impossible.
