Comparison of HPLC a GC-MS method for pesticide determination in soil

Physical and chemical properties of pesticides may vary significantly. Pesticides can be acidic, neutral or alkaline. Some compounds containing phosphorus, other sulfur, nitrogen or halogens. These atoms can be important for the detection of pesticides. Some of the compounds are volatile, others on the contrary not evaporate, which is also one of the important properties for chromatography. The diversity of pesticides actually makes it impossible to create a universal method, which is very impractical in terms of quality and time determination. Because that pesticides are one of the most chemicals in most countries. In the food and drinking water are limits on pesticides in order to avoid negative impact on public health. For residues in soil or surface water limits are set. (Lutz et al., 2006) The objective of this thesis is to evaluate the possibility of using two types of chromatographs with different detectors for the analysis of pesticide. Pendimethalin which was chosen for this work, will be analyzed usinga liquid chromatograph with UV detektor and gas chromatography with mass detector. Both this method will be compared as well two extraction methods (Soxhlet extraction and shaking), and two extraction agents (methanol and acetonitrile). Another point of the work is to provethe hypothesis that, for the determination of pendimethalin in soil is by gas chromatography using a mass spectrometer in comparison with the high performance liquid chromatography time and more expensive, but will achieve a higher yield and a lower detection limit of the pesticide.

Impact of horizontal and vertical pattern of habitats on birds diversity

This diploma thesis follows up the influence of horizontal and vertical structure of biotopes on biodiversity of bird species of Czech Republic of forest and open sites. Vertical structure model was created from digital model of surface of Czech Republic (1st generation) and digital model of relief of Czech Republic (5th generation). Information about horizontal structure were derived by analyzing aerial imagery. Then single variables were detected from these data. Subsequently, it was analyzed which of these variables and which way influence bird diversity. On both types of sites was found increasing bird diversity with increasing diversity of vertical structure. With increasing horizontal structure, however, bird diversity significantly increased only on open sites. The influence of other characteristics was mostly different on bird diversity of open and forest sites, but, for both types of sites, the positive influence had for example edge density of land cover categories or edge density of height categories. Additional part of this thesis was to explore if vertical characteristics of biotopes could influence prediction of selected bird species. Results was different in forest and open biotopes, nevertheless in both types of sites was found potential use of both horizontal and vertical variables.

Monitoring of operation mode in road transport

Evaluation of a management working style

This thesis with a focus on "Evaluation of manager's management style" deals with managing styles that have been defined by its typical characteristics and described in the expertise literature. Individual styles of the senior personnel on their subordinate colleagues affect each individual in the team and also the overall outcome and meeting the deadline for completion of tasks. In the first part of the thesis are described specific types of management styles and their characteristics, typology of managers, their role in management and its history. The second part of the thesis leverages on theoretical knowledge and it evaluates specific control style for the three executives in the company Folklore Garden s.r.o .. These executives manage the same team of subordinates and should reach approximately the same performance results. In the practical part there was used survey with specific questionnaire. This part includes comparison of results referring to the prevailing management style of particular manager in terms of superior and subordinate workers but also from the manager himself. The result is evaluated through a managerial grid model. In the last part of the thesis there are mentioned opportunities for further qualitative and personal development of individual managers to avoid constant personnel changes that are for the company both financially inefficient and time-consuming.

Hodnocení historických konstrukčních spojů během vývoje úložného nábytku, jejich využití a optimalizace v současných podmínkách.

Dizertační práce zpracovává historický vývoj konstrukce a použitých materiálů úložného nábytku od 17. až po začátek 20. století a jej propojení (aplikaci) na současnou nábytkovou tvorbu. Zaměřuje se na rozbor vlastností spojů používaných ve výrobě nábytku. Na vybraných typech spojů nábytku budou vykonané experimentální testy, kde se bude zkoumat vliv rozměrů jednotlivých částí konstrukčních elementů vybraného typu spoje a druhu dřeviny na pevnost nábytkových spojů. Rovněž se bude zkoumat vliv typu použitého lepidla. Vybrané druhy lepidel budou určeny na nábytkové konstrukce. Jednotlivé pevnosti vybraných nábytkových spojů budou analyzovány s výpočtovým modelem pomoci metody konečných prvků. Pomocí počítačové simulace se budou jednotlivé typy nábytkových spojů porovnávat, s cílem najít nebezpečné koncentrace napětí. Tyto údaje poskytnou pro konstruktéry informace, kde je nutné daný materiál zesílit, případně zvětšit geometrické parametry konstrukčních elementů. Předpokládaným výstupem předkládaného projektu je návrh nových geometrických parametrů konstrukčních elementů a korekce dlouhodobě zažitých nedostatků uplatňovaných při tvorbě spojů.

This Bachelor Thesis focuses on motivation as a way of leading people. It deals with a research of motivation in relation to leading underlings from the point of view of a headworker, and searches for important motivation factors which could have an influence on employees´ work. The object of the study is to find out how important motivation is when leading people, and to focus on significance of motivation stimuli from the point of view of head workers. The theoretical part of the study deals with high-quality background literary researchwhich can provide adequate resource for the analytical - research part of this work. The theoretical resource includes information which enables us to understand better the general system of motivation from the point of view of head workers. The theoretical part emphasizes the choice of suitable literature allowing summary and comparison of important theoretical pieces of knowledge. The practical part of this Bachelor Thesis concentrates on thorough analyses of data collected through the qualitative research. The research was carried out by means of a controlled-interview method, with the interview outcomes recorded in structured questionnaires prepared in advance. The research sample group was formed by 8 respondents willing to have their interviews recorded. The interviews were followed by drawing concrete conclusions which summarize findings obtained from the research executed. The conclusion part of the study presents objective recommendations resulting from the research findings.

Staff Adaptation and Training & Development in Selected Organisation

The thesis is focused on the process of adaptation of employees and their development in the Prison Service, specifically Mírov. The main objective of this thesis, considering the outcomes of empirical research conducted to evaluate the control system adaptation and development of employees in terms Mírov and in case of deficiencies, propose appropriate measures. For this purpose it has been through surveys conducted by the prison staff survey of satisfaction with the process and their further development. In the theoretical part are summarized and characterized the concepts and knowledge concerning the process of hiring, orientation, training and personal development of employees. The practical part describes the organization and describes the process of hiring, orientation and training of new employees Mírov. Following a survey and analysis of collected data, their graphical representation and description of the facts. In conclusion sums up the shortcomings identified on the basis of their proposed innovation process of adaptation and development of employees, which can be applied to the whole Prison Service.

Genetic structure of local domestic breeds

A switch from hunting to beginnings of farming and agriculture was a crucial step for humans and for developing civilization. This period was suitable for domestication processes. Dogs were the first domesticated animals at all. Their breeding was connected with settlement and cattle protection and they were used as shepherd and hunting dogs. In some cultures they are even used as food source. Recent trends lead to breeding of new breeds and division of original gene pool into smaller closed groups which lead to an increase of homozygosity and to display recessively inherited diseases. These effects are mostly visible in small local breeds that have a small population size. Global protection of genetic diversity in cattle, poultry and pigs was already addressed in many studies, probably because of the high economic value of these animals. Impact of domesticated carnivores to humans is comparable and due to the growing market in veterinary services it is needed to protect sufficient genetic variability of various unique races. The aim of this work is to compare selected local breeds of dogs from different regions of origin, to evaluate their overall genetic diversity, to determine the occurrence of candidate types of diseases and to compare the frequency of these diseases in selected breeds. Main output is to recommend appropriate methods to protect the gene pool of these unique races. Central European breed (e.g. Český Fousek), subtropical breed (e.g. Sarplaninec), tropical continental breed (eg. Basenji), tropical island breed (e.g. The New Guinea Singing Dog) will be selected. The material will be collected at exhibitions and in cooperation with local farmers. The samples will be processed by classical methods of population genetics in the laboratory of molecular genetics FTZ ČZU.

Undeclared Employment in theTheory and Practise of The Labour Inspectorates

The thesis describes the issue of illegal employment, with emphasis on the analysis of illegal work in the Czech Republic and depending on the control activities carried out by labor inspectorates. Using the characteristics of the basic concepts (dependent work, illegal labor, labor inspectorates) with the help of current judgments given area, the current status and the possibility of the activities of labor inspectorates. The practical part deals, through interviews among the employees of labour inspectorates and experts, the specific situation of control activities and proposes solutions to streamline and harmonize the theory and practice of inspection activities in the illegal employment in the Czech Republic.

Management Accounting as a Source of Information in a Chosen Company

This thesis is focused on the role of management accounting in company management decision making in selected accounting entity. It is divided into three parts The theoretical part clarify the relationship between financial and management accounting and defining them within individual business processes. It also explains the economic structure and describing the cost centers, the costs breakdown from different perspectives, costing system, business planning and budgeting. The last part of theoretical background describes the phases of cost control, determination of deviations and improvement steps. The practical part contains general characteristics of the selected entity, description of the current state of the management information system, economic structure and the classification of profit/cost centers. Furthermore, this section describes the revenues of the company, costing system and types of calculations, budgeting and methods to monitor the costs. The last section, named "Evaluation of proposals and solutions", identifies areas with deficiencies and weaknesses. These areas are based on the theoretical knowledge and the findings made in the practical part and are followed by possible solutions, which would lead to the improvement in particular areas.
