Folklore in the Region of Jihlava - Vysočan: set of songs and dances of region Horacko

This thesis is to introduce the folklore in the region Horácko in a hitherto unknown way. Based on the archive files, the aim was to describe the formidable establishment of the folklore group 'Vysočan', its development and promotion in the field of folk dances and songs in the Czech Republic and abroad as well. I tried to explore information that is not available in common tourist brochures about folklore in the city of Jihlava and its surroundings. With this fact are connected the parts of this thesis describing various activities of the folklore group. For instance, 'Horácký bál', which I have introduced in this work and which also provides an extraordinary backstage insight, has been never described as well as 'Jihlavské folklorní léto'. Nevertheless, there would not have been any folklore, if there had not been typical folk costumes. There are several publications concerning this topic, but they are hard to find and what is more, sometimes we can find some misunderstanding information in them. Part of this thesis describing the folk costumes should bring records corresponding to reality and highlight some interesting notes taken from various scientific publications. Analysis of personnel work in the company

The aim of this thesis is to analyze the personnel work in a concrete company -- the comparison of practical application with available theoretical knowledge and the subsequent evaluation related to the proposal of relevant measures that would improve effectiveness of personnel work in the monitored company. The thesis is divided into two parts -- theoretical and practical. The theoretical part contains information about the importance of human resources management, its relation to the overall strategy of the organization and brief characteristics of particular personnel activities. The practical part describes the characteristics of the company, its organizational structure, basic personal data and I continue with analysis of personnel work and personnel activities in the company. At the beginning of each chapter is a procedural scheme under which the analysis progresses. At the end of each chapter I summarize the findings, evaluate the level of personnel work in the company and introduce proposals for optimization of human resources.

Effects Evaluation of Quinazolin Alkaloids from Justicia Adhatoda

Title of rigorous thesis: Effects evaluation of quinazolin alkaloids from Justicia adhatoda The aim of our experiment was the evaluation of a relaxing activity of natural substances obtained from Justicia adhatoda by an in vitro method on an isolated trachea of rats. Specifically, we tested: a standardized ethanol extract from Justicia adhatoda, vasicine and vasicinone. As a standard substance, we used theofylline. Theofylline was also used for inducing maximal relaxation of a trachea. Our experiment was carried out on rat males of Wistar tribe, weight between 250-300 g. The scheme of our experiment was in following sequence: inducing trachea contractions by giving a carbachol solution (10-5 M), a cumulative intake of separate doses of a tested substance ensued; finally, theofylline (3×10-3 M) was used for inducing maximal trachea relaxation. Every measured value of relaxation of tested substances was compared to maximal relaxation by theofylline and issued with percentage. Acquired values were compiled by GraphPad Prism 5 program. Offer analysis of the travel agency Melissa Travel

Bachelor thesis pursues the offer analysis of the travel agency Melissa Travel. The opening part is based on theoretical conception, where all the basic terms are defined. There is specified the market area, where the power belongs to the travel agency. The advantages coming out of free market are highlighted. There are determined the elementary legislative conditions, that necessarily must be followed by travel agency. The term "offer" is defined and clarified both on the general and detailed level. Practical part is based on product analysis. The product is meant by services, that are prepared by the travel agency Melissa Travel and offered to it's clients. There are investigated basic services and it's complements as well as product price, that is seen as a specific part of the offer. Outer analysis is exploring the ways of distribution and propagation used by travel agency Melissa Travel. On the basis of accomplished analysis the profil of travel agency Melissa Travel is defined and the target type (the client, that travel agency orientates it's activity on) of it's customer is specified.

SVN Proxy

SVN Proxy is an application that behaves like a proxy of a SVN repository. It contains a local repository, which is used by a user for local commits. The local repository enables to synchronize itself with a remote SVN repository on demand. Local revisions can be numbered in a way they never become unsynchronized with the remote repository. The data of the local repository is stored in XML. The application contains the implementation of a diff algorithm and enables to merge versions and to detect conflicts. SVN Proxy is a command line client and its interface is derived from the common SVN client interface. The main emphasis is laid on the easy future development in the design of the application. The design enables to add other new connections to the repository and create other user interfaces including GUI. The application is intended for source text developers who do not want or are not able to send every commit into the remote repository.

Argo, fetch! The history of dog training (cynology) in Liberec region after 1945

The main topic of the thesis is a description of the transformation of the Sport Cynology during the second half of the 20th century in the Liberec region. The sport is based on cooperation between people and dogs. This hobby belongs to the most popular freetime activities in the Czech Republic, even though there is not enough information in Czechoslovak historiography. The aim of the thesis consists in a characterization of development of the Sport Cynology in the era of State Socialism in Czechoslovakia in the second half of the 20th century. I focus on a historical progress of the sport and its significant events, which were crucial for the next development of the sport activity. In the context of the Sport Cynology, "The Association for Cooperation with the Army" is mentioned because Czechoslovak cynology had to become a part of the association in 1953. In the first section of the paper, focused on history of the cynology, I use archival materials, contemporary magazines and information got from interviews with narrators. I apply qualitative research and its specific methods such as analysis of documents and thematically oriented biographical narratives. Práce srovnává styly řízení a vedení manažerů a manažerek v českých firmách. Srovnání bylo provedeno na základě kvalitativního výzkumu. K získání informací pro výzkum bylo použito dotazníkové šetření, kterého se zúčastnili manažeři a manažerky a také jejich podřízení. V práci jsou zkoumány charakterové vlastnosti, měkké dovednosti, odborné znalosti, styly řízení, firemní kultura a celková úspěšnost manažerů a manažerek a analyzovány rozdílné a shodné prvky v jejich manažerské práci.

Haplotyping of oak populations by chloroplast sequencing reveals origin and homogeneity of populations

The aim of this publication is to present new method of DNA isolation from young\noak leaves that enables efficient amplification of trnD-trnT region of chloroplast\nDNA and direct sequencing of crude PCR reaction mixture. Sequence data of trnDtrnT region have been used to examine Quercus robur and Q. petraea populations variability. High discrimination power of this method is demonstrated, comparable with previously used restriction analysis of four similar chloroplast DNA regions. Variable positions in trnT-trnD chloroplast DNA fragment are mapped and their use for oak haplotyping indicated. As an example, 20 Czech Republic oak populations were analyzed and their history, geografical origin and homo/heterogeneity were revealed.

Establishing Mutual Links among Brain Structures

The Human brain consists of mutually connected neuronal populations that build anatomically and functionally separated structures. To understand human brain activity and connectivity, it is crucial to describe how these structures are connected and how information is spread. Commonly used methods often work with data from scalp EEG, with a limited number of contacts, and are incapable of observing dynamic changes during cognitive processes or different behavioural states. In addition, connectivity studies almost never analyse pathological parts of the brain, which can have a crucial impact on pathology research and treatment. The aim of this work is connectivity analysis and its evolution in time during cognitive tasks using data from intracranial EEG. Physiological processes in cognitive stimulation and the local connectivity of pathology in the epileptic brain during wake and sleep were analysed. The results provide new insight into human brain physiology research. This was achieved by an innovative approach which combines connectivity methods with EEG spectral power calculation. The second part of this work focuses on seizure onset zone (SOZ) connectivity in the epileptic brain. The results describe the functional isolation of the SOZ from the surrounding tissue, which may contribute to clinical research and epilepsy treatment.

Characterisation of novel serpin TILIr and its relatives from the superfamily of serine protease inhibitors from \kur{Ixodes ricinus} tick

Novel gene, named TILIr ( TIL Ixodes ricinus), that encoded yet unknown protein with TIL domain characteristic to proteins that belong to trypsin inhibitors family was discovered and isolated from hard tick Ixodes ricinus. Two isoforms of novel gene were identified and studied in this project. As the result of this study recombinant TILIr protein was produced in bacterial expression system and analysis of its functional characteristics was conducted. The protein belonging to family of trypsin inhibitors represent a potential candidate for anti-tick vaccine component.
