Optimization of calcium chloride concentration for removal of polysaccharide contamination during plant DNA isolation

The greatest difficulty in isolating DNA is the presence of contaminants that cause side effects. Polysaccharides are the most common contaminants in fruits. They can distort the results in spectrophotometric determination of purity or act as inhibitors in PCR analysis together with other substances (for example, proteins or phenolic substances). The aim of this work was to investigate the influence of different concentrations of calcium chloride on the process of DNA isolation. In the experimental part, DNA from the apple was isolated using different concentrations of calcium chloride. The isolation was carried out four times, and each time the sample was adjusted in different ways. It was found that the isolation method used works only with a sample that has been lyophilized. Isolation of DNA from fresh fruit provided very low yields. Probably, this was due to the large water content in the sample, and the proportion of the solid component was smaller. Subsequently, PCR analysis and electrophoresis were performed to determine the amplifiability of the isolated DNA. Two sets of primers with different specificity were used for this analysis. Amplifiability was confirmed only when using primers specific to apple DNA when using 100 mM solution of CaCl2. Other samples have been amplifiable using both types of primers. Probably, samples isolated using a 100 mM solution of CaCl2 had a larger amount of inhibitors that do not affect all PCR reactions equally, which may also indicate a small effectiveness of this amount of CaCl2.
Documentation of optical network route using low-cost lidar equipment

This work aimed to test the potential benefits of handheld lidar for optical network route documentation. For this purpose, a lidar embedded in an Apple iPhone 14 Pro mobile phone was used. The work provides insight into the issues of both the data acquisition by this device and the subsequent processing and evaluation of the route, including the surrounding topography. All the procedures have been documented and their findings written into the text to facilitate familiarity with the equipment used. Problems encountered during scanning and data processing were outlined. Also, comparisons were made with currently used methods. The evaluation of the accuracy of the measured data was carried out by independent measurement of the features using the GNSS method. Based on the acquired data, documentation of a part of the newly surveyed route was made at the selected location according to the guidelines of the optical network administrator. The outputs of this thesis can be used to discuss the possible use of the tested scanner today as well as in the future.
RSS Reader for iOS

The aim of this work is to create a mobile application for iOS operating system for easy following, reading and storing/archiving of stories from news sites and blogs accessed through web feeds in RSS format. The application is implemented in Swift programming language. Apple's Core Data application framework was used for data persistence. The work consists of theoretical and practical parts. The theoretical part describes the RSS format and principles of programming an application for iOS operating system. The practical part contains description of design, implementation and testing of the application. The resulting application provides user with a simple instrument to work with RSS channels and stories, and by following the Apple Human Interface Guidelines the application fits naturally in iOS ecosystem.

Tato práce bude zaměřena na ohodnocení jazyka v recenzích mobilních telefonů. K tomuto účelu bude použita recenze Apple iPhone 7. Apple má širokou nabídku produktů, jmenujme počítače, tablety, hodinky nebo mp3 přehrávače. Mobilní telefony jsou ale jedním z nejpopulárnějších produktů. Z tohoto důvodu, jsou recenze mobilních telefonů perfektním zdrojem pro tuto tezi. Účel této práce je analyzovat a demonstrovat prostředky ohodnocení jazyka v reklamní mluvě a odhalení strategií a jazykových prostředků použitých k ovlivnění čtenáře, při apelování na city, morálku a estetiku chápání. Předpokládám, že recenze jsou zaujaté díky použité formě jazyka, která může obsahovat osobní názory autora nebo metody manupulace se čtenářem. Tyto metody mohou být použity při stylizaci textu, mohou apelovat na emoce nebo sociální zázemí čtenáře nebo mohuu být použity při rozhodování o tom zda a jaké informace v recenzi poskytnout. Tato teze ověří, zda je tato hypotéza platná.
Cooking Advisor for iOS

The bachelor thesis deals with the design and implementation of a mobile application for iOS devices that allows easy sharing of recipes. The design was developed in diagrams or Figma tool. The implementation is in swift language using CoreData, CloudKit, VisonKit, SwiftUI frameworks. The focus in the design and implementation was on usability, extensibility and synchronization with cloud storage in sync with persistent storage. The thesis discusses iOS application development tools, analysis of each technology, design and implementation of the application and finally testing of the application.
Library of UI Components for Developing Applications in iOS

In this thesis I analyse and implement a set of frequently used components that are not part of the standard set of user interfaces. Then I implement these components using the UIKit and SwiftUI libraries. In this work I also focus on the implementation of strongly typed data models. In the end I verify the implementation of the set of components on a demonstration application.
Influence of Storage Conditions on Metabolic Profile of Apples

The goal of presented dissertation has been complex analysis of changes in content of fatty acids, enzymatic and low-molecular antioxidants in apples and related evaluation of perception of originators of storage diseases for apples stored in various conditions. Main part of the work has been dedicated to study of the impact of storage of apples in modified atmosphere with reduced amount of oxygen and in reference “normal” atmosphere for six months. Next part of the work has been dedicated to study how apple storage in common, consumer affordable, conditions (storage in a cellar, in a refrigerator, and in room temperature) affects content of low-molecular antioxidants. For testing has been selected apple kinds Jonagored, Golden Delicious, Idared, Šampion, and Granny Smith. Within the scope of this work has been optimized the method for determination of fatty acids in plant material with higher content of wax. The measured values imply the apples are valuable source of many important nutrition substances like vitamins, provitamins and antioxidants. During the storage process these substances exhibit considerable protective function. Long-term storage, mainly in the atmosphere with reduced amount of oxygen (FAN), enables preservation of majority of these important nutrition substances depending on the kind of apple and conditions of storage. Freezing process is conservative to apples (mainly in the presence of protective substances), while in the process of drying the values of all monitored antioxidants decrease depending on temperature and conditions of drying. Upon the choice of the storage method one must consider nutrition, sensoric characteristic and consumer demands.
iOS Application for Festival Visitors

The aim of this thesis is to implement iOS application aggregating information about festivals to one place. Application should provide general overview about individual festival events and help users with time planning. Thesis is describing process of creation from user interface design and system architecture to implementation with final testing. The result is client-server application providing web interface to organizers for any kind of festival registration which is later available in the mobile application. Server is implemented in Django framework, client application in framework SwiftUI.
Determination of athenticity of plant foods by molecular techniques

The aim of presented diploma thesis was to determination of authenticity of fruit baby foods for early infant feeding using molecular methods. In the experimental part, isolation kit was used for isolation of plant DNA from fruits (strawberry, apricot, raspberry, apple) and from six commercial fruit products for children. Isolated DNA was characterized and verified using PCR methods with primers specific for plant rDNA (ITS2). Specific primer pairs were designed to amplify DNA for the detection of one fruit species. Primer specificity was assessed with four fruit species. A mixture of fruit puree from the two fruits was used to determine the sensitivity of the multiplex PCR assay. Six commercial fruit products were evaluated to verify the applicability of the multiplex PCR assay. The methodology of molecular detection of fruit DNA by qPCR and multiplex qPCR (duplex) includes approaches, which enable to detect two fruits (strawberry-raspberry, apricot-apple) in one reaction and thus reduces time and money requirements.
Sport Assistant for iOS

The aim of this thesis was to create a mobile phone application for iOS, that would be used for recording all sport activities of a person. The application can record activity using GPS or count the exercise manually, automatically at time intervals or by motion sensor. For recognition of moves while using motion sensor was developed a library called Motion Recognizer. The application uses HealthKit framework. The application also offers an opportunity of planning a sport activity by using Trainer objects. It uses Core Data framework while working with data and it backs up the data on iCloud.
