Study of Formation and Elimination of Acrylamide in Food Matrix during Heat Treatment.

Acrylamide (AA) is a probable human carcinogen and undesirable contaminant which is produced by the reaction of reducing sugars with asparagine in plant foods during their thermal treatment above 120 °C. AA is most often determined by GC-MS and LC-MS/MS in isolates from the matrix in a wide range of foods. According to our observations, AA intake from food is higher among young people (from 1.8 to 3.8 µg/kg bw/day), which is consistent with the estimations of JECFA FAO/WHO from the year 2006. Considering the health risk, it is recommended to reduce AA formation in food during its processing, in particular exploiting the available experience. The aim of this thesis was to extend the knowledge of the possibility of AA elimination in selected types of thermally processed foods. The study was focused on cereal foods that contribute significantly to AA exposure, especially bread and sweet biscuits. The whole AA content in the bread is in the crust, which represents 5-15% of the bread. Crust of home-made bread contains approximately 30-75 µg/kg, however the marketed bread contains 2 to 10 times more of AA. This is due to the composition of bread mix, preparation conditions and baking. For maintaining the quality of home-made bread during the dry mixture shelf-life, optimization of bread mixtures was designed by increasing of yeast content, which proved positive effect on the reduction of AA content at sufficiently high activity of the yeast. Monitoring of AA content in assortment of sweet bakery products showed higher levels of AA in diabetic biscuits containing fructose instead of sucrose. Three of them even exceeded the reference value (500 µg/kg) more than 1.5 times for commodity "cookies". Elimination of AA by applications of the enzyme asparaginase has been designed for minimal interference in technology of their production. The concentration of the enzyme and the appropriate method of its use in industrial environment have been tested previously in model systems. In optimized conditions of the enzyme application, AA content in diabetic biscuits was reduced by more than 40% without affecting the organoleptic properties of the final product. Effect of the antioxidants on AA formation was also part of the study. AA content in gingerbread was reduced efficiently by the use of fennel, anise, cloves, vanilla and white pepper (by about 9-21%). Conversely, coriander and cinnamon significantly increased its content (by 18-54%). Since correlations between the DPPH• radical quenching activity of the spice extracts and AA content was not observed, the final content of AA was probably influenced by the chemical composition of spices and reactivity of the individual components in the matrix. Investigated methods appear to be suitable ways of elimination AA in some foods; however their specific use must be optimized with regard to the composition of the food, processing and the technology used. Estimated impact of application of the above-mentioned methods to the overall elimination of AA exposure showed that its intake in high school students from the Czech and Slovak Republic can be reduced on average by 10%. This decrease is a success to reduce the possible risk of cancer disease by eating foods with a high AA content. It is also important piece of information for food producers for further development of relevant methods for AA elimination which would help to reduce the AA intake from foods even more.
The Gallery of Brno Grand Prix

The main task of this thesis was to create an architectural study of The Gallery of Brno Grand Prix based on previous urban study from previous semester works. The proposal is focused on the iconic circuit’s former glory restoration, on providing new content for raceway complex and creating decent and presentable place for the presentation of the rich history of Czech and Czechoslovak motorsport. The result is a study of an object that meets these needs while respecting the historical and morphological values of the place. The building creates new themed attraction and creates the potential for further development of near circuit.
The Repechy Guest-House

Bachelor’s thesis is based on a studio project created in the summer term of the third year of the undergraduate studies. The task was the renewal and the extension of the farmhouse for an accommodation facility and other services. The farmhouse is situated in a small village Repechy near the town of Prostejov and is complemented by two other unused buildings. In the second half of the 20th century, rectangular shaped farmhouse with a yard was roofed, interior walls were partially demolished and interior space was used for keeping the cattle. It had a big impact for the current condition of the object. Aggressive environment and high humidity, for which structures were not designed, significantly accelerate the degradation process. Later, the cattle was moved away and the original farmhouse is deteriorating until these days. Currently, the object and its surroundings are in a bad condition. Most of the constructions are damaged and it is not possible to use them anymore. Northern tract is the only part worth protection. There are preserved precious historical chambranles and other plastic decoration. Surroundings is overgrown with wild vegetation. The aim was to preserve and bring out valuable parts of the original object and find them appropriate use under specified conditions. The proposal deals with the removal of three sides of a rectangular farmhouse and preserving the northern tract in the form of a torso. Extension is based on the original farmhouse. Extends the original built up area boundary to use existing structure and creates the first volume, which symbolizes the traditional static form. Rotated duplicate of the first volume then characterized a modern and dynamic attitude in architecture. The final design of an extension is created by synthesis of both volumes as an evidence of the coexistence of a traditional and a modern form. Znojmo – Castle, rotunda and brewery

Urbanistic and architectural study of future development of castle area in Znojmo. Main focus will be on creation of sustainable and vital part of the city with connections to landscape, townscape, castle, old brewery and romanesque rotunda of st. Catherine. Adaptation of historic buildings of castle and brewery for cultural and social center of town. Reconstruction and conversion will be based on theoretical work from previous semester. Urbanistic goal is creation and redefinition of public spaces of castle garden, rotunda hill and Hradni street and its integration with historic town center of Znojmo and valley of river Dyje.
PUBLIC CITY SPACE – RUDOLFOV

The main goal of my work was find and define a form of content for potential reconstruction of the castle Rudolfov. The purpose was a make this castle a good place for serving to city. This thesis is a last from analyzes and proposals for dealing with the city Rudolfov and its public space, which I developed in the whole masters degree: City - Vision for the Future. My work has been divided into four semesters. In first one, i have made a detailed analysis of the city Rudolfov and identifying the problem areas and main city problems, which I also discussed. In the second semester the work was focused on new study for new city center. In the third semester, I continued working on urban development studies of the northern slope with relation to the newly created center. For the thesis I chose the troubled and valuable place of the city with a cultural monument that formerly served as barracks and today it is looking for a new use. All studies were designed to contribute positively to the future development of the town Rudolfov.
Software for CCD camera used for electroluminescence

This thesis studies the optimization of diagnosis defects of the photovoltaic cells in the darkroom by the help of new operating software for the used CCD camera. First the present situation and the possible solutions are analyzed. Then the attention is paid to the theory of light-sensitive CCD sensors, their construction, their principles of operation and the theory of scanning images. The thesis also focuses on the analysis of the function of the operating libraries which were provided by the equipment producer. The key part is the development of the application for the camera G2-3200 of the "Moravske pristroje" company which significantly accelerates the work with the camera in the darkroom.
THE ARCHEOLOGICAL MUSEUM - REBUILDING OF ST.HYPPOLITS AREAL IN ZNOJMO

The main goal of this project is to find new use of former monastic grange for archaeological base and exhibition combined with multipurpose building containing gallery space, workshop room, accommodation and restaurant. The courtyard should become new public space (during opening hours). Znojmo-Hradiště (also known as Hradiště sv. Hypolita) is an important archaeological site with numerous finds from the period of the Great Moravia. The monastery and the grange are situated in the centre of former fortress. The complex of the monastery and all its buildings with few exceptions are cultural monument.
Gallery of modern art

The conceptual design of the Gallery of Modern Art is created with the aim to build the timeless social and cultural center, situated in urban conservation zone, which is in the connection with the cultural axis of the existing round avenue of Brno. The proposed building is located in the Moravian Square, in the place of the existing park. It is a neoplasm in the historic space, which is adding the uniqueness of surrounding by its concept. This proposed building will reflect the most modern architectural, building and construction elements.
Olomouc Central European Forum

The presented diploma thesis was elaborated as an architectural study of a Central European Forum in Olomouc (SEFO). Campus SEFO will be created as an reconstruction of the Museum of Modern Art (MUO) in Denis street and building in a neighboring vacant lot. The proposal involves urban, architectural, operational layout, design and material solutions objects in spatial context. Within SEFO and MUO they are created each operation - stand-alone units. Objects SEFO and MUO are interconnected. It is necessary to respect the separation of publicly accessible areas of compartments accessible only by employees. Architectural study includes space for exhibitions, library, multi-purpose space with facilities, vestibule usable for exhibition openings and other cultural activities, chamber music performances, as well as facilities for education, technological facilities of the building, the depositary (transport and central), photo studio restoration studio, office space, locker rooms and restrooms personnel. SEFO specific aim of capturing the diverse manifestations of visual culture of Central Europe after World War 2, the building's permanent exhibition, acquisition activity, temporary exhibitions, including larger medium-shows (eg. The biennial or triennial), discussion forums and other supporting cultural events.
Thonet vila Korycany

This Bachelor’s thesis is based on an architectural study made in winter semester of the third year of bachelor study program on the topic of “Thonetova vila v Koryčanech”. The task of this study was to find out new and suitable working and spatial solution of current building of the factory owner Thonet’s family house. The building should be used for holding public events, e.g. weddings, exhibitions or public screening. Small ceremony room, wine room, exposition of Thonet and his factory for bentwood furniture are parts of the building. Two housing units for temporary living are also designed in the attic. Another part of the compound is house of culture, which is not part of this project. There is an original park in the surroundings of the family house. The project considers some park changes on the town’s land. The compound should be more open to public, show its visitors parts of the factory owner Thonet’s work and other natives of Koryčany. It should be more used by the citizens for relaxing or education together with park too. The work is divided into documents, constructional study, constructional part of the project documentation for construction and architectural detail.
