Pollens of Selected Alergenic Plants and their Didactic Use in Teaching Biology at High Schools

In my thesis I deal with the pollen of allergenic plants, the origin and proces of allergy to plant pollen and the inclusion of these topics in the teaching of plant biology in high schools. In the theoretical part I focus on topic of pollen allergy. First, I generally describe allergy and its origin, causes of allergic diseases, environmental factors and the development of allergic disease. I also briefly address the naming and registration of allergens and I also mention the so-called cross-allergy. Great emphasis is placed on pollen allergy, which is devoted to a larger unit, divided into individual subchapters dealing with the physiological function of the nose, followed by an allergic reaction of type I, an allergic reaction that occurs on the nasal mucosa. Furthermore, subchapters mentioning the classification of allergic rhinitis, diagnosis and pharmacotherapy, including additions to the drug groups used in the treatment of pollen allergy. Another larger unit is devoted to the structure of the plant's blossom as a source of allergen, which includes the anatomy of the blossom parts and a description of the pollen grain. This bachelor thesis deals with the business plan for FORALL s.r.o. company pro-viding services in gastronomy. The thesis consists of two parts. The first, theoretical part explains the general terminology and defines the types of business en-tities. Also, it describes general requirements on a business plan. The second, practical part presents particular business plan of the company including company description, market analysis and financial plan. The purpose of this thesis is to find out whether the company might succeed on the market. The conclusion of the thesis is that this business plan is useful basis for foundation of a new restaurant.

The causative factors for food-allergic reactions are proteins that are termed allergens. Allergenic proteins usually have high nutritional value for the major part of the population but threaten the health of a subset - the food-allergic consumers. The severity of allergic reactions varies from mild urticaria to potentially fatal anaphylactic shocks. At the moment, the only proven therapy for food allergy is a strict avoidance. This implies that people with a food allergy need to know whether the food products they purchase are indeed free from the ingredient that can trigger an allergic reaction in their body. Labeling of allergens has the potential not only to identify foods that are unsafe for allergic consumers but also to increase the choice of safe food products. Legislators and regulatory agencies have recognized the importance of labeling with regard to protecting the allergic consumer. As a result of this, in many jurisdictions in different parts of the world new legislation specifically aimed at the protection of allergic consumers has been put in place. This legislation has resulted in a mandatory labeling of the 13 most commonly allergenic foods (plus sulfites).

Due to high incidence, medical and socioeconomic burden and impact on individual quality of life and productivity, allergic disorders are a crucial issue for 21st century immunology. Much still remains to be elucidated, particularly regarding the very early processes in allergy development. In order to introduce timely, effective preventive measures, novel, more reliable predictive factors of allergy risk also need to be established. Dysregulation of proper balance between the branches of immune response, particularly unwarranted dominance of Th2, is the underlying cause of allergy. After birth, new immune balance needs to be established to prepare the neonate for adequate reactivity towards newly encountered environmental stimuli. Regulatory T cells (Treg) play a central role in finely setting this balance and inducing tolerance towards harmless environmental antigens, including allergens. Interactions with external factors, most importantly microbiota, modulate this process during the early postnatal "window of opportunity." Analysis of cord blood Treg of children of allergic mothers uncovered decreased presence of function-associated surface markers and lower production of IL-10. Furthermore, decreased proportion of Helios- induced Treg was observed in children with higher risk of allergy....

7 Abstract The thesis is focused on the pollen of selected allergenic plants. Thesis is structured into two parts., pollination and fertilization , development of the pollen grain , its structure and chemical composition. The end of the theoretical part, dedicated to allergies, is divided into subsections dealing with First, teoretical part describes the flower and its morphology, discusses the pollen grain, stamen anatomical structure immunity and allergies, naming and registration of allergens and cross- pollen allergy . The practical part contains an analysis of curriculum for students of secondary schools. An integral part of the work is a survey of teachers in secondary schools, in which method of questioning was used. The aim of this questioning was to determine the presence of solved issue in terms of representation of pollen and pollen allergens in textbooks for secondary schools , as well as the inclusion of this issue in the official curriculum, under the National Programme for the Development of Education in the Czech Republic. Based on the results of a questionnaire survey of secondary school teachers, teaching materials have been developed in the form of two worksheets for teaching botany and guidance on laboratory exercises in plant biology.
Validation of PCR-RFLP method for determination of lactose intolerance

The lactose intolerance is the most common food intolerance occurring worldwide. Individuals having the lactose intolerance cannot produce the laktase enzyme in thein small intestine. This enzyme is able to digest the lactose contained in milk. There are several wals for this disabilitty. The practical part of this bachaleor thesis is focused on the genetic origin of the lactose intolerance. In the population of Europe we can find as the most frequent the two polymorphism of the MCM6 gene that affect the production of laktase C/T13910 and G/A22018. In principle it concerns the new gene station which ensure the production of laktase enzyme even in adult age. The aim of this bachaleor thesis is to creat the research based on the recent knowledge Publisher in the specialized literature. There are gin to be describe all type sof the lactose intolerance and the ways of its diagnostics, futhermore e.g. the princupli of Alleny, the used laboratoř methods, the rule sof validation methods etc.. The pros and cons of every testing approach will be explained. The practical part is aiming at the implementation of the validation of the genetics test used for petting down the lactose intolerance and creating the validation protokol that will serve as a base of my bachaleor thesis. The genetic Cheb-up is based on the PCR-RFLP principle, within the metodology the laboratoř procedures being used in the genetic lab will be described. The validaton will flow into a detection if the applied method is sufficiently valid fot the diagnostics. The genetic analysis will focus on the two selected polymorphism C/T13910 and G/A22018. The sample with a familiar genotype that has already been verifie by sekvenation will be used for validation.
Allergen labelling in catering facilities

The new EU food legislation established an obligation to indicate allergens to prepared meal and defined specific ways of providing this information. The aim of this work is to look for possibilities of allergen labeling on menu. This work is devoted to the analysis of customer satisfaction with individual ways of information about allergens. In conclusion, there is a recommendation that includes the requirements of European legislation and the opinions of customers of catering facilities.
Bronchial asthma in the practice of paramedic

Astma bronchiale is a lifelong chronic respiratory illness which affects all the age categories of population. The aim of this final work is to obtain knowledge and to analyse paramedics´ approaches used during a diagnosis and providing prehospital urgent care to patients affected by this illness.The bachelor´s work is divided into a theoretical and a research part. The theoretical part deals with the anatomy of the respiratory system, process of breathing, diagnosis and treatment in prehospital urgent care.The research of this work was realized by using recorded interview with paramedics. The aim of this research is to show the reader more viewpoints to this issue and to outline the experience of paramedics with treatment of this illness. This research demostrates that paramedics get in contact with the asthma bronchiale several times in a month, they are familiar with this illness and they know methods of its treatment. The result of the work shows how difficult the diagnostics of this illness is. Differential diagnostics also points to broad-spectrum of illnesses. Eventually, we can also enroll a wrong feedback between emergency service and medical facility. Results of this work in the theoretical part help the reader to get objective information about this matters, diagnostics and treatment of this illness in prehospital urgent care. The main contribution of this work should also be the view to the methods, approaches and experiences of paramedics during the treatment of the asthma bronchiale in prehospital urgent care. The output of the bachelor´s work is a brief manual for paramedics showing how to proceed during diagnostics and treatment of the asthma bronchiale.
