The preparation of recombinant tau protein and it's usage for the detection of Alzheimer disease

Currently a grate emphasis is being put on accurate and early diagnosis of Alzheimer's disease (AD), which is important for the introduction of treatments that could postpone the onset of the disease. Antibodies against tau protein appear to be suitable biomarkers for early diagnosis of AD. Therefore, this work deals firstly with preparation of human recombinant tau protein in bacteria and its subsequent use in determining levels of antibodies in blood serum of patients with AD and in normal older persons. A preparation of the tau protein in sufficient purity was achieved for the antibodies measurment by ELISA method and results were statistically analyzed using nonparametric methods. Results were compared with data from Mgr. Jany Švarcové obtained by measuring antibodies in serum of patients with AD and normal elderly using an commercial bovine tau protein. According to the analysis there are differences between the data obtained from human and the bovine tau protein. It was proven that patients with AN have lower levels of antibodies against tau protein than healthy seniors. Recombinant human tau protein was also used to immunize rabbits. The ELISA method confirmed the creation of antibodies against human tau protein in rabbits.

Laboratory markers of sepsis

Sepsis and its complications are currently the most frequent cause of death owing to multi organ failure at intensive care units. The aim of this thesis was to evaluate the most suitable usable marker with sufficient diagnostic sensitivity and specificity for early catch of sepsis development. For my thesis a chosen data collection patients of intensive care units and patients of neonatology department in Nemocnice a.s. (Hospital Inc.) in České Budějovice was used. In patients, determinations of following concentration parameters were carried out: PCT, CRP, IL-6. For diagnosis and evaluation of clinical state change in relation to sepsis PCT is convenient. For monitoring of steady course and clinical state it is useful and sufficient to watch dynamics of CRP. In newborns the sensitivity PCT decreases in sepsis probably due to immaturity of liver and immunity system. That is why according to experience gained in Nemocnice a.s. (Hospital Inc.) České Budějovice it is more convenient to determine IL-6 instead of PCT.

Range and Variety of Daily Motor Fitness Game Activities of Early School Childhood

Cílem mé diplomové práce bylo zmapovat využívání různých pohybových a sportovních her u dětí mladšího školního věku ve vztahu k jejich dennímu režimu. Dále zjistit rozsah a obsah těchto herních pohybových aktivit na základních školách u dětí mladšího školního věku. Zjistit, kolik času věnují děti pohybovým aktivitám a podle jakých kritérií si děti volí činnosti v jejich volném čase. Dalšími cíli bylo zmapovat rozsah a obsah pohybových her u vyučujících. Zjistit jaké sportovní a pohybové hry hrají učitelé s dětmi nejraději a nejčastěji, a jaké prostorové možnosti ke hraní sportovních a pohybových her učitelům škola a okolí školy může nabídnout. Studie se dále zabývala porovnáním výsledků mezi školami na vesnicích a ve městě na okrese Znojmo. Toto srovnání jsem provedla na základě vyhodnocení údajů získaných dotazníkovou metodou a z informací obsažených v odpovědích dětí a rodičů v rámci realizovaných řízených hovorů. Na základě splnění úkolů diplomové práce mohu konstatovat, že jsem stanovený cíl diplomové práce splnila.

Fortification architecture in 10th and 11th century in Central Europe. Early medieval fortification of Žatec.

The master's thesis intends to depict the situation of fortification constructions in Central Europe in 10th and 11th centuries. It emphasizes relations of the Czech area and west part of Central Europe. The first part analyzes four new excavations of the Žatec fortification, dist. Louny, especially the excavations in the line of the bailey fortification. The line of young hill fort period fortification of the acropole serves as a basis for further study of the topics in question. The valuable site illustrates many phenomena which are described in the theoretical part. It surveys origins of fortification technologies, their development in the studied period and determines the position of the Žatec fortification in the corresponding context. The study for lack of ceramics focuses mainly on the analysis of stratigraphic situations and compares all available stratigraphic features which come form the excavation of the garden of the Municipal Theatre. The thesis tests one of ways how to process field data from fortification excavations and studies appropriate possibilities of further employment of the presented methods in analogous situations. Brandýs nad Labem-Vrábí (central Bohemia), a late Hallstatt - early La Tene settlement

My bachelor work is concerned with the archeological finds processing from Late Hallstatt to Early La Tne age. The finds have been discovered during rescue archeological research in poly- cultural residential area in Vrábí- Brandýs nad Labem. The aim of my work is to make systematical and synoptic processing of archeological materials, subsequently explicate the finds and place the residential area in the context of time and place. The emphasis is placed to utilize the findigs with maximum efficiency and to make it possible to compare archeological data from other localities.

Importance of the exercise of parents and children for the development locomotive skills and social competentences in toddler and early preschool age

The diploma thesis deals with the exercise of parents and children (PACES - parents and children exercise simultaneously) and its effect on toddlers and children of early pre-school age. It examines the development of locomotive skills and social competences of this age category from both theoretical and practical point of view. The introduction of the thesis focuses on the history of such exercise, or on psycho- social and physiological development of children. On this basis it formulates some rules for this type of exercise. The practical part verifies the benefits of the exercise by testing, measuring locomotive skills and comparing them with those of children that do not participate in the PACES. It maps the formation of children's social competences via socialisation games. The opinions of participating parents represent another view on the exercise of children and parents. The conclusion of the thesis puts the results of the survey into a wider context of research and measurement made on this topic in the past. 2

Jakub František Prokyš The bachelor work Jakub František Prokyš is intent on a successful painter who lived in the 18th century. He originally came from Slaný but his life fate took him into culturally developing South Bohemia. It was a great chance for him. Strongly flourishing Schwarzenberg's family were establishing their position in Bohemia not only by buying new areas but as well as by improving their former and newly gained dominion. That is why the Schwarzenbergs employed a lot of important architects, painters and sculptors. Jakub František Prokyš was among them. Everything has not been cleared up about Prokyš´ destiny up today. There are a lot of questions which answers we are still looking for. Basically, until to his coming to South Bohemia we do not have real data about his life and apprenticeship. Even in his South Bohemian life many questions are hidden. It is the most remarkable to follow how respectable person could become from a quite unknown artist. He got among notable people. He earned enough money to be able to buy his own house and take care of his family during his life. However, professional success was changed by family tragedies. He was a great fresco painter as well as he could paint even altar paintings. It is strange that his very early works are considered to be his best ones. Those were ordered by Joseph Adam Schwarzenberg. The most space is devoted to those works of art in my bachelor work. The aim of the work is my attempt at closer outlline of Prokyš´not well recognized life.

Feed-forward neural networks and their application in data mining

The goal of data mining is to solve various problems dealing with knowledge extraction from huge amounts of real-world data, the quality of which might be disputable. Neural networks can help with the solution due to their generalization capabilities. While working on data mining projects, we have essentially the following two objectives in real-world applications of feed-forward neural networks. To obtain applicable results, it is crucial to provide the networks with well-prepared data. However, it is equally important to choose the right training strategy for the networks themselves - including network architecture, parameter settings or the training algorithm. One of the most important ideas behind these steps is namely to prevent "over-training". The final network should recall unknown examples as well as possible. There are plenty of techniques with different approaches to the solution. It is possible to modify the data, these comprises modifying the range of the data or its dimension, adding noise to the data, etc. Yet another way is the modification of the neural network by structural learning with forgetting, weight decay or early stopping. Statistical investigation of CT pulmonary angiography in terms of development and depending on age and sex in District Kroměříž 2006-2010

CT pulmonary angiography is, together with ventilation-perfusion scintigraphy, the main diagnostic methods for recognition as pulmonary embolism. This is a very dangerous disease, which may end up even in death. Its share of deaths in hospitals, including subsequent ? complications, is around 4%. Therefore, emphasis is placed on early diagnosis. Clinical and laboratory symptoms are unspecific (including D Dimer).(10) Pulmonary embolism after ischaemic heart disease and arterial hypertension the frequency of its occurrence, the third most serious cardiovascular diseases. Cause of death in pulmonary embolism is usually the exhaustion of the right heart, seeking to overcome suddenly increased the resistance of the pulmonary circulation. This is the pulmonary artery or its branches is embolism blood clot-trombus, resulting in the venous system or in the heart, a saddle embolus, patched up a significant part of the lung. The most common place of trombs, which embolizated to the lungs, are deep veins of lower limbs if they are affected by the flebotrombolitis.(16) In addition to the acute embolism are frequent and chronic embolism. They are equally dangerous. By gradually and subtly clog small blood vessels, reduces the total cross-sectional area of open pulmonary artery, which have blood flow, and since the volume of blood must be maintained even in the smaller section of the vascular bed, should permanently increase the blood pressure in a small circulation, which is also dangerous.(16) In my work is the methods of mathematical statistics processed file 1966 CT pulmonary angiogramy (CTPA), carried out on the CT work in Kroměříž in the years 2006-2010. The work is focused on the statistical survey of the influence of age, sex, and the time evolution of the CTPA during that period.

The Importance of Timely Diagnosis of Dyscalculia

The bachelor work is focused on diagnosing dyscalculia. Its aim is to highlight the consequences of the late diagnosis of dyscalculia for a child and its relatives. The work includes information about dyscalculia, its diagnosis and teaching methods. The work contains five cases of late diagnosing of dyscalculia, case studies, an evaluation of the consequences of the late diagnosis for the child and its relatives. There is also a comparison of the differences in the development of the child with early and late diagnosed dyscalculia and outlined remedies. The work includes the research about the awareness of dyscalculia and an information availability about it. There is a research of a frequency of the term dyslexia vs. dyscalculia in contemporary mass media too. The aim is to notice the lack of awareness of dyscalculia among teachers and non-teaching public, a summary outlining the implications and potential sources of improvement.
