The impact of modern communication tools on international supply chains
Hricko, Ján ; Černohlávková, Eva (advisor) ; Olšanová, Květa (referee)
The subject of my thesis is to clarify main changes in national and international trade that could have been caused by modern communication instruments. Some of these changes have already been used by few retail sellers of light products in Slovakia. Nevertheless such changes are nowadays more and more likely to be seen also among wholesale dealers. I focus on current conditions of international trade and its possible way of development in future in theoretical part of my work. I did not omit even some examples of modern technologies that have been successfully implemented in our daily lives or are likely to be used in a foreseeable future. As a next step I chose to analyze internet communication and its advantages for either seller or customer. I assume that, as far as internet communication is concerned, it is progressively becoming interactive and therefore there is a greater demand on implementing new changes as fast as possible. The next point of my work is to describe typical representatives of market with light products and to analyze their current functions in international trade and ways in which they use their modern communication tools. In conclusion if we did not take a few well-known and also some new sellers into consideration, it could be said that modern technologies mentioned above have not yet been used by "classic" sellers. These sellers are not capable of implementing such methods quickly enough and this could, in my opinion, result in a considerable problem in future.

Homelessness – Res Communes (Local Housing Policy Anticipation)
Dobrovičová, Iveta ; Kučerová, Eva (advisor) ; Kateřina, Kateřina (referee)
Summary The thesis is dedicated to solving the problem of homelessness, from the vantage point of synergy of social policy tools, mainly the housing policy. The author s aim has been to find out what methods are adopted to prevent and solve homelessness in Prague, whether the steps taken by the state or other subjects active in the capital provide help to the group of the socially excluded individuals in the process of their reintegration into the society of people with homes. The theoretical part deals with the crisis of the welfare state concept, based on solidarity of the wealthier classes with the socially deprived groups. After examining expert sources, the accent is put on definition of social policy participants on the part of the state and other subjects that are instrumental in integration of persons temporarily or permanently excluded from the consumption of the fundamental social value, housing. Last but not least, this work defines the phenomenon of homelessness in its key aspects: in the history of its emergence and causes, its perception by the public, typology of homeless persons, and in the problem with a qualified estimate of the number of the affected. The practical part is dedicated to a closer examination of the social exclusion of inhabitants in the interaction with the responsible persons from the ranks of social workers, curators, housing authority clerks, providing expert help to the socially deprived groups among Prague inhabitants. This research, done in the form of interviews, is completed by a questionnaire survey carried out directly with the people concerned the homeless. The contribution of this work consists in this combination of both perspectives, the basis of which is knowledge of the homeless environment, acquired via the author s voluntary work for a lowthreshold treatment program center Naděje in Prague.

Bachelor's thesis deals with organic food. The first chapters are definitions of terms that are closely associated with this topic. Chapters include concepts as organic farming, focusing especially on plant cultivation and animal husbandry because these two industries produce organic food. In the following chapters we find a description of manufacture, inspection, marking and prices of organic food. The important part is devoted to the franchise concept Sklizeno. The aim of this part is make a business strategy for the company and draw up proposal most advantageous locations in the Prague for the placement store Sklizeno. Bachelor's thesis used method of describing a quantitative method (used in the form of interactive and field survey). The aim of these methods is obtain sufficient information about areas of Prague and its population, which serves as a material for selection of a suitable location. Conclusion contains a business strategy for franchising project Sklizeno and proposal suitable area to locate their store.

Hollow cone valve capacity in the restricted outflow conditions
Nehudek, Adam ; Haluza,, Miloslav (referee) ; Šulc, Jan (advisor)
The thesis aims to determine the influence partial or complete submerge of hollow cone valve by outflow on its capacity without space restrictions, by outflow into discharge chamber and by outflow into water tunnel of circular cross section. The valve capacity is for mutual comparability purposes usually characterized by discharge coefficient. Research works have been performed on a spatial hydraulic model of the valve with inlet diameter D = 67 mm and an apex angle of a cone 90°. Values of the discharge coefficient obtained from measurements were statistically analyzed. The dependence of discharge coefficient, expressed by energy head, on submerged level was approximated by an exponential function. Exponent value of this function express the dependance rate of valve capacity on downstream water level. This procedure didn’t demonstrate signifiant dependance downstream water level on hollow cone valve capacity, only on tests at outflow to water tunel showed a low increase in the discharge coefficient partly due to the influence of kinetic energy of the stream flowing out of the tunnel on the energy head and also due to underpressure. Independence of discharge coefficient value on valve head has been prooved for head greather than 232 mm. Also was defined 3 stages of interaction outflow jet from valve and downstream water, some of them may cause on real waterworks structures unfavorable situations (may affect excessive load on valve by pressure pulsations). The presented results allow better evaluation of bottom outlets capacity at higher water levels, when the valves are flooded. Significant effect of sufficient aeration (especially in long water tunnels) on valve capacity was also confirmed. Taking into account on the specifics of hollow jet valves and cone valves, it is possible to generalize some of this resultos on them as well.

A teacher as a victim of violence
Tomášek, Jan ; Pelikán, Jiří (advisor) ; Černíková, Vratislava (referee) ; Koťa, Jaroslav (referee)
The thesis is focused on violence against teachers. It is a broadly discussed topic because of several cases of teachers assaulted by a student in the Czech Republic or abroad, sometimes with fatal consequences. Usually there is an opinion that youth violent behaviour is on increase and therefore educational profession is becoming a risk one. The thesis suggests there is some kind of distortion in sources of such information which such opinions are based on. It relates both to news coverage and crime statistics. There is also a problem to define violence or aggressive behaviour. Moreover, we should consider them in a close relation to the work of a teacher, ideally as a type of so called workplace violence. The thesis explores causes of violence against teachers. Not only with respect to perpetrator but also regarding the victim and his or her behaviour or specific features typical for interaction between teachers and students. Empirical research is the most important source of relevant information related to the extent and patterns of violence against teachers. Unfortunately, there are only few studies focused on the problem. This is the reason why the discussion of the problem is usually based on guesswork or speculations but not on the real knowledge. The author of the thesis tries to summarize outcomes...

In vitro-cultures of medicinal plants - V
Sedlářová, Marcela ; Tůmová, Lenka (advisor) ; Martin, Jan (referee)
The elicitation of the therapeutically important cultures in vitro is a metod used for increasing the production of secondary metabolites. The effectiveness of elicitation depends on a complex interaction between the elicitor and the plant cell. The elicitation is influenced by many factors: the elicitors specificity, their concentration, the treatment interval, the growth phase of cell culture, the composotion of nutritional medium and the light. A wide array of external stimuli are capable of triggering changes in the plant cell which leads to a cascade of reactions, ultimately resulting in the formation and accumulation of secondary metabolites which helps the plant to overcome the stress factors. This work the influence of UV radiation on the flavonoids production in the callus cultures Genista tinctoria in vitro investigated. The highest production of flavonids was obtained at genistin under the exposure of callus culture to UV radiation of 254 nm wave lenght for the time 300 s with taking of sample after 48 hours. This callus culture grew on SH medium. The content of genistin was 3,03 %. Keywords: elicitation, elicitor, secondary metabolites, UV radiation, Genista tinctoria

The repair of DNA damage induced with suphur mustard and its relationship to the cytotoxicity
Jílková, Martina ; Vopršalová, Marie (advisor) ; Herink, Josef (referee)
The aim of this study was to investigate the induction and repair mechanisms of DNA damage caused by sulphur mustard in specific cell lines and their relationship to cytotoxicity of sulphur mustard. Sulphur mustard is a chemical warfare agent of the blistering agent category, which can be misused in local conflicts, terrorist attacks and during liquidation of its storage. Sulphur mustard is an alkylating agent, which interacts with a wide range of cellular macromolecules including DNA, RNA and proteins. Sulphur mustard forms single - strand breaks, monofunctional guanine and adenine adducts, as well as interstrand cross-links involving the two guanines in interaction with DNA. This study is aimed at cross-links. We used the Single Cell Gel Electrophoresis (SCGE, comet assay) for their detection. It is a method of evaluation of single cells ambeded in agarose. It is used for detection of DNA single strand breaks in a single cell. In our modification of the method we determined cross-links, which make the alkaline DNA unwinding impossible. That is why we had to induce a standard number of single strand breaks in DNA by styreneoxide (at a certain concentration and exposure time) before the comet assay. We studied DNA repair mechanisms of specific DNA lesions using specific cell lines with clearly...

Effect of compounds with antioxidant properties on glycation of aspartate aminotransferase
Bacílková, Eliška ; Boušová, Iva (advisor) ; Dršata, Jaroslav (referee)
Publishing place: Charles University in Prague, Faculty of Pharmacy in Hradec Králové Author: Eliška Bacílková Title: Effect of compounds with antioxidant properties on glycation of aspartate aminotransferase Supervisor: PharmDr. Iva Boušová, Ph.D. In this diploma thesis, I was concerned with the direct and indirect effects of uric acid, aminoguanidine, and hydroxycitric acid on enzymatic activity of aspartate aminotransferase and on its glycation by methylglyoxal. The catalytic activities were measured by kinetic UV method. Stabilities of aminoguanidine and uric acid, their interactions with the molecule of enzyme and influence on formation of glycation products were observed by absorption spectroscopy. Although aminoguanidine decreased enzymatic activity in higher concentration, it showed positive indirect effects on glycation of aspartate aminotransferase and it also decreased amount of rising glycation products. Hydroxycitric acid did not have negative direct effect on enzyme activity and was partially able to protect enzyme from glycation by methylglyoxal. Uric acid exerted neither negative direct nor positive indirect effects. Aminoguanidine and uric acid were stable during experiment and reacted with molecule of protein, which was expressed by modifications of its UV-VIS spectra in the area...

The study of interaction of platinum cytostatics with BCRP drug transporter using MTT cytotoxicity test
Horká, Markéta ; Čečková, Martina (advisor) ; Štaud, František (referee)
AAbbssttrraacctt The resistance of oncological diseases to cytostatic treatment is considered as one of the most serious problems of cancer treatment. One of the mechanisms by which cancers develop resistance is the overexpression of efflux transport proteins that limit intracellular concentrations of substrate agents. This work is focused on study of the interaction between clinically used platinum cytostatics (cisplatinum, carboplatinum and oxaliplatinum) and the efflux transport protein - BCRP. The experiments were made on MDKCII (parental cell line) and MDCKII-BCRP (cells were transfected by the gene of BCRP) cell lines. The results of the experiment with oxaliplatinum did not confirm any effect of BCRP on viability of BCRP transfected cells. Cisplatinum and carboplatinum tests provided evidence that BCRP transporter plays an important role in resistance development. The cells that were transfected by the gene of BCRP showed remarkably enhanced viability than parental MDCKII cells. However, the inhibition of BCRP transporter by specific inhibitor fumitremorgin C did not affect the viability of MDCK-BCRP cells. Such results suggest that the resistance of transfected cells was developed by other mechanism. Enhanced resistance could be caused by EGFP which is used as a reporting protein in preparation of...
