 Comparsion of the blood lactate level of an elite judokas during training and competition loads Objectives: The aim of this study was to compare the lactate level of an elite judokas during training and competition loads and to identify potential relationships with other entering variabilities. Methods: The chosen method was empirical, ie collecting data from its own research and statistical representation of the data and linked to the context with other studies. Results: It was found that lactate levels during competition load increases dramatically over training load and pulse rate has no statistical significance as an indicator of the accumulation of lactate. The role of lactate shuttle in ischemic-reperfusion injury of rat heart adapted to chronic hypoxia

Adaptation to hypoxia is a well-known phenomenon increasing myocardial resistance to ischemia-reperfusion (I/R) injury as an appropriate physical exercise which improves the contractile function of the heart. Lactate is a major energy substrate for the heart muscle during physical activity and hypoxia. The metabolism of lactate was and still is associated with muscle fatigue, but in the last decades it has been considered its significant modulating function of metabolism during exercise at cellular level and whole organism level. It has been shown that its effects might be similar to the effects of hypoxia and its oxidized form, pyruvate, has the cardioprotective effects. The aim of this study was to compare the expression of LDHA and LDHB isoforms between left and right ventricle in the cardioprotective scheme of adaptation to hypoxia. Another objective/goal was to determine the left ventricular response to I/R insult in the perfused heart model adapted to hypoxia compared with the normoxic controls on/at the expression level of both LDH isoforms. 3 ABSTRACT Title: Comparison of lactate level at the same load in a swimming treadmill and on a bicycle ergometer Subject: 6 students (3 girls and 3 boys) from the first year of Bachelor's study at the Faculty of Physical Education and Sport of the Charles University at the age of 20,5 years ± 3 years took part in this research. Goal of the research: To compare demands of the load in the swimming flume with the same load on the bicycle ergometer, mainly at the level of the anaerobic threshold. Methods: We tested students by the method Critical Swimming Speed (CSS) for finding out their anaerobic threshold. Then students swam physical tests in the swimming treadmill - flume. The test consists of three 6 min parts. The first part of the swimming test was under the level of the CSS, the second part was at the level of the CSS and the third part was above the level of the CSS. We used the device for measuring the beat frequency (sport-testers) for obtaining information about changes of the heart rate (HR) during tests. Afterwards we accomplished tests on the bicycle ergometer, where we maintained HR at the same levels as in the load in the flume. The use of MR spectroscopy in neurosurgery

Proton MR spectroscopy is a non-invasive tool for measuring in vivo concentrations of several metabolites. The aim of this thesis was to test its applicability and reliability in neurosurgical praxis. In the first part of the study multiple MR spectroscopy methods were applied in a group of patients after surgery and oncologic treatment for high-grade glioma to test which method performed best in discriminating recurrent tumor from radionecrosis in the presence of a new enhancing lesion. The best diagnostic yield was achieved by comparison of choline, creatine and lactate between lesion and contralateral side (sensitivity 93.3%, specificity 78.6%). Creatine was significantly decreased in patients compared to controls. The inhibiting effect of ongoing oncologic treatment on cerebral and tumoral metabolism makes differential diagnosis trickier. Therefore, a diagnosis of radionecrosis assessed during ongoing radio- and chemotherapy should be confirmed after its completion. In the second part of the study MR spectroscopy data was compared with MR hippocampal volumetry and transcranial doppler examination in a cohort of patients with unilateral occlusion of the internal carotid artery. The lactate level assesment in various body fluids plays an important role in the diagnostics of mitochondrial disorders in children. However, the interpretation of lactate level is often difficult due to its unspecificity and variability even in particular mitochondrial disorders. Three specific aims have been stated in this PhD Thesis: 1. To analyse the role of lactate examination in the differential diagnosis between children with mitochondrial disorders and children with other diseases. 2. To study the lactate level differences in various mitochondrial syndromes. 3. To characterise the clinical and laboratory data of neonates with mitochondrial disorders and to suggest new diagnostic algorhytms. Clinical and laboratory data from patients hospitalized in the Department of Pediatrics were collected. Laboratory methods were provided in the cooperation with the Mitochondrial laboratory of the Department of Pediatrics and Institute of Inherited Metabolic Disorders. The study with lactate levels in 107 patients documented that brief seizures lasting less than 2 minutes did not increase lactate concentration in the CSF. Title: The Comparison of Results of Diagnostic Methods of Anaerobic Thresholds in Rowing Aims: The aim of the work is to compare the most used diagnostic methods of anaerobic thresholds on a rowing machine. Methods: This diploma thesis is a quantitative empirical research. To compare diagnostic data was used the Concept 2 rowing machine. The rowing machine was also used as an instrument for evaluation of five types of test methods for the anaerobic threshold diagnostics and the sixth control test, respectively. Exanimated functional values were determined using a HR monitor Garmin Fenix 6 and a Lactate scout lactometer. Exponential regressions in MS Excel were used to evaluate lactate curve tests. The homogeneous sample group consisted of 10 club rowers (n = 10). Results: We used basic statistical methods for data analysis, paired t-test, and correlation analysis (Pearson's correlation coefficient). Comparing the functional values of the monitored diagnostics, we found that the descending time protocol of the lactate curve was the most accurate methods of anaerobic threshold diagnostics for the control twenty-minute test. Biochemical parameters of energy metabolism in cerebrospinal fluid in inflammatory and non-inflammatory CNS diseases.

The basic examination of the cerebrospinal fluid provides a quick orientation in the diagnostic algorithm of CNS diseases about the nature of the pathological process. The current evaluation of cytology and biochemical parameters of glucose metabolism reports about the cells present and at the same time about the level of their activation. The aim of the work was to investigate the relationship between biochemical and cytological findings in cerebrospinal fluid in a large data set (n = 8 178), or confirm the hypothesis of a significant alteration in the Coefficient of Energy Balance (CEB) in cytological classes typically accompanying oxidative burst of phagocytes. CEB was subjected to analysis and comparison with other energy parameters (lacto-glucose ratio, gluco-lactate ratio, concentration of lactate and glucose in cerebrospinal fluid). The relationships between blood and cerebrospinal fluid glucose and lactate concentrations were investigated. CEB values were statistically significantly different in cytological groups, which reflected purulent inflammation, tumor involvement or infection by potential intracellular pathogens, the hypothesis was confirmed. Title: Comparison of internal loading on the organism by double poling and diagonal striding during the sprint on the roller ski Objectives: The aim of the diploma thesis is to compare the internal loading using the Cortex Metamax 3B and the SenzoStar GL devices in a two-hundred meters long section performed by the maximum effort between the double poling and diagonal striding about the 10.4% climb on roller skis. Methods: The diploma thesis is conceived as an empirical field research, the aim of which is to compare a double poling and diagonal striding in terms of internal loading on roller skis during sprint. The test subject will be a group of cross-country skiers and long-distance skiers. Results: Using the measured values of the internal loading and the measured time, we found differences during comparing the diagonal striding and double polling in specific values of SF, VO2 / kg, VE. the conclusion was that diagonal striding is faster than double poling to the uphill on roller skis. Title: Effects of single beta-alanine dose on performance at submaximal load Objectives: The aim of this study was to investigate the effects of single beta-alanine dose on blood-lactate level in capilary blood and whether it will affect the speed of passing the test. Methods: A double-blinded cross-over study conducted on 16 male students of Military Department at the Faculty of Physical Education and Sports of Charles University (mean ± SD: weight 79,31 ± 6,97 kg; height 181,56 ± 6,36 cm; age 21,63 ± 1,45 yrs). The study was performed in Military Sport Centre Pilotů 217/12, 161 00, Prague. Each volunteer underwent two performance tests after consuming either placebo (maltodextrine) or beta-alanine (50 mg/kg). Time needed to complete the test and blood-lactate concentration before and during 5th and 10th minute after completion was measured. Volunteers also filled a short questionnarie regarding subjectively percieved load and side effects, and whether they were administered beta-alanine (BA) or placebo (PLA). For verification of normal distribution were used Q-Q Plots and statistical significance of BA use was determined using student's t-test. Effect of alkaline water consumption on sports performance

Title: Effect of Alkaline Water Consumption on Sports Performance Objectives: The aim of the work was to find out the differences in performance after three days of intake of alkaline water in comparison with intake of water with normal pH. Methods: The thesis is conceived as an experimental study. A total of 12 participants aged 19-28 participated in the study. Volunteers were students of the UK FTVS military field. The testing included repeated crate outputs (Step-test), reaction rate tests on visual and auditory stimuli. Before and after exercise, blood lactate value, specific urine weight, and body diagnostics were recorded using a bio-impedance device. The study lasted a total of 3 weeks. Testing was performed with weekly spacing in controlled drinking regime of common or alkaline drinking water, always 2 days before testing. The obtained data were then analyzed and processed. Results: The average power of the Step-test participants in the first part of the measurement for both tests after the use of plain water is 718,8 W, compared to the average power of the participants after the use of alkaline water, which is higher and reaches 848,8 W.
