Molecular analysis in cases of inherited diseas

Urcení vztahu mezi onemocnením a jeho molekulární podstatou je jedním z hlavních cílu lékarské genetiky. V rámci studie: "Molekulární analýza vybraných dedicne podmínených onemocnení" byly popsány postupy, které vedou k objasnení molekulární podstaty u onemocnení s již známými odpovednými geny i u onemocnení, u kterého byl odpovedný gen teprve identifikován. Studie prispela k rozšírení znalostí zkoumaných dedicných onemocnení. Zavedené postupy molekulárních analýz jsou soucástí diagnostiky na Ústavu dedicných metabolických poruch v Praze. Výsledky techto analýz slouží k prevenci, prognóze i lécbe onemocnení a u postižených rodin byla umožnena prenatální diagnostika. Výsledky byly publikovány v nekolika domácích i zahranicních odborných casopisech a prezentovány na mezinárodních konferencích. Nekteré z publikací byly dále citovány.

Molecular factors of artherogenesis in chronic kidney disease with experimental model of chronic inhibition of Nitric Oxide Synthesis and its Effect of HMG-CoA reductase inhibitoin

The experimental part of the thesis covered the markers involved in nitric oxide deficiency model in rats. The analogue of arginine N-nitro- L -arginine-methyl ester ( L -NAME) was used. L -NAME administration resulted in sustained elevation of BP, decreased GFR, and in higher albuminuria compared to control animals. The administration of hypolipidemic drug - atorvastatin to Wistar rats was studied. The statin used caused lower albuminuria and prevented decreases in GFR in L -NAME rats without having an impact on hypertension. CAV1 protein expression was similar in all groups of rats. In contrast, VEGF expression and RhoA activity was increased in L -NAME-treated animals, and normalized with coadministration of atorvastatin. Clinical part of the thesis focused on the analysis of markers of oxidative stress in obese and non-obese patients with CKD and after kidney transplantation. The patients with CKD had elevated serum concentrations of AGEs, sRAGE and ADMA with the maximal concentrations of AGEs in the patients on hemodialysis. The successful kidney transplantation decreases the AGEs, sRAGE and ADMA concentrations but not to normal value according to general population. Internet marketing is currently one of the most important components of marketing. However, Internet marketing as a whole is much more important than mere webdesign, mere search engine optimization (SEO) or a simple PPC advertising. Most companies have recently made it throught with a website and that was all their presentation on the Internet. However, the internet is one of the fastest growing technology today and the number of its users is growing every year. Constantly there are new technologies that allow us to have a better user experience and also bring us new ways of presentation and promotion. Most of the companies make a mistake early on and underestimate the preparation, don't know their goals, customers or competitors. Internet marketing is the process and the contents of this process is the recognition, analysis, design, implementation, and ultimately verify that you achieve the goals of the organization. In the theoretical part it is defined internet marketing and its advantages over offline marketing. Briefly sum up the history and major milestones of Internet marketing that shaped its present form. Further, it's described current trends in Internet marketing and detail expands upon the selected Internet marketing tools and tools for evaluating the success of campaigns. The practical part deals with the proposal of marketing strategy in the company Imperium Finance. The main objective is to analyze the requirements of the company Imperium Finance, from analysis to draw specific conclusions and devise a marketing strategy that will support the vision and strategy of the company in the following period. All steps were carried out within the company Imperium Finance on real data from active clients and tested in action. The result is a proposal of a marketing strategy that will drive Imperium Finance, and a marketing plan for the company. The biggest benefit of this work is so real use theoretical knowledge in practice.

The possibilities of use of the robotic system da Vinci for treatment of localized prostate tumor

Cancer of prostate is the most frequent male malignity and also it is the second most common cause of death in Europe and in the USA. There are 2 228 new cases reported in the Czech republic per year. The probability of occurrence of this disease is one to six during whole life. There is a good success in detection of his disease in the early stage due to development of screening methods. In such case the radical prostatectomy is very successful method of treatment. Besides of operation methods like open radical prostatectomy and laparoscopic prostatectomy there is a new operation method using robotic system da Vinci. The aim of this machine is to bring the advantages of the miniinvasive surgery together with keeping intuitive handling like in open surgery. First da Vinci system was bought and installed in the Central military hospital in Prague, Czech republic in 2005. The common knowledge about using this method is very low in both general public and specialists. Nor patients, neither their doctors often know about possibility of surgery using da Vinci system. Possibilities of prevention against burn-out syndrome for helping professionals working with children

This bachelor thesis: Possibilities of prevention against burn-out syndrome for helping professional with children is divided into two parts - theoretical and practical. Theoretical part is split into three chapters. The first one is dedicated the burn-out syndrome, its definition, symptoms, circumstances and ways that can lead to the syndrome. It is dedicated to phases in which the syndrome takes place and there are also described people who are threatened by the syndrome more than the others. The second chapter is dedicated to stress. The last one is dedicated to the prevention of burn-out syndrome in private and work life. The aim of the practical part is a quest to find knowledge and approach of helping professional working with children to the burn-out syndrome and its prevention. Component of the practical part of the thesis is a small research that was accomplished through interviews with twelve employees of Základní škola pro zrakově postižené in Prague. Research sphere concerned the burn-out syndrome in general, the prevention of burn-out syndrome in private and work life.

Prevention of HIV/AIDS and House of Light in Prague During 2005 - 2008

In meiner Arbeit widme ich mich der Problematik HIV/AIDS. Ich versuche auf Erheblichkeit der Probleme und auf die Hauptfragen der Prävention HIV/AISD in der Tschechischen Republik aufmerksam machen. Der Zielpunkt meiner Arbeit ist die Beschreibung und die Bewertung von Dienst für HIV-positive Menschen und für AIDS-Kranken. Diese Dienste ermöglicht Prager "Dům světla". Ich beschreibe die Entwicklung in den letzten Jahren, Erfolge, Probleme und auch Voraussetzungen für weitere Verbesserung der Prävention in der Tschechischen Republik. Die ersten Kapitel sprechen von den Hauptfragen des Phenomens HIV/AIDS und von der Prävention in der Tschechischen Republik. Nächste Kapitel betreffen die Tätigkeit der "Tschechischen Gemeinschaft AIDA Hilfe", die der Begründer von "Dům světla" ist. Kapitel 6 - 10 sind der Tätigkeit von "Dům světla" gewidmet und zugleich sprechen von Veränderungen, zu denen in der letzten Jahren kommt. In der letzten Kapitel 11. vergleiche ich die Stellungen der Klienten zu der Tätigkeit von "Dům světla" und ich spreche davon, welche Dienste in Jahren 2005 - 2008 möglich waren. Early Baroque Altars in Prague

ENGLISH ANNOTATION This paper discusses Prague altars that were crafted in the years of early-baroque between 1620 and 1680. It contains a brief outline of the historical period during which - in relation to counter-reformation and recatholization after the defeat of the Estate uprising in 1620 - baroque style had been fully accepted in Czech lands. The church, supported by the state, had made it its goal to restore catholicism in Czech lands and for this purpose it also used the means of arts. The baroque church combined all kinds of arts in its complexity. The main attention of its builders was paid to altar constructions, most often wooden ones. This paper then analyzes the history and typology of the altar and its components - the altar table, the retabulum (altar leg) and the tabernacle (sanctuary). The overall development of early-baroque altars dated from late-renaissaince mannerism to the beginning of top-baroque is discussed, together with the metamorphosis of the altars' architectural concepts, the artistic perfection of the altars and of their decorative elements. Resource-based classification of habitats of Central European macro-moths

To be used as a predictive conservation tool, classification of animal habitats should rely on actual resource requirements of individual species. Shreeve et al. (2001) illustrated this for British butterflies (J. Insect Conserv. 5, 145-161). To test this approach for a group with less-known distribution and biology, I produced a resource-based classification of habitats of Central European moths. I considered macrolepidopteran families except the megadiverse Geometridae and Noctuidae. Our data consisted of 179 binomically-coded traits for 164 species. I used PCA to ordinate the moths according to their resources, and compared resulting classification with external ecological data. The moths formed five groups: (i) associated with dense arboreal vegetation; (ii) associated with grasslands of herb layer; (iii) associated with sparse woody vegetation; (iv) early successional hawk moths; and (v) lichen-feeding arctiids. The classification was also robust after deleting hostplant taxonomy. Groups (i), (ii) and (iii) sustained control for taxonomic position, whereas (iv) and (v) did not. The groups differed in the representation of biogeographic elements, size-classes of distribution ranges, pest species, and Red-listed species. Endangered species were over-represented in (ii) and (iii) and underrepresented in (i). These preliminary results illustrate the utility of this resource-based approach for predictive inference in conservation.

The settlements and agriculture of the margins in the later middle age and early new age

This doctoral thesis presents the study of the rural milieu in the Later Middle Ages and the Early New Ages, especially with regard to its economic and settlement aspects. As a useful source of specific information we chose enclaves of margins since they are connected with contrasting settlement situation. This contrast served us well as a starting point for a discussion of more general features. The dissertation is divided into four parts. The first part deals with general problems of intensive agricultural exploitation of margins. In the second part, we bring forward a concrete example and discuss agricultural systems, which were employed on the enclave from the Early New Ages, namely, on the aeolian sands near Sadská (Central Bohemia). The third part is devoted to the beginnings of the medieval settlements in this particular region. This part of our research we based on written sources. The subject of the fourth part is the analysis of all relief remains represented by the deserted medieval village Kří (near Sadská) situated on the aeolian sands. It was this deserted village, together with the interest in relief remains of medieval settlements hitherto preserved in the landscape, that gave incentive to write this dissertation.
