Speciation analysis of selenium in selenized yeast
Motlová, Tereza ; Vitoulová, Eva (referee) ; Diviš, Pavel (advisor)
The aim of the theses was determination of selenium species in yeast Saccharomyces cerevisiae cultivated in medium with added inorganic form of selenium (Sodium Selenite). Concentrations of Sodium Selenite in cultivation medium were 0,1; 1; 10 and 100 mg.l-1. Cultivation was undertaken in fermenting tub for period of 72 hours. Cultivated yeasts were extracted by use of enzymes and subsequently the species of selenium in particular parts of yeasts were determined. In order to determine selenium species, the method of high-performance liquid chromatography in combination with atomic fluorescent spectrometer and technique of hydride generation was used. Having analysed different fractions of the yeasts Saccharomyces cerevisiae it was ascertained that during cultivation the sorption of selenium occurred in form of Se4+ in cell membranes while in cytoplasm no inorganic forms of selenium were found. Furthermore, it was stated that yeasts Saccharomyces cerevisiae are able to metabolically change inorganic forms of selenium to organic forms (selenomethionine), while these forms are present in cytoplasm and they are likely to be bound to proteinic structures of cell membranes. An increase of concentration of Se4+ in cell membranes could be observed as a result of increasing concentration of Sodium Selenite in cultivation medium. In proteinic structures the concentration of organic selenium forms increased only to concentration 10 mg.l-1 of Sodium Selenite in cultivation medium.
Taxonomic submission of Saccharomyces yeast from selected matrices
Mašitová, Lucie ; Vadkertiová, Renata (referee) ; Vránová, Dana (advisor)
This thesis explores the optimizing of the methods relevant to the cultivation, isolation, and identification of individual yeast strains from selected matrices, using the molecular biology methods. Grape berries, vine-leaves and soil from vineyard have been used as matrices. As yeasts are an integral part of fermentation processes, they are used for making of wine, the organoleptic charakteristics of which they influence. For identification of yeast strains the PCR-RFLP method has been used. Specifity of yeast DNA sequences of certain species has been used for this analysis. These spacers have been amplificated through the use of PCR and consequently they have been subjected to a restrictive analysis with specific restrictive endonucleases. These fragments have been detectioned through horizontal electrophoresis. In the literature background research section the basic informations about viticulture, yeasts, their cultivation and separation, PCR, restrictive analysis and horizontal electrophoresis is presented.
Aromatic substances of wine
Stávková, Markéta ; Vespalcová, Milena (referee) ; Ruibar, Jiří (advisor)
This bacherols thesis consists of two parts, of theoretical part and practical part. The theoretical part delimits wine-producing regions in Czech republic, explains growing of grapevine. The thesis is concerned with the soil classification, amount of mineral element and decribes the quality and aroma of a wine. It´s briefly concerned with bouquet of wine during wine production. Deals with chemical changes in must during fermentation using yeast. The practical part is concerned with soil analysis of vineyard Rajhradské Klášterní. The goal of this experimental part was to finding out amout of mineral elements in samples and their comparison between the four vineyard. Aromatics are analyzed by gas chromatography and sensory analysis.
The effect of pesticides on the grapevine microflora
Damborský, Pavel ; Pořízka, Jaromír (referee) ; Omelková, Jiřina (advisor)
The bachelor thesis of theoretical character contains information about an effect of pesticides on grapevine microflora. At the beginning there is a description of general pesticides characteristic, their classification, toxicity and a brief overview. Further it describes illnesses and pests that often attack grapevine. The next chapter deals with grapevine microflora. It defines morphology, taxonomy and reproduction of microorganisms most often occur on grapevine and during technological processing of wine. The last chapter is focused on effect of pesticides on microorganisms during alcoholic and malolactic fermentation.
Raw materials and microorganisms to distilled beverage production
Jechová, Iva ; Vitoulová, Eva (referee) ; Babák, Libor (advisor)
The production of distilled ranks among established fermentative production. Than the first – quality spirits is became from raw material, it is obliged to change ranks processes. The important from the processes are alcoholic fermentation, which proceeds owing to mikrobiological cultures, distillation and subsequent rectification and at not least ranks aging distilled, for which is necessary selected appropriate container. For all processes the granted conditions are appointed, under which the processes would ocur, so resulting distilled achieved the first – quality and the best sensory character. Therefore this work does one’s best map these single processes and it is largely intent on fermentative processes.
Brewing industry
Artýszková, Jana ; Illková, Kateřina (referee) ; Omelková, Jiřina (advisor)
The aim of the theoretical part of the thesis is the study and the treatment of the problem of the manufacturing of beer, focused on the czech conditions and classical Czech technology. Within the theoretical part the history of this branch of industry together with the modern methods, enabling greater productivity of beer production, are summarized. Experimental part of the thesis was carried out in a small brewery in Czech republic, producing aproximately 10 000 hl per year. In this brewery the selected parameters were studied not only in the course of cooking and fermentation of beer, but during the output control in racking to the drums and plastic bottles, as well. The studied parameters were as follows: the sugar content of front and wort, the control of microbiological contamination in fermenting cellar and in beer-storage cellar, the content of CO2 by last stage of fermentation and pH- value and color by the end-product.

The evaluation of horse nutrition with chosen preparats in sommer and winter season
HANETŠLÉGROVÁ, Petra
ABSTRACT The aim of this work was to provide a detailed breakdown of horse nutrition, to mention the importance of dietary supplements. The main part deals with the basics of horse nutrition, the bare necessities of nutrients, the proportion of maintenance rations and the technique of feeding. Afterwards, I outline the properties and effects of varied herbal plants such as garlic, hemp, hyssop, white horehound, coltsfoot and oats. Flaxseed, biotin, zinc, yeast and probiotics are dietary supplements improving the quality of fur, horsehair and hoofs. The feed will be analysed by standard methods. As far as horse excrement is concerned, we are going to observe its shape, colour, consistency, pH value and smell. The assessment of horse excrement will serve as a judgement about the activity of digestive system. The ethology observations will provide information on daily activity of a horse, on particular activities, to which the horse gives its attention during a day, and the amount of time it dedicates to them. As far as horses with respiratory disorders are concerned, we will observe the signs of their health problems and the rate of these problems occurring both at rest and in action. The information will be completed by the measurement of body proportions useful to body weight calculation. The statistical and economic methods will be selected in terms of the results accomplished during the research.
Studie úloh TIF34/eIF3i a TIF35/eIF3g subjednotek eIF3 v iniciaci eukaryotické translace
Janoušková, Olga ; Nielsen, K. H. ; Valášek, Leoš
Translation initiation is a multiple-step process involving a large number of soluble eukaryotic initiation factors that ultimately leads to formation of the 80S ribosome with Met-tRNAiMet based paired with the start AUG codon of mRNA. Among the initiation factors, eIF3 draws a special attention since it was shown to promote several critical steps such as recruitment of the eIF2GTPMet-tRNAiMet ternary complex and mRNA to the 40S ribosomal subunit and was implicated in cancer
