Alternative fuels for transport

The thesis Alternative fuels for transport is dealing with the topic of economic competitiveness and environmental advantages of alternative fuels in comparison to classical oil fuels in transport. First, the main transport related problems are discussed in detail, including the dependency on oil, emissions from transport and their impact on human health and the environment. Then, the different types of alternative fuels are presented, including the legal framework, possibility of tax exemption and subsidy financing. The comparison of emissions of alternative and classical fuels follows the logic of the whole life cycle of a fuel, using the Well-to-Wheel analysis as method. Emissions of greenhouse gases and emissions of pollutants of local importance are observed. The cost comparison is based on experience in public bus transport in the Czech Republic. Fixed, variable and total costs are compared for classical diesel, biodiesel, natural gas, LPG, hydrogen and an electric vehicle. The purpose of the study is to decide which of the alternative fuels represents a cost competitive option in comparison to classical diesel and also provides a reduction in total emissions. The results of the study are designed to help Krkonošské Metro Association to choose a suitable alternative fuel for a project in rail transport.

Laboratory diagnostics of micrometastases in breast cancer patients

Introduction: The presence of circulating tumor cells (CTC) in the peripheral blood has been associated with worse prognosis and early relapse in breast cancer patients. CTC determination in the peripheral blood has been considered as a liquid biopsy. The aim of this project was to analyze the presence of CTC followed by their molecular characterization with the potential use not only as a new biomarker for real-time monitoring of therapy efficacy but also as a suitable tool for patient's stratification and individualization of treatment for breast cancer. Methods: A total of 54 patients with diagnosed early breast cancer were enrolled into a prospective study. Ten millilitres of peripheral blood were sequentially collected to test for the presence and characterization of CTC during the follow-up of patients. CTC isolation and detection was performed by AdnaTest BreastCancer™ (AdnaGen AG, Germany), which is based on the detection of EpCAM, HER2 and MUC1 specific transcripts in enriched CTC- lysates. cDNA from isolated CTC has been further used for newly optimized qPCR assays for breast tumor and therapy resistance associated genes: TOP1, TOP2A, CSTD, ST6GAL, KRT19 and reference gene actin. qPCR results have been analyzed by Genex software (MultiD Analysis). Biomass boilers

This bachelor´s thesis concerns with the biomass boilers and the biomass as a renewable source of energy. The first part informs us about renewables sources of energy, concerns with biomass and with production of fuel from biomass. The second part describes the biomass boilers. Bellow it shows the possibillities of heating, which the todays market is able to offer us. It mainly deals with the posswibillities of haeting of biomass and compares the most economical boiler, which is used for different fuel. The calculating part is specialized in comparing of biomass boilers with gas boiler from economical standpoint.

Epigenetic regulation of retroviruses.

The human genome is the site of integration of the retrovirus HIV-1, ERVWE1 (syncytin-1) is its stable part. Thus as an integral part of the genome, they are under influence of important genome based epigenetics regulations, such as DNA methylation, histone methylation and acetylation. Promoter DNA hypermethylation and histone deacetylation leads to establishment and maintenance of latent state of the HIV-1 virus and formation of the latent reservoir in the CD4+ memory T-cells. This process leads to severe problems during HIV-1 infection treatment by antiretroviral therapy (HAART), as the HIV-1 latent reservoir remains unaffected. Moreover DNA hypermethylation in the promoter of syncytin-1 directs its transcriptional silencing, which is important in tissue specific regulation of this fusogenic protein. In physiological conditions syncytin-1 expression is observed only in placental cells, where the DNA methylation of promoter is decreased. Higher expression level of syncytin-1 was also observed in several other tissues, such as testes, where it is tightly coupled with germ-cells tumors and syncytin-1 promoter hypomethylation. Russian view on energy security

The thesis is dedicated to the Russian view on energy security. Its aim is to evaluate how successful is Russian energy policy in sustaining energy and economy security in longterm perspective, and how well it deals with current and future potential problems of energy sector. The topic became of interest due to recent oil price drop, which led Russian economy to crisis and gave rise to discussion on its dependency on fossil fuel exports. First chapter is dedicated to definition of energy security of Russia as an exporting country. Next chapter is dedicated to SWOT analysis of current state of energy sector and defines its strengths, weaknesses, opportunities and threats. Last chapter evaluates how successful is Russian energy policy in relation to defined energy security and performed SWOT analysis.

Context of macroeconomic development and energy sector in the Czech Republic and Austria in the period 2000 - 2012

The aim of this diploma thesis is to verify a connection between macroeconomic development and energy sector. This connection is examined with the data from the Czech Republic and with the data from Austria, both in the years 2000 - 2012. The theoretical part describes energetics as a natural monopoly and a resource of externalities. It includes the theory of monitoring the business cycle and financial analysis of companies. The first half of the practical part compares macroeconomic development of the Czech Republic and the ČEZ group. The similar comparison is made with Austria and the Verbund company. Second half of the practical part explains how economic policies influence the Czech energetics. This impact is being studied in terms of legislation, regulations, monetary and fiscal policy. The practical part ends with rating and valuation of costs that arise from the main energy resources. The link between energetics and macroeconomic development was discovered and proved thanks to the doubled analysis used in the thesis.

Semi-automated assessment of micromechanical properties of the metal foams on the cell-wall level

Metal foams are innovative porous material used for wide range of application such as deformation energy or sound absorption, filter material, or microbiological incubation carrier. To predict mechanical properties of the metal foam is necessary to precisely describe elasto–plastic properties of the foam on cell–wall level. Indentation with low load is suitable tool for this purpose. In this paper custom designed instrumented microindentation device was used for measurement of cell-wall characteristics of two different aluminium foams (ALPORAS and ALCORAS). To demonstrate the possibility of automated statistical estimation of measured characteristics the device had been enhanced by semi-automatic indent positioning and evaluation procedures based on user-defined grid. Vickers hardness was measured on two samples made from ALPORAS aluminium foam and one sample from ALCORAS aluminium foam. Average Vickers hardness of ALPORAS foam was 24.465 HV1.019 and average Vickers hardness of ALCORAS was 36.585 HV1.019..

Computational Simulation of Mechanical Tests of Isolated Animal Cells

Buňka tvoří složitý biologický systém vystavený mnoha mimobuněčným mechanickým podnětům. Hlubší pochopení jejího mechanického chování je důležité pro charakterizaci její odezvy v podmínkách zdraví i nemoci. Výpočtové modelování může rozšířit pochopení mechaniky buňky, která může přispívat k vytvoření vztahů mezi strukturou a funkcí různých typů buněk v různých stavech. Za tímto účelem byly pomocí metody konečných prvků (MKP) vytvořeny dva bendotensegritní modely buňky v různých stavech: model vznášející se buňky pro analýzu její globální mechanické odezvy, jako je protažení nebo stlačení, a model buňky přilnuté k podložce, který vysvětluje odezvu buňky na lokální mechanické zatížení, jako třeba vtlačování hrotu při mikroskopii atomárních sil (AFM). Oba zachovávají základní principy tensegritních struktur jako je jejich předpětí a vzájemné ovlivnění mezi komponentami, ale prvky se mohou nezávisle pohybovat. Zahrnutí nedávno navržené bendotensegritní koncepce umožňuje těmto modelům brát v úvahu jak tahové, tak i ohybové namáhání mikrotubulů (MTs) a také zahrnout vlnitost intermediálních filament (IFs). Modely předpokládají, že jednotlivé složky cytoskeletu mohou měnit svůj tvar a uspořádání, aniž by při jejich odstranění došlo ke kolapsu celé buněčné struktury, a tak umožňují hodnotit mechanický příspěvek jednotlivých složek cytoskeletu k mechanice buňky. Model vznášející se buňky napodobuje realisticky odezvu síla-deformace během protahování a stlačování buňky a obě odezvy ilustrují nelineární nárůst tuhosti s růstem mechanického zatížení. Výsledky simulací ukazují, že aktinová filamenta i mikrotubuly hrají klíčovou úlohu při určování tahové odezvy buňky, zatímco k její tlakové odezvě přispívají podstatně jen aktinová filamenta. Model buňky přilnuté k podložce dává odezvu síla-hloubka vtlačení ve dvou různých místech odpovídající nelineární odezvě zjištěné experimentálně při AFM. Výsledky simulací ukazují, že pro chování buňky je rozhodující místo vtlačení a její tuhost určují aktinová povrchová vrstva, mikrotubuly a cytoplazma. Navržené modely umožňují cenný vhled do vzájemných souvislostí mechanických vlastností buněk, do mechanické úlohy komponent cytoskeletu jak individuálně, tak i ve vzájemné synergii a do deformace jádra buňky za různých podmínek mechanického zatížení. Tudíž tato práce přispívá k lepšímu pochopení mechaniky cytoskeletu zodpovědné za chování buňky, což naopak může napomáhat ve zkoumání různých patologických podmínek jako je rakovina a cévní choroby.

The strategic importance of Serbia to the European Union

Doctoral dissertation presents an analysis of the Serbian economy in the mutual relations between the European Union (EU) and the Western Balkans. The assumption that the EU and Serbia, as an integral part of the Balkan region, have interests in the partnership and cooperation is the baseline of the research. The following aspects have been singled out, by method of ranking, which is based on achieving the highest turnover in the last six years: trade, foreign direct investment and energy. Gravity model was used to analyze routes of trade and energy. A models of trade import and export between Serbia and the EU 28 member countries and the countries of the Western Balkans, as well as models of imports and exports of energy between Serbia and the EU 28 member countries and the countries of the Western Balkans, were used in order to assess the potential of trade and energy trade. Method of trend line and trend extrapolation of foreign direct investment (FDI) was used in order to determine the degree of the presence of FDI in Serbia and their predictions, which could be an indicator of investment climate in Serbia. The conclusion is that Serbia has strategic importance as a candidate for membership in the EU because of its regional importance. Entering the Western Balkan countries in the EU should not happen without Serbia, because Serbia has a regional importance, which arise from the economic, historical, cultural, infrastructural and geopolitical links with countries in the region, of which some are already members of the EU.
