Možnosti vinifikace hroznů odrůdy Frankovka pro VOC Modré Hory
Hloušek, Petr
In the thesis entitled The possibilities of vinification of grapes of the variety Frankovka for VOC Blue Mountains, the primary objective was to evaluate the influence of autochthonous yeast on the commercial by fermentation in an automatic vinifier, an open and closed vessel. Next, find out by ripening the wines in a wooden barrel and an impervious container whether it suits more maturation with increased or minimal amount of oxygen. The result of this work was then the sensory analysis of the wine was quantified into a 100 point table according to the OIV as well as the aromatic profile and profile of the structure and strength of the wines, which were then processed into the graphs. Experimental samples were analyzed by individual parameters - anthocyanins, total polyphenols, antioxidant capacity and color parameters.
Dozrávání meruněk podle látkových složek při rozdílné teplotě plodu těsně po sklizni
Nekužová, Nikola
Two apricot varieties that were harvested at the beginning of climacteric phases were evaluated. The apricot varieties used were harvested on the plot of the Horticultural faculty in Lednice. This is a variety of Leskora harvested on 22 June 2016 and Betinka variety harvested on 1. 7. 2016. The fruits were stored at 1°C, 5°C and 20°C for 9 days. Mass losses, soluble suspensions, titration acids and penetrometric strength were assessed after each three days of storage, and physiological parameters were assessed by ethylene production and breathing intensity (CO2 production).
Hodnocení odrůd meruněk při dozrávání na stromě
Kochová, Eliška
This diploma thesis on the topic of "Evaluation of apricot varieties during ripening on the tree" describes in the theoretical part the characteristics of apricots, significant material components of apricots fruits and methods of their determination, as well as the ripening of apricots and changes of storage substances during ripening and storage. The diploma thesis deals with the changes in the material composition during ripening on the tree and storing for 4 and 8 days at 20 °C in three stages of ripeness (unripened, ripe, overripe). In the practical part two varieties of apricots were monitored ('Leskora', 'Goldrich'). The source of the fruits varieties was the school orchard at the Faculty of Horticulture in Lednice. Changes of fetal body weights, weight of stones from fruits, fruit firmness, soluble solids content and titratable acids content were regarded. Further, was determined ethylene production and carbon dioxide production. Ripening index was calculated from the values of fruit frimness, soluble solids and titratable acids. The measured values were presented in the form of spreadsheets and graphs and were evaluated. The achieved results are valid for particular year and location.
Development of new technology of sand autoclaved aerated concrete with using of secondary raw materials
Ondříčková, Pavlína ; Suchý, Peter (referee) ; Drochytka, Rostislav (advisor)
Autoclaved sand aerated concrete is a modern building material with high thermal insulation properties. Only sand is used as the primary silicate component of aerated concrete in the Czech Republic. For the more favourable economic and ecological properties of aerated concrete, the use of secondary raw materials is used in this work. The secondary raw materials examined included fluidized bed and fly ash, slag and glass recycled. The aerated concrete composite with secondary admixtures was developed under hydrothermal conditions of a laboratory autoclave for 7 and 12 hours at 190°C. From the secondary raw materials tested, a 10% glass recycler additive yielded the highest strength. Other additives that have a positive effect on mechanical properties include SAKO and Oslavany. From the results of the work it is evident that the use of secondary raw materials increases the strength, improves the rheology of the mixture and supports the formation of tobermorite.

Transcriptional activity of the genes characterizing developmentally competent cytoplasm in bovine oocytes.
Pešanová, Denisa ; Němcová, Lucie (advisor) ; Kalous, Jaroslav (referee)
4 Abstract The antral follicle provides a specialized microenvironment or niche, which is necessary for production of high quality oocyte. The developmental competence of bovine oocyte is influenced by the follicle size. Oocytes originated from medium or larger follicles (≥6 mm) have greater developmental competence (ability to develop to the blastocyst stage). The changes in cytoplasmic factors, for example mRNAs, could explain differences in oocyte developmental potential. Using Bovine Oligonucleotide microarrays the differences in gene expression profiles of oocytes at germinal vesicle and MII stages from medium (MF, 6-10 mm) or small (SF, 2-5 mm) follicles were characterized. The aim was to find differencies between oocytes diverse developmental competence. The expression fold change between the two experimental groups was in 61 genes. Subsets of 15 differentially expressed genes were validated by quantitative RT-PCR. Before maturation, significant differences were confirmed at the level of ATP5C1, MAP3K13, MTRF1L, TAF1A and UBL5. Subpopulations of oocytes were classified according to atresia of cumulus cells and follicle size. We determined the level of 12 individual transcripts after maturation. ATP5F1 remained stable in all experimental groups of oocytes. The level of BRD7 transcript remained stable...
Psychological and spiritual guide through middle-age crisis
Klimeš, Václav ; Lachmanová, Kateřina (advisor) ; Bartůšková, Ludmila (referee)
The thesis "Psychological and Spiritual Guide through Middle-Age Crisis" contains a synthesis of knowledge about middle-age crisis from the interdisciplinary point of view of psychology and spiritual theology in its first part. The fundamental opinion declares that middle-age crisis is not something to be suffered through but, in spite of all its demands, it is an opportunity and challenge for fuller and more authentic life, for maturation of a man and for getting closer to oneself and to God. In the second part of the thesis, knowledge and procedures of psychotherapy, crisis intervention and pastoral theology are used to concretize and extend the conclusions of the first part in a more practical way; hence, the paper can be used as a guide for those who are undergoing this period of life or those who want to accompany someone else on this way. The thesis is intended primarily, but not exclusively, to Christians who lack education in the psychosocial area and who is middle-age crisis a personal topic of themselves or of people around them for-be it their relatives or friends, or, in case of pastoral workers, clients in their professional care. This work is rather singular in the Czech language due to its conception, scope and synthesis of its starting points as well as their elaboration. Keywords...
Dispersed reinforcement influence on the maturation of polymercement materials
Zaťko, Petr ; Jakubík, Aleš (referee) ; Bydžovský, Jiří (advisor)
The diploma thesis is focused on polymercement materials with dispersed reinforcement. The aim of this work is development of suitable polymercement mixture and monitoring of the impact of dispersed reinforcement on the course of maturation of these materials and physico-mechanical properties. The effect of high temperatures on the properties of the mortar is also examined. Emphasis is placed on the use of alternative resources and byproducts. Mixed binder of cement and slag and admixture of microsilica was used. Recycled cellulose was used as dispersed reinforcement and was compared with commercial polypropylene fibers.
Comparison of individual development of oocytes maturation of sterlet (Acipenser ruthenus) females during the prespawn period at different temperature conditions
KNOWLES, Jindřiška
The aim of presented work was a study of the effect of temperature on the development of mature oocytes of sterlet during prespawn period. At the turn of 2013 and 2014 was conducted our experiment with females of sterlet. Experimental fish were divided into two main groups. The first group was exposed to the heated water and a second group was kept in natural water temperature. Four biopsies were carried out during attempt. The samples were measured, and then was calculated oocyte polarization index (PI). For more accurate results processing fish were divided into two subgroups within both groups. 1st subgroups had at first biopsy PI 12.5 % and 2 subgroups PI> 12.5 %. In less mature females the effect of temperature was statistically significant (p < 0.05) while in more mature females the opposite was true (p > 0.05). From a total assessment follows that the temperature had no significant importance for the rate of oocytes maturation (p > 0.05). Experimental results indicate that the heating method can be used to accelerate the maturation especially in less mature fish and for synchronization spawning.
