Novel protease in the reduced mitochondria of Giardia intestinalis.

The proteom of Giardia mitosomes is represented by twenty proteins, which are imported from the cytoplasm, as mitochondrial genome is missing. The N-terminal sequences, which target proteins to mitosomes are removed upon translocation and the remaining protein is folded into its native comformation. Out of three types of mitochondrial processing peptidases only functional half of dominant matrix peptidase is present in the mitosomes of G. intestinalis - feature unique among all eukaryotes. Based on the proteomic analyses, we identified novel putative mitochondrial peptidase (peptidase X) in G. intestinalis mitosomes. Overexpressing the protein with hemagglutinin tag we confirmed its mitosomal localization. Using specific polyclonal antibody combined with carbonate extraction and protease protection assay we have specified the distribution of the protein in the mitosomal subcompartments. Based on these results, peptidase X is anchored in the inner mitosomal mambrane via its C-terminus facing the mitosomal matrix. Peptidase X has mitochondrial targeting sekvence on the N-terminus, which is also capable of targeting protein into yeast mitochondria. The aim of the dissertation thesis is the development of modern extraction methods for the sample purification and for the analytes isolation and preconcentration from various biological materials, including biological fluids (plasma, serum, vaginal lavages) and plant material. Modern approaches have a lots of advantages such as a lower consumption of sample and organic solvents, extraction speed-up, better sample purification, matrix removal and lower influence of matrix effects. Extracts from modern microextraction methods can be analyzed by separation methods including ultra-high performance liquid chromatography and supercritical flluid chromatography coupled with mass spectrometry. The separation methods enable fast high-throughput analysis with sensitive and selective detection. Creativity and innovation in self-awareness and self-management

This bachelor's work addresses the theme of self-awareness and self-management, their creative methods and implementation. The aim of this work is to represent and express the importance of knowing one's self and to show the important role of self-management in the every day life of people. Different practical tools and methods which can contribute to higher performance and success are described as well. The first part of this work contains theoretical knowledge about creativity and innovation, self-awareness and self-management and the assessment of effective goals; the emphasis is dedicated to one particular tool and method which could contribute to the improvement of people's performance and perfection. The practical part of this bachelor's work continues with working out the theoretical part and is founded on research which aims to confirm whether students in universities are concerned about self-awareness and self-management; and in the cases where they are, to which degree are they using these particular tools and methods. Following the results, an interpretation of the research is formulated in advantageous suggestions and recommendations.

Methods of reduction of excessive biomass of phytoplankton

The aim of this study was to prepare review of methods to combat over development of phytoplankton. The work also includes a critical evaluation of usability, efficacy and risks of these methods. The methods to combat over-development of phytoplankton can be divided into direct and indirect. While the direct methods acts directly against phytoplankton, indirect methods are based mostly on reducing the amount of nutrients available for phytoplankton growth. Both of these methods can be further divided to biological, physico-mechanical and chemical. The direct methods of physico-mechanical include ultrasound, flushing and dilution, direct chemical methods include the use of chemical algaecides, coagulants and flocculants. Direct biological methods are especially divided by the principle of parasites(viruses,bacteria and fungi), competitors (algae), allelopathy (extracts from higher plants),predation (protozoa and herbivorous fish). The indirect physico-mechanical methods include extraction and overlapping sediment. Chemical methods are meant by the binding of nutrients to insoluble compound in the sediment and the biological methods are meant use of communities of bacteria for mineralization of sediments. The next part of the work is devoted to the characteristics of algae and cyanobacteria, as well as factors affecting the formation and development of algal blooms. In this part of the work I focus mainly on cyanobacteria, their toxins and the effect they have on the individual components of aquatic ecosystems.

Optimization of conditions for derivatization of fatty acids and perfluorinated organic acids with methylchloroformate

Statistical methods designed for optimization were applied for selection of optimal experimental conditions of derivatization process for perfluorinated acids, PFA (C8, C10 ,C12), fatty acids, FA (C8, C12, C14) and their mixtures. The calibration curves and the extraction efficiences for system acetonitrile/heptane were measured for all studied acids. The Plackett-Burman screening was performed for obtaining the significant experimental parameters from their original series and significant ones were remitted to optimization via central composite design, which takes a second order effects and mutual interactions in between parameters into account. Optimal conditions of derivatization were found for PFA, FA and their mixtures. The principle of the search of optimal selective derivatization conditions for one group of acids in the presence of the other is based on the search of optimum on the response surface where the dependent variable is either sum or difference of acid group responses normalized to common internal standard. It was found that FA influence total response more than PFA and that in the case of demand for selective derivatization of PFA in the presence of FA in the sample, the most significant parameter is the volume of methylchloroformate added. Stanovení obsahu residuí kofeinu v povrchových vodách metodou HPLC s UV detekcí

The occurence of caffeine residues was investigated in superrficial waters. The aim of this study was to obtain a first overview about contamination of waters from Mondego River and from source fontains near Coimbra city ( Portugal) with caffeine. The samples were preconcentrated by SPE ( Solid Phase Extraction) with Oasis polymeric column procedure and analysed by Liquid Chromatography (LC) with UV detektor ( 280nm). Mobile phase consist of 190 ml acetic acid glacial 100% and 810 ml of LC water (adjusted to pH 3.0 with acetate sodium) A total of twelve samples were collected in different site of Mondego River and from fountains near Coimbra city during three month's periods. A total of twelve samples of caffeine were analysed under the conditions described and none of them showed to be contaminated with caffeine.

Side products from power industry as the raw material base for the future

This work reacts to the worldwide problem related to looking for alternative sources of raw materials. The work deals with analyzing samples of high temperature and fluid fly ashes produced by power plants in the Czech Republic, followed by experimental verification of possibilities to separate some desired components. Based on the elemental and phase compositions, the possibility of aluminum, titanium, and iron extraction to sulfuric acid solution and subsequent selective precipitation using suitable separeation methods was designed and tested in a laboratory scale. Separation of selected elements from the fly ash matrix was carried out at room temperature and then at the boiling point of the used solvent.

Modular Document Supply System

Článek projednává o modulárním Document Delivery systému vyvíjeném v STK. Vývoj byl podpořen projekty LI200028 (VPK) a LI01018 (portál STM). Tento systém staví na zkušenostech získaných vývojem a provozem Virtuální polytechnické knihovny. Je implementován v jazyce Java, z důvodů snadné přenositelnosti. Systém neobsahuje katalog dokumentů, požadovaný dokument musí být vyhledán mimo tento systém a je možné předat metadata o vyhledaném dokumentu a jeho umístění v OpenURL adrese při zadání objednávky. Účastnící sytému musí být registrování a mají v systému svůj finanční účet. Objednávku na digitalizaci dokumentu a zprostředkování výtisku kopie si zadává účastník systému samoobslužně web browserem. Objednávka je uložena v databázi čekajících objednávek, a periodicky dotazována digitalizačními místy. Pro toto je digitalizační pracoviště vybaveno zvláštním komunikačním a digitalizačním programem. Digitalizační pracoviště dokument zdigitalizuje a uloží do souboru .pdf a předá zpět do databáze. Tento soubor .pdf je určený pouze k tisku a je chráněn proti změně. Uživatel si ho stáhne ze své www stránky chráněné heslem a vytiskne na své lokální tiskárně. Současně s těmito procesy probíhá účtování ceny zakázky, která je odečtena z konta uživatele a přičtena na konto knihovny, která pořídila kopii dokumentu. Systém se nazývá modulární, protože umožňuje komunikaci s jiným instalacemi téhož systému a tak předávat objednávky do jiných knihoven vybavených tímto systémem (obdoba požadavku MVS). Objednávky a data se předávají pomocí standardizovaných XML souborů. Cílem je dosáhnout kompatibility i mezi heterogenními systémy DDS.

Analýza a návrh webového rozhraní pro uživatele služeb typu Document delivery jako součást Analýzy implementace DDS do AKS NPKK

Práce popisuje vytvoření návrhu nového webovského rozhraní e-PK pro nově vytvářený modul ?document delivery? automatizovaného knihovního systému KP-WIN SQL. Návrh byl vypracován na základě analýzy stávajícího rozhraní e-PK a porovnání e-PK s rozhraním Virtuální polytechnické knihovny. Dále byly zakomponovány návrhy, potřeby a přání servisního centra e-PK za využití dynamicky generovaných stránek a přístupu k databázi.

Poskytovatelé informací jsou rozděleny na: instituce Evropské unie, státní orgány České republiky, specializované instituce zřízené státem, tiskové agentury, profesní sdružení, producenti specializovaných databází, databázová centra, specializované nakladatelství a knihovny
