Modification of electroinsulating varnish with ground mica
Drápal, Aleš ; Frk, Martin (referee) ; Rozsívalová, Zdenka (advisor)
This master's thesis focuses on composite systems based on varnish combined with different filler ratios of micronized mica. The aim is to analyse impact of the filler on dielectric properties of the varnish, i.e. relative permittivity and loss factor as functions of frequency as well as charging and discharging currents as functions of time. Dielectric mixture formulas are applied on relative permittivity values. Calculated and measured values are compared.

Activating antitumor immune response using bispecific fusion proteins
Chytrá, Gabriela ; Vaněk, Ondřej (advisor) ; Černá, Věra (referee)
Natural killer (NK) cells are lymphocytes of the innate immune system that recognize and eliminate transformed and potentially harmful cells in a mechanism termed immunosurveillance. Malignant cells strive to escape immunosurveillance, and if successful, oncological disease develops. To restore immune recognition, immunotherapy utilizing NK cell-directed therapeutic fusion proteins can be employed. Therapeutic fusion proteins target tumour markers expressed on the surface of malignant cells and, at the same time, stimulate immune response through binding to NK cell activating receptors, for example receptor NKG2D or NKp30. A relevant example of a tumour marker is the HER2 receptor, which is often overexpressed in several types of cancer, most notably breast carcinoma. This thesis describes the preparation of several bispecific fusion proteins with potential use in immunotherapy. Bispecific fusion proteins consist of an NK cell activating ligand (ligand MICA or B7-H6) and nanobody targeting selected tumour marker (receptor HER2), which are connected by flexible glycine-serine linker. The constructs of fusion proteins were prepared in two configurations - with nanobody located on the N-terminus and the ligand on the C-terminus and vice versa. In addition, bispecific fusion proteins introducing...
Preparation of a polymeric therapeutic targeting CD20 positive B-lymphomas and NK cells
Hejl, Maxmilián ; Vaněk, Ondřej (advisor) ; Bělonožníková, Kateřina (referee)
Malignant transformation of B-cells is manifested by a marked increase in the number of surface markers 20 (CD20, cluster of differentiation 20). In studying this trend, chimeric monoclonal antibodies targeting CD20 were introduced to induce apoptosis in B-lymphomas. Since the introduction of the first therapeutic monoclonal antibody rituximab, many others have been developed, with some still used to treat B-lymphomas today. Unfortunately, in many cases, resistance to these drugs is developing, and therefore the development of new types of therapeutics is still relevant. This work aims to develop a polymer-protein macromolecular conjugate capable of inducing apoptosis in CD20 positive leukemia cell lines. For this purpose, we work with biologically active vectors, so-called anti-CD20 nanobodies. This is a variable binding domain derived from the "heavy chain only" antibodies found in, e.g., llamas or camels. Compared to conventional antibodies, nanobodies are approximately ten times smaller, but their binding affinity for the antigen is not altered. For this reason, nanobodies are ideal candidates for attachment to a polymeric carrier, where poly-N-[2-(hydroxypropyl)methacrylamide] (pHPMA) was chosen in this work. The transpeptidase reaction catalyzed by recombinant sortase A, which recognizes and...
Decentralized Finance from the Perspective of Czech Financial Regulation and MiCA Proposal
Koliba, Viktor ; Sejkora, Tomáš (advisor) ; Kotáb, Petr (referee)
1 Decentralized Finance from the Perspective of Czech Financial Regulation and MiCA Proposal Abstract Decentralized Finance constitutes a new system for allocating resources by using irreplicable digital data that can be processed within a predefined framework of automatic operations based on blockchain technology. Its development has brought not only new economic opportunities but also a number of risks and legal issues. Since Decentralized Finance is in its early stages, relevant legal questions relating to its positioning within the existing and forthcoming legal frameworks are still not firmly established yet. This thesis aims to answer those relating to the applicability of selected Czech financial regulations and the European Commission's Proposal for a Regulation of the European Parliament and of the Council on Markets in Crypto-assets, as well as validate the hypothesis that the wide scope of the MiCA Proposal could influence the ecosystem of Decentralized Finance. First, the important technical foundations of Decentralized Finance and their specifics were outlined, and relevant financial applications were established for further assessment. Subsequently, the applications were compared with the scope of the chosen Czech financial regulation, namely the Act on Payment System, the Act on Currency...
Financial Aspects of Global Payment Systems
Moravec, Jiří ; Kohajda, Michael (advisor) ; Boháč, Radim (referee) ; Papoušková, Zdenka (referee)
Financial Aspects of Global Payment Systems Abstract This dissertation summarizes findings on Digital Asset's development, which would fit under the era of Blockchain 1.0. We analyzed and synthetized available resources focusing on the following areas: (i) historic aspects of Digital Assets, (ii) technical solutions of Digital Assets, (iii) actual use of Digital Assets, (iv) abuse of Digital Assets, (v) Digital Assets' legal integration, (vi) Digital Assets as a global payment system. Analyzing history of Digital Assets, we summarize that Digital Assets were developed with the intent to liberate payment systems from existing financial supervision. Once such system became functional it was immediately abused. In connection with technical solutions, we find that as technical complexity of Digital Assets (especially the lack of the trusted third party) diminishes protection of Digital Assets users, it incentivizes criminal activity. Consequently, Digital Assets are vastly abused for different criminal purposes, including development of services dedicated to criminal activity, such as Dark Web Marketplaces or Digital Assets Mixer. Further, Digital Assets are used for payments on minimal scale, and the retail use if practically nonexistent. Currently, Digital Assets' legal integration is slow and fractional;...
Proposal for a Regulation on Markets in Crypto-Assets: Impact on Currently Available Stablecoins
Vilímek, Matyáš ; Sejkora, Tomáš (advisor) ; Kotáb, Petr (referee)
1 Proposal for a Regulation on Markets in Crypto- Assets: Impact on Currently Available Stablecoins Abstract On 24 September 2020, the European Commission introduced its Proposal for a Regulation on Markets in Crypto-Assets (MiCA). In addition to regulating general crypto-assets, so-called crypto-asset service providers, and market abuse in relation to crypto-assets, MiCA also lays down rules for stablecoins, i.e., crypto-assets which aim to maintain a stable value. This thesis analyses how MiCA, if passed into law as introduced by the Commission, would impact the stablecoin market as it exists today, and how this would affect the ways people already use stablecoins. It does so by asking and answering these questions: How do different types of stablecoins function? If MiCA were applied to these stablecoins, would they be compliant? If not, which rules would they not comply with? If the rules were not changed, could we realistically expect such stablecoins to adapt? If they would not adapt and would cease to function, how would it change the way people currently use stablecoins? In its core, the thesis selects five stablecoins to represent the broader stablecoin market and then describes how they function. These stablecoins are Tether (USDT), TerraUSD (UST), Magic Internet Money (MIM), Fei USD (FEI), and...
Utilization of mica separated from washed kaolin
Vaculík, Josef ; Ptáček, Petr (referee) ; Opravil, Tomáš (advisor)
The Master’s Thesis aims to laboratory test the possibilities of utilization mica separate, which arises during the process of floating kaolin as a by-product together with sand. Mica and sand are then separated by flotation or vibration. The mica separate thus formed was subjected to analysis. XRD and heating microscopy methods were used for analysis. Subsequently, experiments with mica separation as a filler in composites based on epoxy resins were set up and performed. Furthermore, the separate was tested as a part of plasters and visual building elements. Last but not least, the separate was mixed into the ceramic, which was then subjected to firing in the selected mode. The diploma thesis provides a comprehensive overview of the above-mentioned possibilities of using mica separation, which is based on a sufficient amount of experimental data.
